Summary: Auto-allo-SCT may improve progression-free survival and complete response compared to tandem auto-SCT in patients with HRMM, but it does not significantly improve overall survival. Patients undergoing auto-allo-SCT have a higher risk of transplant-related mortality and non-relapse mortality.

Summary: The results of this study suggest that BUMEL-based conditioning regimen can lead to longer progression-free survival (PFS) and similar overall survival (OS) compared to HDMEL in patients undergoing auto-HSCT for multiple myeloma. While there were no significant differences in treatment response and duration of engraftment between the two groups, the BUMEL group had lower rates of gastrointestinal toxicity, whereas the HDMEL group experienced less mucositis and infection.

Summary: This study conducted a comprehensive systematic review and meta-analysis to investigate the efficacy and safety of autologous hematopoietic stem-cell transplantation (AHSCT) in patients with multiple sclerosis (MS). The results showed that AHSCT led to a significant decrease in Expanded Disability Status Scale (EDSS) score and annualized relapse rate. Most patients remained relapse-free after AHSCT and the overall survival rate was high.

Summary: Front-line high-dose therapy followed by autologous stem cell transplantation (ASCT) is still a beneficial approach for newly diagnosed multiple myeloma (MM) patients in the era of novel agents. However, the benefit of HDT/ASCT in terms of progression-free survival (PFS) and overall survival (OS) remains inconsistent. We conducted a systematic review and meta-analysis to evaluate the advantages of upfront HDT/ASCT and identified factors influencing survival outcomes.

Summary: This study investigated taste changes in patients undergoing autologous hematopoietic cell transplantation using chemical gustometry. The study found that taste testing was feasible, taste changes peaked when blood counts were lowest, and most patients recovered their taste by 100 days after transplantation. Taste changes correlated with lower food intake and higher levels of chemotherapy. Future work will focus on using personalized chemotherapy doses to reduce taste changes and matching patients' taste preferences with their diets.

Summary: This study systematically summarized the quality and evidence of multiple meta-analyses on the treatment of MM, including autologous stem cell transplantation. The findings revealed that the included meta-analyses were affected by possible bias and lacked protocol and registration. The methodological quality of the meta-analyses ranged from very low to moderate, and most outcome indicators were of low quality.

Summary: Multiple Myeloma (MM) is a common cancer of the bone marrow that remains incurable despite advancements in therapy. While Daratumumab has improved survival rates, heavily pretreated patients who progress on this treatment have poor clinical outcomes. High-dose chemotherapy followed by autologous stem cell transplantation (ASCT) may be a viable treatment option for selected patients who are refractory to Daratumumab.

Summary: Multiple sclerosis (MS) is a chronic disease of the central nervous system that can lead to lifelong disability. Despite various treatment options, some patients still face severe symptoms and long-term disability risk. Recent studies have shown that autologous hematopoietic stem cell transplantation (AHSCT) is a highly effective and relatively safe treatment option for highly active MS.

Summary: AutoHSCT in MM patients leads to significant changes in NK cell subsets and phenotype, with patients showing lower frequencies of the mature NKG2A-CD57+ NK cell subset experiencing better clinical outcomes. This suggests that the degree of NK cell maturation post-autoHSCT affects the disease progression and treatment outcomes in MM patients.

Summary: The study aimed to evaluate the safety and efficacy of tandem autologous hematopoietic cell transplantation in patients with multiple myeloma. The results showed that the conditioning with TMI in sequence with high-dose melphalan had very good toxicity profile with encouraging early efficacy, with overall and progression-free survival probabilities at 5 years being 74% and 55%, respectively.

Summary: This study provides insights into the understanding and response of multiple myeloma (MM) patients to treatment changes during the COVID-19 pandemic. The research identifies beliefs and perceptions regarding autologous stem cell transplantation (ASCT), the impact of COVID-19 on fear and anxiety levels, emotional responses to treatment disruption or delay, perceptions of care, and the importance of social support. These findings will help guide adjustments in healthcare service and counseling approaches to align with patient priorities and expectations.

Summary: The effect of autologous hematopoietic stem cell transplantation (auto-HSCT) versus conventional chemotherapy or allogeneic hematopoietic stem cell transplantation (allo-HSCT) on the survival of patients with advanced follicular lymphoma (FL) is uncertain. This study included 13 studies and found that auto-HSCT improved overall survival (OS), progression-free survival (PFS), and event-free survival (EFS) compared with conventional chemotherapy. In addition, auto-HSCT resulted in longer OS and lower non-relapse mortality (NRM) compared with allo-HSCT.

Summary: The optimal consolidation strategy for newly diagnosed primary central nervous system lymphoma (PCNSL) remains controversial between autologous hematopoietic cell transplant (auto-HCT) and whole-brain radiotherapy (WBRT). A systematic review and meta-analysis of randomized controlled trials was conducted, revealing no significant differences in survival outcomes between auto-HCT and WBRT consolidation, but a higher degree of neurocognitive decline associated with WBRT. Individualized decision-making based on age, frailty, and co-morbidities is recommended.

Summary: Autologous hematopoietic cell transplantation has been the standard approach for patients with relapsed/refractory diffuse large B cell lymphoma. However, the introduction of CAR T cell therapy has changed the management of these patients, especially with the approval of CD19-directed CAR-T therapy in the second-line setting for high-risk groups. The American Society of Transplantation and Cellular Therapy has formulated consensus recommendations to address the lack of consensus on the role and sequencing of HCT and cellular therapies in DLBCL.

Summary: Chimeric antigen receptor T-cell therapy (CAR-T) is a significant advancement in managing aggressive relapsed or refractory B-cell lymphomas, but frequent relapses remain a major challenge. There is substantial variability in assessing and managing post-CAR-T failures across transplantation and cellular therapy programs. A survey was conducted among lymphoma, transplantation, and cellular therapy physicians in the United States to determine their practice patterns for the detection, diagnosis, and management of CAR-T failure in diffuse large B-cell lymphoma. The survey revealed significant differences in surveillance, diagnosis, and treatment approaches for CAR-T failure.
