Summary: This study found that MTA1 gene has significantly increased cytoplasmic expression in non-keratinizing squamous cell carcinoma and vulvar carcinoma of the condylomatous type compared to keratinizing squamous cell carcinoma and vulvar carcinoma of the verrucous type. High nuclear expression of MTA1 was associated with advanced tumor stage and higher FIGO grading. In addition, tumor samples without nuclear expression of MTA1 had significantly lower disease-free survival.

Summary: PRAME is a tumor antigen that can distinguish between benign nevi and malignant melanomas, and is also expressed in non-melanocytic tumors. This study found that PRAME expression is correlated with higher metastatic risk and lower metastasis-free survival in primary uveal melanoma. It suggests that PRAME can be included in immunohistochemical panels to predict metastatic risk and stratify patient outcomes.

Summary: This study systematically explored a new mechanism by which FOXP3 inhibits breast cancer metastasis through the FOXP3-MTA1 pathway. It was found that FOXP3 downregulated MTA1 expression in breast cancer cells by directly inhibiting MTA1 promoter activity, affecting the invasion and metastasis abilities of breast cancer cells. Clinical data showed a significant negative correlation between the expression levels of FOXP3 and MTA1 in breast cancer.

Summary: This study investigated the impact of MTA1, SOX4, and EZH2 on the prognosis of patients with ESCC. The results suggest that MTA1 may serve as a prognostic and diagnostic marker for ESCC, and that MTA1-SOX4 signaling is associated with prognosis in ESCC. However, no significant association was found between SOX4 and EZH2 in this study, indicating the need for further investigation into the function of the MTA1-SOX4-EZH2 axis and the interactions of the proteins involved.

Summary: For melanoma treatment, early diagnosis and complete resection of the primary tumor are crucial, and the detection of factors related to metastasis is also essential. This study investigated 30 Japanese patients with Stage I or II melanoma to identify factors statistically associated with metastasis. The analysis revealed that older age at onset and positive immunohistochemical expressions of Nestin and Fascin were significantly related to metastasis. Double immunofluorescence staining was performed to observe Nestin and Fascin expression at different stages. This is the first study to report the presence of Nestin and Fascin double-positive melanoma cells.

Summary: Our study aimed to explore the genes correlated with metastasis in Medulloblastoma (MB) through analyzing differentially expressed genes and pathways. We identified 10 genes, including FAIM2, that were significantly associated with survival of MB patients. FAIM2 expression was decreased in metastatic MB samples, and it harbored mutations, amplifications, and deletions. Pathway analysis showed that FAIM2 high expression group had significantly activated and inhibited pathways compared to FAIM2 low expression group. FAIM2 was tightly correlated with metastasis in MB patients and low expression was associated with poor prognosis.

Summary: Chordoma patients may benefit from immunotherapy, as their unique immune microenvironment shows PD-L1+ immune cells playing a negative prognostic role. Higher CD8, presence of PD-L1+ immune cells, and higher vascular density are adverse prognostic factors, with only PD-L1+ immune cells retaining prognostic significance in multivariate analysis. Further studies in this field and the potential beneficial role of immunotherapy in chordoma patients are warranted.
