OAT10/SLC22A13 Acts as a Renal Urate Re-Absorber: Clinico-Genetic and Functional Analyses With Pharmacological Impacts

Summary: By studying the dysfunctional variant of OAT10 and renal handling of urate, we found that carriers of the dysfunctional OAT10 variant have lower serum urate levels and higher urate excretion values, indicating that OAT10 dysfunction increases renal urate excretion. Functional and immunohistochemical analyses of OAT10 further confirmed its role in the kidneys, and we also discovered that losartan and lesinurad might lower urate levels by inhibiting OAT10.

Post-hoc analysis of pegloticase pivotal trials in chronic refractory gout: relationship between fluctuations in plasma urate levels and acute flares

Summary: Gout flares increased in patients treated with pegloticase during the first 3 months of treatment but then decreased significantly in months 4-6. Fluctuations in plasma urate levels, particularly in monthly responders, were associated with flare occurrence. Significant factors associated with flares were the treatment group and absolute change in plasma urate before flares according to multivariate linear regression analysis.

Drug Repurposing for Parkinson's Disease: The International Linked Clinical Trials experience

The review provides an overview of the history, structure, process, progress, and examples of agents selected and evaluated by the program.

The effect of dietary approaches to stop hypertension and ketogenic diets intervention on serum uric acid concentration: a systematic review and meta-analysis of randomized controlled trials

Summary: This study investigated the effects of two nutritional interventions (DASH diet and ketogenic diet) on serum uric acid levels. The results showed that the DASH diet significantly decreased serum uric acid levels and may be recommended for hyperuricemia states such as gout. However, the ketogenic diet did not have a significant effect on serum uric acid levels. Further research is needed to determine the effects of the ketogenic diet on serum uric acid levels.

New insight into the management of renal excretion and hyperuricemia: Potential therapeutic strategies with natural bioactive compounds

Summary: Hyperuricemia is associated with various comorbidities and is caused by increased production and/or underexcretion of uric acid. Dysregulation of xanthine oxidoreductase (XOD) and urate transporters is a major reason for hyperuricemia. There is a need for new nephroprotective drugs with improved safety profiles. This review summarizes the characteristics of major urate transporters, the pathogenesis of hyperuricemia, and potential mechanisms of action of natural compounds with antihyperuricemic and nephroprotective potential.

Prodromal Parkinson's disease: hype or hope for disease-modification trials?

Summary: The ultimate goal of PD research is to find treatments that can slow down or stop the progression of the disease. Past disease-modification trials in PD have failed due to various factors, including wrong intervention choices, inadequate trial designs, and target populations. Prospective studies have provided sufficient evidence for defining diagnostic criteria for prodromal PD, and different approaches have been studied in observational cohorts. This review discusses recruitment strategies, trial designs, trial durations, and sample sizes for disease-modification trials in prodromal PD cohorts, and offers an outlook on how this goal can be achieved in the future.

Uric acid-lowering effect of dotinurad, a novel selective urate reabsorption inhibitor, in hypertensive patients with gout or asymptomatic hyperuricemia: a pooled analysis of individual participant data in phase II and III trials

Summary: Dotinurad effectively lowers serum uric acid levels in hypertensive patients with gout or asymptomatic hyperuricemia, especially at a dose of 4 mg, with consistent long-term results. The drug has a high achievement rate of over 80% in reaching the target serum uric acid level of <= 6.0 mg/dL in both short-term and long-term analyses.

Associations between vitamin C intake and serum uric acid in US adults: Findings from National Health and Nutrition Examination Survey 2011-2016

Summary: This study found negative associations between dietary vitamin C and SUA levels and hyperuricemia risk among US adults. Summary: Renal hypouricemia (RHUC) is a pathological condition characterized by extremely low serum urate and overexcretion of urate in the kidney. In this study, a familial case of RHUC type 2 was described, and an intronic GLUT9 variant, c.1419+1G > A, was identified as the causal mutation that resulted in a functionally-null variant. This finding enhances our understanding of RHUC and the importance of GLUT9 in its pathophysiology.

The clinical efficacy of urate-lowering therapy in acute gout: a meta-analysis of randomized controlled trials

Summary: The meta-analysis found that using ULT agents can lower serum uric acid levels and improve medication adherence in patients with acute gout attacks. Summary: This study aimed to explore the relationship between the decrease in serum urate and the risk of gout flares during urate-lowering therapy (ULT) initiation. The results showed that the degree of decrease in serum urate is affected by baseline serum urate, and higher baseline serum urate and greater decrease in serum urate increase the risk of gout flares.

Summary: The study results show that prenatal exposure to phenobarbital may have negative effects on children's mental abilities, especially in verbal IQ and full-scale IQ. Compared to other drug groups, children exposed to PB had lower verbal and full-scale IQ scores, but there were no significant differences in the performance of children exposed to CBZ compared to those exposed to PB or PHT.

Dissecting the Domains of Parkinson's Disease: Insights from Longitudinal Item Response Theory Modeling

Summary: This study applied longitudinal item response theory (IRT) models to analyze the motor symptoms in patients with Parkinson's disease and found distinct progression patterns in tremor and nontremor domains over time and in response to treatment. Levodopa treatment showed a greater slowing effect on tremor severity compared to inosine treatment. Furthermore, longitudinal IRT analysis proved to be a valuable statistical method for monitoring changes in different but related domains.

Virtual Tai Chi program for patients with irritable bowel syndrome with constipation: Proof-of-concept feasibility trial

This study examines the role of parenting self-efficacy and family resources in mediating this relationship. The results suggest that parenting self-efficacy mediates the relationship between child behavior problems and parenting stress, and this effect is moderated by family-level resources.

Validation of a predictive model for obstructive sleep apnea in people with Down syndrome

Summary: Detecting obstructive sleep apnea in individuals with Down syndrome is crucial for preventing comorbidities and understanding its impact on mental health. However, laboratory-based polysomnograms are often not feasible for this population. A previous prediction model showed promise in identifying individuals who may not have significant apnea and can avoid a diagnostic polysomnogram. This study aimed to validate these findings in a new group of participants with Down syndrome, but a reliable screening tool for OSA was not achieved. Current healthcare guidelines recommend ongoing monitoring for OSA in individuals with Down syndrome.

The results of this study will provide information for future efficacy trials and may serve as a cost-effective solution to reduce stress in caregivers of persons with ADRD.
