Summary: This study identified a rare PTGS1 variant associated with a bleeding phenotype and alterations in platelet reactivity and eicosanoid production in a large consanguineous family. The specific loss of platelet COX-1 activity was demonstrated for the first time in humans, providing important insights into platelet function and eicosanoid metabolism. Despite the absence of thromboxane A(2) formation by platelets, urinary thromboxane A(2) metabolites were in the normal range, suggesting the need for caution when using them as markers of in vivo platelet function.

Summary: The study demonstrates a synergistic relationship between P2Y(12) receptor antagonists and adenosine signaling, with their efficacy partly dependent on the presence of endogenous adenosine. Despite reported differences in their effects upon adenosine reuptake, both prasugrel and ticagrelor show similar synergistic effects on platelet inhibition in the presence of adenosine or NECA.

What can we learn from senescent platelets, their transcriptomes and proteomes?

Summary: Research on the natural aging process of platelets has attracted much attention in recent years. Previous studies have shown an association between the proportion of newly formed platelets in the circulation and the risk of thrombosis. However, these observations were mainly demonstrated in patient groups with underlying systemic changes. Recent technological advances have allowed in-depth analysis of differently aged platelets from healthy individuals, revealing extensive changes in the transcriptome and proteome of aged platelets. These changes result in deteriorated platelet function, impairing their ability to participate in hemostatic responses.

Summary: Arterial tonometry and vascular calcification measures are valuable in assessing cardiovascular disease (CVD) risk. This study examined the relationship between arterial tonometry, platelet function, arterial calcium, and aortic diameter. The results suggest that higher arterial calcium may be associated with increased platelet reactivity, while larger aortic diameter may be linked to lower platelet reactivity. These findings indicate the potential interplay between subclinical atherosclerosis, platelet activation, and vascular structure.
