Summary: RNA viruses rely on RNA-RNA interactions involving genomic structural elements for viral replication and translation. In this study, intra- and intermolecular RNA-RNA interactions in the 3' UTR of the West Nile virus genome are observed, with intermolecular interactions forming molecular dimers. Functional analysis suggests a relationship between 3' UTR dimerization and viral translation efficiency. These findings imply the existence of a network of RNA-RNA interactions involving 3' UTR structural elements that regulate viral translation.

Summary: West Nile virus (WNV) and Usutu virus (USUV), two related flaviviruses, can cause severe neurological disease in animals and humans. The immune response and immunopathogenesis of these viruses are poorly understood. In this study, mice primed with low doses of USUV showed protection against lethal challenges of WNV, even without detectable neutralizing antibodies. Furthermore, mice immunized with a chimeric virus containing USUV's E protein were also protected against WNV. These findings contribute to understanding the interaction dynamics during sequential infections of these flaviviruses.

Summary: Mutations in the precore and basal core promoter regions of the HBV genome are associated with fulminant hepatitis and HBV reactivation. This study found that these mutations might enhance viral replication and directly induce liver damage. PC/BCP mutant infection in mice resulted in the accumulation of HBsAg in the endoplasmic reticulum, leading to apoptosis through unfolded protein response in HBV-infected hepatocytes. These mutations may be associated with liver damage in patients with fulminant hepatitis or HBV reactivation.

Summary: This study examined the potential of BCL-2 inhibition in active HIV infection in vivo for the first time. It showed that venetoclax significantly enhances target cell killing induced by Fas ligand, TRAIL, and perforin/granzyme B, and synergistically enhances autologous NK and CD8 cells' killing of target cells. In a humanized mouse model, venetoclax monotherapy effectively reduces plasma viremia and restores CD4:CD8 ratios to normal levels. However, it also suggests that venetoclax should be combined with antiretroviral therapy to reduce toxicity.

Summary: In this study, chimeric HPV:HIV VLPs were constructed by inserting HIV-1 P18I10 CTL peptide and T20 anti-fusion peptide into the HPV16 L1 capsid protein. The chimeric VLPs induced both HPV16- and HIV-1-specific immune responses, making them a potential vaccine platform for controlling HPV16 and HIV-1 infection.

Summary: Influenza A viruses in swine reservoirs constantly evolve and can occasionally infect humans with potential human-to-human transmission. This study compared the pathogenesis and transmission of diverse swine-origin viruses and human seasonal viruses using ferret models. The results showed that variant viruses were more frequently isolated from the lower respiratory tract and had lower airborne transmission capability compared to human-adapted viruses.
