Summary: Human Cytomegalovirus Immediate-Early protein 1 (IE1) is a multifunctional protein that interacts with the SUMO signaling pathway, specifically binding to SUMO1 through SUMO-Interacting Motifs (SIMs). IE1 plays a crucial role in inhibiting PML SUMOylation and transactivating viral promoters, with the non-covalent interaction with SUMO contributing to the latter activity. Transactivation and inhibition of PML SUMOylation are independent functions of IE1.

Summary: Pentamidine disrupts the S100P-53 complex formation by targeting the shared binding site of S100P and p53, leading to re-activation of the p53-21 pathway and inhibition of cancer cell proliferation. This suggests a potential new avenue for cancer therapy targeting the S100P-p53 interaction.

Summary: Here, we characterized the interaction between p53 and FOXO4 using NMR, chemical cross-linking, and analytical ultracentrifugation. Our findings highlight the importance of this interaction for the stability of the p53:FOXO4 complex and shed light on the intertwined functions of p53 and FOXO4 in cellular homeostasis, longevity, and stress response.

Summary: The study investigates the interaction between β-catenin and CBP/p300, and determines that the C-terminal region of β-catenin binds to the TAZ1 and TAZ2 domains of CBP/p300. The results provide insights into the gene regulation mechanism of β-catenin and offer a framework for developing methods to block β-catenin dependent signaling.

Summary: The study revealed that the FGFR/RACK1 complex regulates senescence in LSCC cells by promoting the degradation of p53 through the formation of a complex with MDM2. Inhibition of FGFR or knockdown of RACK1 induced cell senescence, while inhibitors targeting MDM2 and p53 cooperatively inhibited colony formation and metastasis of LSCC cells. This understanding could have important implications for the treatment of LSCC.

Summary: The study reveals that CCAAT/enhancer binding protein alpha (CEBPA) interacts with the long splice isoform DNMT3A, inhibiting its activity by blocking its access to DNA substrate. Recurrent tumor-associated CEBPA mutations disrupt the DNMT3A association, causing aberrant DNA methylation. This study sheds light on the functional difference between DNMT3A isoforms and the regulation of de novo DNA methylation, providing a potential therapeutic strategy for CEBPA-mutated leukemia using DNA-hypomethylating agents.

Summary: This study reveals that human cytomegalovirus (HCMV) inhibits the MR1 pathway and disrupts the MR1:MAIT cell axis through the viral protein gpUS9. The interaction between this virus and MAIT cells in the context of viral infection is not well characterized.

Summary: This study identified a novel interaction between TP53(R273H) and BCAR1, where BCAR1 translocates from the cytoplasm to the nucleus and binds to TP53(R273H) in a manner dependent on SRC family kinases (SFKs), promoting cancer cell invasion. High BCAR1 expression was associated with a poorer prognosis among patients with mutant TP53, highlighting the potential therapeutic approach of disrupting the TP53(R273H)-BCAR1 binding in TP53(R273H)-harbouring cancer patients.

Summary: UL36 encoded by human cytomegalovirus not only inhibits apoptosis but also suppresses immune signaling by targeting IRF3 directly. Although UL36-mediated inhibition of apoptosis enhances immune signaling, its immunosuppressive activity counterbalances this effect.

Summary: This study reveals the important role of long non-coding RNA CASC11 in the progression of prostate cancer. CASC11 promotes tumor proliferation and migration by promoting the cell cycle and competitively interacting with YBX1 to block the p53 pathway. The use of poly (beta-amino ester)/si-CASC11 nanoparticles enhances the antitumor effect by inhibiting CASC11 expression. This study provides a potential therapeutic strategy for the treatment of prostate cancer patients.
