Core Binding Factor Beta Plays a Critical Role by Facilitating the Assembly of the Vif-Cullin 5 E3 Ubiquitin Ligase

The HIV-1 virion infectivity factor (Vif) targets the cellular cytidine deaminases APOBEC3G (A3G) and APOBEC3F (A3F) for degradation via the host ubiquitin-proteasome pathway. Vif recruits a cellular E3 ubiquitin ligase to polyubiquitinate A3G/F. The activity of Vif critically depends on the cellular core binding factor beta (CBF beta). In this study, we investigated the Vif-CBF beta interaction and the role of CBF beta in the E3 ligase assembly. Vif-CBF beta interaction requires an extensive region of Vif spanning most of its amino terminus and zinc finger region, and cullin 5 (Cu15) binding enhances the stability of the Vif-CBF beta interaction. Our results further demonstrate that CBF beta plays a critical role in facilitating Cu15 binding to the Vif/elongin B/elongin C complex. Vif, with or without bound substrate, is unable to bind Cu15 in the absence of CBF beta. These studies support the notion that CBF beta serves as a molecular chaperone to facilitate Vif-E3 ligase assembly.

Association of genetically predicted lipid traits and lipid-modifying targets with heart failure

Summary: This study assessed the association between genetically predicted lipid traits, their modification through licensed or investigational targets, and heart failure. The results showed a significant relationship between genetically predicted LDL-C, TG, HDL-C, and ApoB levels and heart failure, mainly mediated by coronary heart disease. Drug target analysis identified PCSK9, CETP, and LPL as potential targets for heart failure prevention.

Structural basis for recruitment of host CypA and E3 ubiquitin ligase by maedi-visna virus Vif

Summary: In this study, the cryo-electron microscopy structure of MVV Vif in complex with CypA and E3 ligase components was reported. The structure, along with biochemical and functional analysis, revealed both conserved and unique structural elements of MVV Vif and its interaction modes with different host proteins. This provides further understanding of the evolutionary relationship between lentiviral Vifs and their mechanisms for capturing host cofactors for immune evasion activities.

Mental health outcomes in parents of children with a cancer diagnosis in Sweden: A nationwide cohort study

Summary: This study examined the long-term impact of childhood cancer on parental mental health, with a focus on potential sex differences between mothers and fathers. Associations of genetically determined lipid traits and lipid-modifying agents with the risk of diabetic retinopathy: A Mendelian randomization study
