The Cellular Antiviral Protein Viperin Is Attenuated by Proteasome-Mediated Protein Degradation in Japanese Encephalitis Virus-Infected Cells

Viperin is identified as an antiviral protein induced by interferon (IFN), viral infections, and pathogen-associated molecules. In this study, we found that viperin is highly induced at the RNA level by Japanese encephalitis virus (JEV) and Sindbis virus (SIN) and that viperin protein is degraded in JEV-infected cells through a proteasome-dependent mechanism. Promoter analysis revealed that SIN induces viperin expression in an IFN-dependent manner but that JEV by itself activates the viperin promoter through IFN regulatory factor-3 and AP-1. The overexpression of viperin significantly decreased the production of SIN, but not of JEV, whereas the proteasome inhibitor MG132 sustained the protein level and antiviral effect of viperin in JEV-infected cells. Knockdown of viperin expression by RNA interference also enhanced the replication of SIN, but not that of JEV. Our results suggest that even though viperin gene expression is highly induced by JEV, it is negatively regulated at the protein level to counteract its antiviral effect. In contrast, SIN induces viperin through the action of IFN, and viperin exhibits potent antiviral activity against SIN.

Summary: In this study, a population-based hiPSC drug screening platform is established to assess toxicity. Thirteen HLA-homozygous super donors are identified after recruiting 1,000 healthy donors and screening for high-frequency HLA haplotypes, representing at least 477,611,135 of the global population. The differentiated hiPSCs into cardiomyocytes and neurons demonstrate their utility in a high-throughput toxicity screen. Dose-dependent toxicity and functional modulation of hit compounds are validated, and reproducible in vivo drug toxicity results using mouse models are shown. This study shows the feasibility of using a population-based hiPSC drug screening platform to study inter population differences in drug toxicity and adverse drug reactions.

Human Platelet Lysate Induces Antiviral Responses against Parechovirus A3

Summary: This study found that the infection of Parechovirus type-A3 (PeV-A3) was reduced in human platelet lysate (hPL)-cultured cells compared to fetal bovine serum (FBS)-cultured cells. Mechanistic analysis revealed that hPL stimulates the type I interferon (IFN) antiviral pathway, resulting in the activation of related transcription factors and the expression of antiviral genes. Enhanced PeV-A3 replication was observed in hPL-cultured cells treated with STAT-1 inhibitor and STAT1 shRNA. These findings suggest that hPL may have potential antiviral effects.

Summary: Ciclesonide blocks coronavirus-induced cytokine production by increasing I kappa B alpha protein levels and decreasing p65 nuclear translocation, and it can be used in the treatment of COVID-19 patients. The combination of Ciclesonide and dbq33b synergistically decreases cytokine levels and inhibits coronavirus replication. Therefore, this combination therapy is a promising treatment option for COVID-19 patients with high viral loads and NF-kappa B-mediated cytokine storms.
