Differential Expression of Extracellular Matrix Constituents and Cell Adhesion Molecules between Malignant Pleural Mesothelioma and Mesothelial Hyperplasia

Introduction: Malignant pleural mesothelioma (MPM) is a highly aggressive neoplasm associated with asbestos exposure. Currently, the molecular mechanisms that induce MPM development are still unknown. The purpose of this study was to identify new molecular biomarkers for mesothelial carcinogenesis. Methods: We analyzed a panel of 84 genes involved in extracellular matrix remodeling and cell adhesion by polymerase chain reaction (PCR) array in 15 samples of epithelioid mesothelioma and 10 samples of reactive mesothelial hyperplasia (MH; 3 of 25 samples were inadequate for mRNA analysis). To validate the differentially expressed genes identified by PCR array, we analyzed 27 more samples by immunohistochemistry, in addition to the 25 samples already studied. Results: Twenty-five genes were differentially expressed in MPM and MH by PCR array. Of these we studied matrix metalloproteinase 7 (MMP7), MMP14, CD44, and integrin, alpha3 expression by immunohistochemistry in 26 epithelioid MPM and 26 MH samples from the entire series of 52 cases. We observed higher MMP14 and integrin, alpha3 expression in MPM samples compared with MH samples (p = 0.000002 and p = 0.000002, respectively). Conversely, CD44 expression was low in most (57.7%) mesothelioma samples but only in 11.5% of the MH samples (p = 0.0013). As regards MMP7, we did not observe differential expression between MH and MPM samples. Conclusions: We have extensively studied genes involved in cell adhesion and extracellular matrix remodeling in MPM and MH samples, gaining new insight into the pathophysiology of mesothelioma. Moreover, our data suggest that these factors could be potential biomarkers for MPM.

Adipose tissue in COVID-19: detection of SARS-CoV-2 in adipocytes and activation of the interferon-alpha response

Summary: In fatal cases of COVID-19, a significant proportion of adipocytes have been found to contain the SARS-CoV-2 nucleocapsid antigen, indicating direct infection of subcutaneous fat cells by the virus. Infection seems to activate the IFN alpha pathway and attract infiltrating leukocytes. Due to the large numbers of adipocytes in adults, adipose tissue serves as a significant reservoir for SARS-CoV-2 and an important source of inflammatory mediators.

New Insights in Pleural Mesothelioma Classification Update: Diagnostic Traps and Prognostic Implications

Summary: The 2021 WHO classification of tumors of the pleura introduces significant changes in the codification of mesothelioma, going beyond the three current histological subtypes and including advancements in nuclear grading, architectural patterns, cytological features, and stromal features. The new classification also recognizes mesothelioma in situ as a diagnostic category and emphasizes the importance of immunohistochemistry and fluorescence in situ hybridization (FISH) in establishing the diagnosis of epithelioid mesothelioma. These changes have implications for prognosis and patient management, but also pose challenges in the differential diagnosis of pre-invasive tumors.

Summary: This study analyzed the clinicopathological characteristics of 989 patients with tall-cell variant of papillary thyroid carcinoma (TCPTC) and found that this type of cancer is not exclusively found in older patients. The study also revealed higher rates of local recurrence in the younger and older age groups, and a higher rate of distant recurrence in older patients. The results suggest that the prognosis of TCPTC is age-dependent and further studies are needed.

Dynamic profiling of the immune tumor microenvironment in locally advanced gastric cancer treated with perioperative chemotherapy

Summary: In this study, the impact of FLOT on the immune tumor microenvironment (TME) in gastric cancer (GC) was investigated. The results showed significant changes in immune gene expression after FLOT treatment, particularly in genes related to cytotoxicity, T-cell functions, complement system, tumor necrosis factor-superfamily, cell cycle, and regulation. Moreover, an association between changes in immune cell profiling and tumor regression was observed, suggesting a potential role of immune response in treatment outcomes.
