Summary: Through the combination of deep learning and cross-linking mass spectrometry, an atomic-level model and an integrated processing/assembly pathway for a structural component of the vaccinia virion, protein P4a, has been established. Proteolytic separation of the C-terminal P4a-3 segment triggers a conformational rotation within the N-terminal P4a-1 segment, which is fixed by disulfide-locking and removes a steric block to trimerization of the processing intermediate P4a-1+2. These events lead to the proteolytic separation of P4a-2 and allow the assembly of P4a-1 into a hexagonal lattice enclosing the nascent virion core.

Summary: This study demonstrates the role of the DECREASED ELECTRON TRANSPORT AT PSII (DEAP2) assembly factor in the early assembly of photosystem II (PSII) in plants. It shows that DEAP2 works together with the PHOTOSYNTHESIS AFFECTED MUTANT 68 (PAM68) factor to facilitate the formation of the RC47 intermediate. The loss of both DEAP2 and PAM68 leads to a loss of functional PSII and the inability of plants to grow photoautotrophically.

Summary: Two of the four human ubiquitin-encoding genes express ubiquitin as an N-terminal fusion precursor polypeptide, with either RPS27a or RPL40 at the C-terminus. RPS27a and RPL40 are important for the induction of p53, suggesting their role in the coordination of ribosome production, ubiquitin levels, and p53 signaling. Knockdown of both RPS27a and RPL40 induces p53 and is needed for rRNA production in MCF7 and LNCaP cells, indicating their cell type-dependent role in p53 signaling and consistent role in ribosome biogenesis.

Summary: In recent years, it has been discovered that many cytoplasmic proteins can dimerize while being translated. However, it was unclear whether this phenomenon also applies to integral membrane proteins. This study demonstrates that the inner membrane protein EmrE in Escherichia coli can indeed undergo cotranslational interactions, suggesting that membrane proteins can start to fold and dimerize during the membrane insertion process.
