Complete Genome Sequence of T4-Like Escherichia coli Bacteriophage HX01

Phage T4 is among the best-characterized biological systems (S. Kanamaru and F. Arisaka, Seikagaku 74: 131-135, 2002; E. S. Miller et al., Microbiol. Mol. Biol. Rev. 67: 86-156, 2003; W. B. Wood and H. R. Revel, Bacteriol. Rev. 40: 847-868, 1976). To date, several genomes of T4-like bacteriophages are available in public databases but without any APEC bacteriophages (H. Jiang et al., Arch. Virol. 156: 1489-1492, 2011; L. Kaliniene, V. Klausa, A. Zajanckauskaite, R. Nivinskas, and L. Truncaite, Arch. Virol. 156: 1913-1916, 2011; J. H. Kim et al., Vet. Microbiol. 157: 164-171, 2012; W. C. Liao et al., J. Virol. 85: 6567-6578, 2011). We isolated a bacteriophage from a duck factory, named HX01, that infects avian pathogenic Escherichia coli (APEC). Sequence and morphological analyses revealed that phage HX01 is a T4-like bacteriophage and belongs to the family Myoviridae. Here, we announce the complete genome sequence of phage HX01 and report the results of our analysis.

Physicochemical and functional properties of egg white peptide powders under different storage conditions

Summary: This study aims to evaluate the effects of different storage conditions on the physicochemical and functional properties of egg white peptide powders (EWPPs). The results showed that storage temperature and relative humidity significantly affected the antioxidant activity, secondary structure, solubility, and functional properties of EWPPs. Lower temperature and humidity storage conditions were beneficial for retaining the functional properties of EWPPs.

Summary: The diagnostic value of MDM2 RNA in situ hybridization (RNA-ISH) was evaluated in distinguishing low-grade osteosarcoma (LGOS) and compared with MDM2 FISH and IHC. MDM2 RNA-ISH showed high sensitivity and specificity, with higher sensitivity than IHC.

Study on long-term tracking of rail corrugation and the influence of parameters

Summary: Rail corrugation is a common form of damage in subway tracks. This study analyzed track wear at two sites on the Chengdu Metro and found corrugation on a 350 m radius track, but not on a 610 m radius curved track. A finite element model was used to predict corrugation and results showed that adjusting the material parameters of rail pads could reduce corrugation frequency.

The R-gt(F) value is 0.0535, the wR(ref)(F-2) value is 0.1239, and the temperature is 273(2) K.

Probabilistic machine learning approach to predict incompetent rock masses in TBM construction

Summary: This study proposes a new approach that combines a Bayesian boosting probabilistic model with two parallel machine learning methods for classifying the quality of rock masses. The results show that by using the binary classifier and Bayes boosting model, most of the predictions achieve a probability level greater than 0.9.

T-Toxin Virulence Genes: Unconnected Dots in a Sea of Repeats

Summary: In 1970, a devastating epidemic of Southern Corn Leaf Blight caused significant economic losses in the United States. The outbreak was caused by a highly virulent strain of the fungus Cochliobolus heterostrophus. This study used PacBio long-read sequencing to reveal the structure of the unique DNA responsible for the virulence of the pathogen and identified specific genetic elements involved in the integration of virulence-causing genes. These findings provide a foundation for future research on mechanisms of DNA acquisition from foreign sources.

Enzymatic polymerization of enantiomeric L-3,4-dihydroxyphenylalanine into films with enhanced rigidity and stability

Summary: The authors investigated the effect of the molecular chirality of (L)-3,4-dihydroxyphenylalanine on the improved structural and thermal stability of self-assembled films by tyrosinase-induced oxidative polymerization. The pure enantiomers showed altered kinetics and morphology upon co-assembly, allowing the fabrication of layer-to-layer stacked nanostructures and films with enhanced properties. The (L+D)-racemic mixtures exhibited different molecular arrangements and self-assembly mechanisms, resulting in stronger intermolecular forces and increased elastic modulus.

Optimal Scale-Free Small-World Graphs with Minimum Scaling of Cover Time

Summary: This article investigates the impact of cover time in random walks on network applications, showing that the minimum cover time in complete graphs is N log N. However, complete graphs cannot simulate real-world networks with small average degree and scale-free small-world properties, and therefore the behavior of cover time in these networks is not well understood. By conducting experiments and studying model networks, it is found that sparse networks with scale-free small-world structure also exhibit N log N cover time behavior. This finding deepens the understanding of cover time in real-world networks and has potential implications in algorithm design.

Synthesis and anti-α-glucosidase activity evaluation of betulinic acid derivatives

Summary: A series of betulinic acid derivatives were synthesized and evaluated for their anti alpha-glucosidase activities. All derivatives exhibited stronger activities than betulinic acid and acarbose. Compound 3q showed outstanding inhibitory activity, comparable to 1100 times stronger than acarbose. It functioned as a reversible and noncompetitive a-glucosidase inhibitor. The interaction of compound 3q with a-glucosidase caused conformational and secondary structure changes. Molecular docking simulation and physicochemical parameter assessment were also conducted.

Multi-omics data integration reveals the molecular network of dysregulation IQGAP2-mTOR promotes cell proliferation

Summary: As a tumor suppressor gene, IQGAP2 can affect cell proliferation in multiple tumor cell lines. However, the regulation network of cell proliferation caused by the deficiency of IQGAP2 in cells is still unclear. In this study, we used transcriptome, proteome, and phosphoproteome analyses to investigate the regulatory network of cell proliferation in IQGAP2 knockdown HaCaT and HEK293 cells. Our findings suggest that the dysregulation of the IQGAP2-mTOR molecular network leads to increased cell proliferation. We also demonstrated that IQGAP2 knockdown enhances the phosphorylation levels of AKT and S6K, resulting in increased cell proliferation. Additionally, inhibition of AKT and mTOR partially rescues abnormal cell proliferation by reducing hyperphosphorylation. These results indicate a potential connection between the mTOR signaling pathway and aberrant cell proliferation in IQGAP2 knockdown cells, providing a new therapeutic strategy for patients with IQGAP2 deficiency.

Summary: Biobanks containing FFPE tissues from animals and atomic-bomb survivors are important for understanding radiation exposure effects. Optical imaging is often the only option for these old samples, but it provides no information about radioactive microparticles. This study demonstrates the use of XFM to generate elemental maps and identify radioactive micro-particulates in 35-year-old FFPE canine specimens.

A novel maxillary transverse deficiency diagnostic method based on ideal teeth position

Summary: This study proposed a new diagnostic method for maxillary transverse deficiency and evaluated skeletal Class I and mild skeletal Class III groups. Pre-treatment data from 30 mild skeletal Class III and 30 skeletal Class I patients were collected and uploaded to the Emeiqi Case Management System to design ideal teeth positions. By measuring the width differences at these positions, it was found that there was a significant difference in the molar area between the two groups, which was mainly caused by the difference in mandible width.

Spatial scale analysis for the relationships between the built environment and cardiovascular disease based on multi-source data

Summary: This study employed multiscale geographically weighted regression to examine the relationship between 18 variables and cardiovascular disease. The results showed that multiscale geographically weighted regression was a better fit for the association analysis between the built environment and cardiovascular diseases. The built environment variables related to cardiovascular diseases can be divided into global variables and local variables, and at specific spatial scales, global variables had trivial spatial variation while local variables showed significant gradients.
