Summary: Downregulation of multiple tumor suppressor genes is crucial in cancer formation, with epigenetic modifications playing a significant role. Using nanopore sequencing technology, the study identified potential TSGs in HCC and demonstrated the tumor suppressor function of GCK in inhibiting HCC cell proliferation through lactate accumulation-induced energy crisis. These findings offer valuable insights for further understanding tumorigenesis in human cancer.

Summary: This study reveals that NFAT5 directly regulates the expression of Ranbp3l by binding its promoter, leading to tumor-like phenotype and altered cell behavior and gene expression in RANBP3L-deficient cells. Loss of RANBP3L may promote the development and progression of renal cell carcinoma.

Summary: The protein expression of MAPK10 is significantly downregulated in HCC patients compared to normal tissues, and its negative expression is correlated with advanced tumor stage, more microsatellite nodules, higher serum AFP, and shorter overall survival time in HCC patients. MAPK10 is frequently methylated in HCC, and its methylation is associated with silenced or downregulated expression.

Summary: This study found that mutations in tumor suppressor genes can affect the tumor immune microenvironment, particularly in lung squamous cell carcinoma, where these gene mutations can suppress infiltrating immune and stromal cells, leading to a decrease in immune gene expression. Patients with non-mutated tumor suppressor genes are more likely to have inflamed tumors and respond positively to immune checkpoint blockade therapy.

Summary: This study reveals that SETD2 inhibits beta-catenin activity during the transition from polycystic kidney disease (PKD) to renal cell carcinoma (RCC) by competing with beta-catenin for binding promoters of target genes, ultimately enhancing Wnt/beta-catenin signaling and promoting epithelial-to-mesenchymal transition and tumorigenesis.

Summary: The histone H3K4 methyltransferase SET1B plays a crucial role in the transcriptional activity of HIF target genes, and its loss results in impaired cell growth and tumor formation. Experimental findings demonstrate that SET1B accumulates on chromatin in hypoxia and is recruited to HIF target genes by the HIF complex.

Summary: KDF1 has been identified as a key regulator of epidermal proliferation and differentiation, but its involvement in malignancy remains unknown. This study explored the role of KDF1 in clear cell renal cell carcinoma (ccRCC) through analyzing its expression levels and functions. The results suggest that KDF1 may act as a tumor suppressor in ccRCC by inhibiting proliferation and invasion of cancer cells.

Summary: MOF, a histone acetyltransferase, is downregulated in renal cell carcinoma (RCC), and its downregulation is associated with advanced histological grade, pathologic stage, and distant metastasis of RCC. Ectopic expression of MOF inhibits cell proliferation and promotes cell apoptosis, and suppresses cell migration through inhibiting epithelial-mesenchymal transition. Mechanistically, MOF transcriptionally upregulates the expression of SIRT1, leading to attenuated STAT3 signaling. Silencing SIRT1 restores the biological functions induced by MOF overexpression.

Summary: This study reveals that TRIM3 functions as a tumor suppressor in non-small cell lung cancer (NSCLC) by promoting cell death and inhibiting tumorigenesis. TRIM3 directly interacts with SLC7A11/xCT, leading to its degradation and inhibition of NSCLC development. Low TRIM3 expression is associated with a worse prognosis in NSCLC.

Summary: CpG-island methylation plays a role in ovarian cancer progression and is associated with survival outcomes. Differential promoter methylation of tumor suppressor genes (TSGs) was observed, with RASSF1A hypomethylation and increased aggressiveness in mesenchymal-like OCSPCs. Overexpression of CCND2 and CDKN2B reduced the aggressiveness of mesenchymal-like OCSPCs and may serve as potential therapeutic targets for EOC.

Summary: This study reveals that HAND1 is frequently downregulated in gastric cancer due to promoter methylation, and its low expression is correlated with tumor progression and poor prognosis. High expression of HAND1 is associated with higher 5-year overall survival rates in gastric cancer patients. HAND1 inhibits gastric cancer cell growth and migration through induction of endoplasmic reticulum (ER) stress and apoptosis. The methylation status of HAND1 promoter could be a potential biomarker for gastric cancer.

Summary: Peptides/small proteins encoded by noncanonical open reading frames (ORF) of previously claimed non-coding RNAs have important biological functions but are largely uncharacterized. In this study, we identified and validated a small protein SP0495 as a novel tumor suppressor, frequently inactivated by promoter methylation in multiple tumors. SP0495 functions as a phosphoinositide-binding protein, regulating AKT signaling activation and autophagy. It inhibits tumor cell growth and induces apoptosis, cell cycle arrest, senescence, and autophagy through multiple mechanisms, including the modulation of phosphoinositides turnover and autophagic/proteasomal degradation of autophagy regulators. SP0495 has the potential to be a biomarker for cancer diagnosis and prognosis.

Summary: Selenite is found to induce both apoptosis and ferroptosis in lung cancer cells, especially those with EGFR and KRAS mutations. It activates the p38-ATF4-DDIT3 pathway for ferroptosis induction and influences DNA methylation machinery. Combining selenite with osimertinib or adagrasib enhances the therapeutic effect.
