Synthesis of Chiroptical Molecular Switches by Pd-Catalyzed Domino Reactions

New photochromic switches based on helical alkenes can quickly and efficiently be accessed by Pd-catalyzed domino reactions using a modular approach; this allows a wide variability in product formation with the advantages of a convergent synthetic route. The alkenes have been synthesized in excellent enantioselectivity and their switching properties assessed by stimulation with nanosecond laser pulses at two different wavelengths in over 1000 switching cycles.

Summary: The study identified 37 compounds that bind to the SARS-CoV-2 main protease through a high-throughput x-ray crystallographic screen of two repurposing drug libraries. Among these compounds, one peptidomimetic and six nonpeptidic compounds showed antiviral activity in cell-based viral reduction assays at nontoxic concentrations. The identification of two allosteric binding sites presents potential targets for drug development against SARS-CoV-2.

Protecting redesigned supercharged ferritin containers against protease by integration into acid-cleavable polyelectrolyte microgels

Summary: The integration of ferritin containers into microgels can protect ferritin against degradation by protease and a stimuli-responsive microgel system can be designed by employing an acid-degradable crosslinker. Experimental results showed that around 80% of the ferritin containers were integrated into microgels with approximately 85% and 50% released in buffer pH 2.5 and 4.0, respectively.

Encapsulation of Gold Nanoparticles into Redesigned Ferritin Nanocages for the Assembly of Binary Superlattices Composed of Fluorophores and Gold Nanoparticles

Summary: In this study, protein nanocages were used as nanoscale building blocks for the assembly of gold nanoparticles and fluorescent molecules, resulting in the synthesis of binary nanomaterials with highly ordered structures. The cargo was encapsulated into the protein nanocages using a dis- and reassembly approach. The strong exciton-plasmon coupling between the encapsulated fluorophores and gold nanoparticles was observed in the binary crystals.

Overcoming Non-Specific Interactions for Efficient Encapsulation of Doxorubicin in Ferritin Nanocages for Targeted Drug Delivery

Summary: Due to its pharmacological properties, ferritin (Ftn) is used as a drug delivery vehicle to reduce the cardiotoxicity of doxorubicin (DOX) in chemotherapy. However, DOX encapsulation in Ftn faces challenges such as precipitation and low protein recovery yield. A new strategy using cysteine-maleimide conjugation for DOX encapsulation in Ftn is proposed and shown to be more efficient. The surface charges of Ftn variants affect the cytotoxicity, DOX loading, and cellular uptake mechanism.

Optical Properties of Metacrystals Based on Protein Nanocages

Summary: The optical properties of 3D metacrystals made of gold nanoparticles in protein nanocages are investigated. These metacrystals have good structural and optical quality with sizes in the tens of micrometers range. Through microspectroscopy measurements and model calculations, it is shown that the metacrystals exhibit plasmonic absorption in the green wavelength range and are mostly transparent in the red and infrared ranges. By using empty nanocages as placeholders in the metacrystal lattice, the absorption strength of the metamaterial can be controlled. Measurements on a pyramidal metacrystal demonstrate deflection of visible light, indicating anomalous refraction at short wavelengths and normal refraction at long wavelengths. The refractive dispersion is attributed to the optical dispersion relation of the plasmonic metamaterial.
