β-III Tubulin Levels Determine the Neurotoxicity Induced by Colchicine-Site Binding Agent Indibulin

Summary: Indibulin, a microtubule-depolymerizing agent, shows minimal neurotoxicity and is less cytotoxic towards differentiated neuronal cells. It binds to beta-tubulin isotypes differently and has a lower affinity to beta-III tubulin. Depletion of beta-III tubulin increases the toxicity of indibulin and colchicine, while vinblastine is unaffected. Indibulin inhibits colchicine binding to tubulin and is a colchicine-site binder.

Summary: Compounds targeting tubulin as potential anticancer agents have shown promising efficacy and safety, with CTR-21 and CTR-32 identified as lead compounds that effectively kill a variety of cancer cells, including multi-drug resistant cells, and exhibit high selectivity against cancer cells. These compounds prevent tubulin polymerization and induce cell cycle arrest at G2/M, with CTR-21 demonstrating more favorable metabolic properties and showing synergistic effects when combined with the Bcl-2 inhibitor ABT-737.

Molecular Docking of Cryptoconcatones to α-Tubulin and Related Pironetin Analogues

Summary: Cryptoconcatones A-L is a series of 12 dihydropyrone derivatives isolated from the evergreen tree Cryptocarya concinna Hance, with potential antiproliferative activity against cancer cells. In this study, the interaction of these compounds with tubulin dimers was investigated using molecular docking. Two compounds, cryptoconcatones F and L, showed stronger binding capacity to alpha-tubulin than pironetin, a known microtubule polymerization inhibitor. The study contributes to the understanding of the mechanism of action of cryptoconcatones and may aid in the design of analogues targeting the pironetin site of alpha-tubulin.

Identification of novel microtubule inhibitors effective in fission yeast and human cells and their effects on breast cancer cell lines

Summary: Researchers identified novel small molecules that inhibit microtubule organization through sequential phenotypic screening of fission yeast and human cells. In vitro assays showed that these compounds directly bind tubulin and destabilize the structures of tubulin dimers. One compound in particular, L1, demonstrated preferential potency against human breast cancer cell lines and synergistic effects when combined with the Plk1 inhibitor BI2536, suggesting it as a candidate for cancer chemotherapy.
