Summary: Through genome-wide gene x sex interaction study in a sub-Saharan African population with orofacial clefts, a novel risk locus on chromosome 8p22 with sex-specific effects was identified. The genetic variant at this locus showed different impacts on the risk of cleft lip with or without cleft palate between males and females. The locus contains genes related to craniofacial development and rare coding mutations, supporting its role in the genetic etiology of nonsyndromic orofacial clefts.

Summary: In this study, it was found that mothers exhibited higher levels of parenting stress and satisfaction compared to fathers. Predictors of higher parenting stress included being female, having two children, and experiencing work-home conflict. On the other hand, having no partner and being male were associated with lower parenting satisfaction.

Summary: Hypodontia has a significant impact on dentofacial features, including retruded lips, tooth position, and facial convexity. The severity of the impact is related to the number of missing teeth, with 5 or more congenitally missing teeth having a greater effect on facial aesthetics.

Summary: This study investigates the prevalence of medical conditions affecting patients with cleft lip and/or palate in three cleft units in the UK. The results showed that 38.9% of patients had associated medical conditions. The study highlights the impact of these medical conditions on dental care complexity for these patients.

Summary: Sleep-disordered breathing (SDB) occurs more frequently in children with craniofacial anomalies, and data on SDB prevalence in Thailand is limited. This study aimed to determine the prevalence of SDB and associated risk factors among Thai children with congenital craniofacial anomalies. The study found a higher prevalence of SDB in children with syndromic craniofacial anomalies, with risk factors including overweight, allergic rhinitis, and tonsillar hypertrophy. Understanding the prevalence and related factors can improve care for craniofacial patients.

Summary: This paper investigates the influence of commuting on time use and satisfaction within households. The findings suggest that longer commutes reduce leisure time for men, leading to decreased life satisfaction. However, longer commutes for women reduce household tasks time without impacting satisfaction with task allocation. Intra-household interactions regarding time use also have implications for domains and overall life satisfaction.

Summary: This study compared the outcome of facial symmetry in patients with craniofacial malformations who underwent simultaneous digitally planned patient-specific implant (PSI-) based orthognathic surgery and polyether ether ketone (PEEK) bone augmentation. The results showed that simultaneous PEEK bone augmentation significantly improves facial symmetry compared to conventional orthognathic surgery, even in patients with syndrome-related underdevelopment.

Summary: This study reports a severe case of oligodontia in a female patient with the EDA gene variant c.742-2A>G. The results strongly suggest that this variant is pathogenic and the oligodontia in the patient is exceptionally severe. Early identification of this variant is crucial for planning adequate treatment and follow-up.

Summary: This study investigates the role of the Gnas gene in calvaria development in mice and reveals that loss of Gnas leads to craniosynostosis and activation of Hedgehog signaling is associated with cartilage ossification. Reduction of Hedgehog signaling could be a potential intervention to reduce skull deformity in craniosynostosis.

Summary: Nonsyndromic orofacial clefts are complex congenital malformations with a multifactorial etiology. The face shape hypothesis (FSH) provides a theoretical explanation for the development of this malformation. Studies have shown changes in facial prominence shape during embryonic development in mice and altered post-natal facial shape in individuals at risk for orofacial clefting, revealing genes associated with orofacial clefting. Research on the FSH contributes to our understanding of the etiology of orofacial clefts.

Summary: This review presents the facial, oral, dental or orthodontic findings and diagnostic tools related to fetal alcohol spectrum disorder. It found that there are heterogeneous guidelines and parameters for diagnosis, and there is a need for uniform and objective diagnostic criteria for the orofacial region. A bio database with values and parameters for different ethnicities and age groups should be made available for diagnosis.

Summary: This study conducted a comprehensive chart review of VACTERL association patients from two medical centers in California. The results showed that ear anomalies, microtia, and hearing loss are part of the phenotypic diversity of VACTERL association, and diabetes and twinning may be associated with an increased risk for this phenotype.

Summary: Lymphatic anomalies encompass a range of disorders, from common localized lymphatic malformations to rare complex lymphatic anomalies. Imaging diagnosis of cystic lymphatic malformations is relatively straightforward, but diagnosing complex lymphatic anomalies can be more challenging. Radiologists play a crucial role in the diagnosis and management of complex lymphatic anomalies, and being familiar with the spectrum of entities and their typical imaging findings is important.

Summary: Repair of soft tissue defects in myelomeningoceles remains challenging. This study showed a simplified approach to repair with low reoperation rates and no cases of tethered cord or cerebrospinal fluid leak. Complications were not related to flap type, recommending the use of any tissue type for dural coverage and 2-layer primary closure of the skin.

Summary: Craniofacial anomalies, such as cleft lip and palate, are congenital disorders affecting the cranium and facial bones. The high winged Le Fort I osteotomy is a surgical option that can simultaneously correct midface deficiency and malocclusion in cleft and craniofacial patients while restoring optimal esthetic convexity. This advancement significantly improves malocclusion and aesthetic concerns by reestablishing maximal midfacial support.

Summary: The majority of craniofacial surgeons order preoperative head CTs in patients with craniosynostosis, while only a quarter order postoperative CTs mainly to evaluate surgical outcomes. Surgeons with higher operative volume are less likely to order preoperative head CTs. Consensus guidelines are needed to prevent resource waste and unnecessary radiation exposure in children.

Summary: This study investigated the association between habitual snoring, middle ear disease, and speech problems in children with cleft palate. The study found that a higher proportion of children with cleft palate had sleep problems, middle ear disease, and speech problems. Children with isolated cleft palate were more likely to have habitual snoring and early episodes of middle ear disease. The frequency of middle ear disease with effusion was higher in children with cleft lip and palate compared to those with Robin sequence. The presence of at least one episode of middle ear disease in the last year was associated with an increased frequency of habitual snoring.

Summary: The incidence of cancer worldwide is expected to exceed 22 million annually by 2030, with approximately half of these patients requiring radiation therapy. Current clinical management strategies for radiation-induced bone complications are suboptimal. Research on three therapeutics, amifostine, deferoxamine, and adipose-derived stem cells, has shown promising results in promoting healing and regeneration of irradiated bone.
