Isolation, structure, and antibacterial activity of philipimycin, a thiazolyl peptide discovered from Actinoplanes philippinensis MA7347

Bacterial resistance to antibiotics, particularly to multiple drug resistant antibiotics, is becoming cause for significant concern. The only really viable course of action is to discover new antibiotics with novel mode of actions. Thiazolyl peptides are a class of natural products that are architecturally complex potent antibiotics but generally suffer from poor solubility and pharmaceutical properties. To discover new thiazolyl peptides potentially with better desired properties, we designed a highly specific assay with a pair of thiazomycin sensitive and resistant strains of Staphylococcus aureus, which led to the discovery of philipimycin, a new thiazolyl peptide glycoside. It was isolated along with an acid-catalyzed degradation product by bioassay-guided fractionation. Structure of both compounds was elucidated by extensive application of 2D NMR, 1 D TOCSY, and HRESIFT-MS/MS. Both compounds showed strong antibacterial activities against Gram-positive bacteria including MRSA and exhibited MIC values ranging from 0.015 to 1 mu g/mL. Philipimycin was significantly more potent than the degradation product. Both compounds showed selective inhibition of protein synthesis, indicating that they targeted the ribosome. Philipimycin was effective in vivo in a mouse model of S. aureus infection exhibiting an ED(50) value of 8.4 mg/kg. The docking studies of philipimycin suggested that a part of the molecule interacts with the ribosome and another part with Pro(23), Pro(22), and Pro(26) of L11 protein, which helped in explaining the differential of activities between the sensitive and resistant strains. The design and execution of the bioassay, the isolation, structure, in vitro and in vivo antibacterial activity, and docking studies of philipimycin and its degradation product are described.

Parental Depression Symptoms and Internalizing Mental Health Problems in Autistic Children

Summary: Autistic youth are at risk for internalizing mental health problems, such as depression and anxiety. Parents of autistic youth also experience higher levels of depression compared to parents of typically developing children. This study reveals bidirectional associations between parent depression symptoms and the internalizing problems of autistic youth. Future research should focus on exploring the genetic and environmental pathways that link parent depression and internalizing problems in autistic youth.

Summary: Memnoniella is a fungal genus that produces a variety of biologically active compounds. A specific extract from Memnoniella dichroa CF-080171 showed potent activity against Plasmodium falciparum 3D7 and Trypanosoma cruzi Tulahuen parasites. Eight new meroterpenoids with varying potencies were isolated from the extract, and compounds 1, 2, and 4 exhibited strong antiparasitic activity against the mentioned parasites and cytotoxic activity against HepG2 cells.

Complete Genome Sequence Analysis of Kribbella sp. CA-293567 and Identification of the Kribbellichelins A & B and Sandramycin Biosynthetic Gene Clusters

Summary: In this study, a strain of Kribbella sp. CA-293567 was found to produce sandramycin and kribbellichelins A-B, which are new secondary metabolites. The complete genome sequencing and in silico identification of biosynthetic gene clusters were performed, focusing on the pathways encoding these compounds. A comparative analysis of the biosynthetic potential of 38 publicly available genomes from Kribbella strains was also presented.

Profiles of the parenting experience in families of autistic children

Summary: Parents of autistic children often experience more stress and have worse mental and physical health compared to parents of neurotypical children. However, not all parents of autistic children are distressed. This study examined the different parenting profiles of 183 mothers and fathers of autistic children and found that parents in the adaptive profile, characterized by low stress, high competence, and positive parenting strategies, had children with fewer behavioral problems and less severe autism symptoms. The findings suggest the importance of understanding and supporting diverse parenting experiences in interventions for families of autistic children.

Summary: This study examined the relationships between child social functioning, parenting stress, and Part C Early Intervention (EI) enrollment. The results showed no direct relationship between EI enrollment and parenting stress, but a significant inverse association was found between child social functioning and parenting stress, which was moderated by EI enrollment. The findings suggest potential benefits of timely referral and access to EI for children at risk of developmental delay in historically underserved communities.

Interactions of Different Streptomyces Species and Myxococcus xanthus Affect Myxococcus Development and Induce the Production of DK-Xanthenes

Summary: The co-culturing of microorganisms is a well-known strategy to study microbial interactions in the laboratory. In this study, the effects of the interaction between nine Streptomyces species and predator bacteria Myxococcus xanthus were investigated. It was found that certain Streptomyces species induce mound formation of M. xanthus on complex media. Moreover, the study identified siderophores produced by S. griseus and molecules produced by S. peucetius that affect the behavior of M. xanthus.

Xerophytic Lichens from Gypsiferous Outcrops of Arid Areas of Andalusia as a Source of Anti-Phytopathogenic Depsides

Summary: In this study, six species of lichens from gypsum areas in southern Spain were tested for their potential as sources of new antifungal metabolites. Three lichen extracts showed antifungal activity, and ten metabolites were identified, including a new derivative. These metabolites exhibited broad-spectrum antifungal activity against various phytopathogenic fungi.

Oxepinamides L and M, two new oxepine-pyrimidinone-ketopiperazine type nonribosomal peptides from Aspergillus californicus

The structures of these compounds were characterized by spectroscopic analysis, and the absolute structure of compound 1 was determined by ECD calculation. The antibacterial and cytotoxic properties of compound 1 were evaluated.
