Summary: This study investigates the targets of ICP0 in HSV-1 infection and its roles in neuronal cells. Mass spectrometry analysis identified various proteins interacting with ICP0 in Neuro-2a and 293T cells. Further experiments confirmed that SNX9 and OTUD4 restrict the replication of ICP0-null virus in neuronal cells, and OTUD4 enhances the expression of type I interferon during infection with ICP0-null virus.

Summary: Herpes simplex virus 1 (HSV-1) utilizes the viral E3 ubiquitin ligase, ICP0, to counteract the antiviral effects of interferon (IFN)-beta by altering the cellular signaling and gene expression. A specific 77 amino acid region in the N-terminal half of ICP0 facilitates HSV-1's resistance to IFN-beta and may interact with the posttranslational modification SUMO.

Summary: This study aimed to determine whether HSV-1 can be transmitted through TNTs and to investigate the effect of inhibiting the Arp2/3 complex on the intercellular transmission of HSV-1. The results showed that HSV-1 can be transmitted through TNTs and that inhibiting the Arp2/3 complex reduces the number of TNTs and the spread of HSV-1. This finding provides new insights into the transmission mode of HSV-1 and suggests a potential new antiviral target.

Summary: Despite annual vaccination efforts, influenza continues to cause significant morbidity and mortality worldwide. To improve vaccine efficacy, researchers are exploring the use of antibodies that can activate immune cell effector functions, such as ADCC. Evidence suggests that ADCC is crucial for protection against influenza and other infectious diseases, leading to the development of innovative vaccine strategies such as using Delta gD-2 as a vector for a broadly protective influenza vaccine.

Summary: SLFN5 is identified as an ICP0 target that binds vDNA during HSV-1 Delta ICP0 infection. ICP0 mediates ubiquitination of SLFN5, leading to its degradation and repression of HSV-1 transcription. This study demonstrates that viral countermeasures can overcome SLFN antiviral activity.

Summary: In this study, we found that aMPV/C infection induces MAVS degradation through the ubiquitin-proteasome pathway at the post-translational level. Furthermore, specific amino acid sites were identified as pivotal in the formation of polyubiquitin chains during MAVS degradation. E3 ubiquitin ligases targeting MAVS were also identified, with RNF5 shown to be involved in MAVS degradation in aMPV/C-infected cells.

Summary: This study revealed that the activation of NLRP3 inflammasomes triggers Gasdermin D-dependent pyroptosis in mouse microglia, leading to inflammation caused by HSV-1. The expression levels of NLRP3 and IL-1 beta were significantly increased in a mouse model of herpes simplex encephalitis. These findings provide potential treatment targets for viral inflammation.

Summary: Endoplasmic reticulum-associated degradation (ERAD) is a protein quality control pathway that removes misfolded proteins from the ER. Overexpression of ERAD factors inhibits the degradation of specific substrates. This study reveals a potential retrotranslocation mechanism independent of ERAD E3 complexes.

Summary: Neurotropic viruses such as herpes simplex virus (HSV) can infect neurons and cause severe diseases. HSV-induced neuronal cell death is mediated by gasdermin E (GSDME) and involves endoplasmic reticulum stress, caspase activation, and mitochondria-dependent apoptosis. The necrotic neurons release alarmins, triggering inflammatory responses in microglia.

Summary: This study investigated the interaction of human parainfluenza viruses type 1 (hPIV1) and type 3 (hPIV3) with human airway cells. The results showed that hPIV1 infection downregulated cholesterol-related genes, induced ISGs stronger than hPIV3, and led to ubiquitination and degradation of 3-hydroxy-3-methylglutaryl-coenzyme A reductase, disrupting formation of cholesterol-rich lipid rafts in infected cells. These findings suggest that hPIV1 has the ability to regulate cholesterol biogenesis, which along with ISGs, contributes to the establishment of quiescent infection.

Summary: This study characterized the clinical markers of herpes simplex virus (HSV) infection among patients with pemphigus vulgaris. It was found that male sex, presence of fissures, hemorrhagic crusts, erosions with angulated margins, linear erosions, and raised erythrocyte sedimentation rate were significantly associated with HSV infection. Hemorrhagic crusts and linear erosions were identified as independent predictors of HSV infection.
