Low expression of LOC285194 is associated with poor prognosis in colorectal cancer

Background: The long non-coding RNAs (lncRNAs) study has gradually become one of the hot topics in the field of RNA biology. One lncRNA which has attracted attention is LOC285194, a lncRNA demonstrated the potential tumor-suppressor role in osteosarcoma. The aim of this study was to examine the expression of LOC285194 in colorectal cancer (CRC) patients and to investigate the relationship between this lncRNA levels and existing clinicopathologic parameters and patient survival. Methods: The expression of LOC285194 was detected by quantitative real-time polymerase chain reaction in pairs of tumorous and adjacent normal tissues of 81 colorectal cancer patients with a follow-up of 5 years, as well as in three colorectal cancer cell lines and normal intestinal mucous cell line. Then, we analyzed the potential relationship between this lncRNA levels in tumor tissues and existing clinicopathological features of CRC, and clinical outcome. Results: The relative expression levels of LOC285194 was significantly lower in tumor tissues (p < 0.001) and colorectal cancer cell lines compared with adjacent normal tissues and normal intestinal mucous cell line. In addition, low expression of LOC285194 was correlated with larger tumor size (p = 0.015), higher tumor stage (p = 0.034), and more distant metastasis (p = 0.046). Kaplan-Meier analysis indicated that patients with low LOC285194 expression had a poor disease free survival (p = 0.010). Moreover, multivariate analysis showed that decreased expression of LOC285194 was an independent predictor of disease-specific survival. Conclusion: Our data indicate that LOC285194 might be a novel prognostic indicator in colorectal cancer and may be a potential target for diagnosis and gene therapy.

A Lipid Metabolism-Based Seven-Gene Signature Correlates with the Clinical Outcome of Lung Adenocarcinoma

Summary: In this study, a prognostic prediction model for patients with LUAD was developed based on the expression profiles of lipid metabolism-related genes. Three molecular subtypes were identified in LUAD, and a seven-gene signature was confirmed as an independent prognostic factor. The established gene signature can be used as a new prognostic marker for predicting survival in patients with LUAD.

The role of cancer-associated fibroblasts in tumorigenesis of gastric cancer

Summary: Despite progress in cancer treatment, gastric cancer (GC) still has a poor prognosis. Recent research has shown that the malignant behavior of cancer is not only caused by tumor cells, but also mediated by the activity of cancer stroma and regulated by molecular networks in the tumor microenvironment (TME). Cancer-associated fibroblasts (CAFs) are abundant in the stroma and play a significant role in the malignant development of GC. CAFs can modulate the biological properties of tumor cells through various mechanisms, affecting the initiation and progression of GC. However, there is heterogeneity in the cellular origins, phenotypes, and functions of CAFs in the TME of GC, and the molecular mechanisms are not well understood.

Evolutionary Trajectories of Primary and Metastatic Pancreatic Neuroendocrine Tumors Based on Genomic Variations

The study provides novel insight into genetic changes in metastatic tumors of PanNETs.

Transcriptomic characterization and construction of M2 macrophage-related prognostic and immunotherapeutic signature in ovarian metastasis of gastric cancer

Summary: Transcriptomic sequencing and follow-up analyses revealed unique molecular and immune features of paired normal mucosae, primary and ovarian metastatic tumors. A novel OM- and M2 macrophage-related signature, M2GO, was established and served as a promising prognostic and immunotherapeutic biomarker in gastric cancer patients. The M2GO score was significantly associated with clinical outcomes, molecular and immune features, and differential responses to immunotherapy.

High PROX1 expression combined with low SIRT3 expression predicts poor prognosis in CRC patients.

Enhanced BCAT1 activity and BCAA metabolism promotes RhoC activity in cancer progression

Summary: In this study, the researchers identified a relationship between branched-chain amino acid metabolism and cell growth and motility through RhoC signaling. They found that enhanced BCAT1 activity, caused by a mutation enriched in gastric cancer, leads to increased production of a metabolite that activates RhoC signaling. The study also found that the BCAA-derived metabolite can directly bind to the small GTPase protein RhoC and enhance its activity. The researchers also discovered that the drug candesartan can inhibit BCAT1 activity, leading to decreased RhoC activity and cancer cell motility.

Summary: The present study investigates the mechanism of gambogenic acid (GNA), a natural compound, on apoptosis and ferroptosis in colorectal cancer (CRC). The results show that GNA inhibits tumor growth by suppressing the proliferation of CRC cell lines and induces ferroptosis by activating endoplasmic reticulum stress and ROS production. Furthermore, GNA regulates iron metabolism to further induce ferroptosis.
