Background CD133 has been suggested to be a cancer stem cell (CSC) marker in various types of cancers. The present study assessed the relationship between CD133 expression and clinicopathological features of gastric cancer. In addition, the prognostic value of CD133 for gastric cancer was evaluated. Methods In total, 100 advanced gastric cancer patients who received curative gastrectomy and adjuvant chemotherapy were included. CD133 expression was determined by immunohistochemistry and clinicopathological results, including survival, were analyzed. Results CD133 was expressed in 23% of advanced gastric cancer patients (23/100). CD133 expression was significantly associated with serosal exposure (P?=?0.036), venous invasion (P?=?0.047), well and moderate differentiation (P?=?0.002), and intestinal-type Lauren classification (P?=?0.001). CD133-positive patients had a significantly worse 5-year disease-free (28.1% vs. 65.8%, P?=?0.002) and overall (47.5% vs. 74.0%, P?=?0.037) survival rate than those who were CD133-negative. A multivariate analysis suggested that CD133 expression significantly affected the 5-year disease-free and overall survival. Conclusions CD133 may play an important role in chemoresistance and recurrence, thus representing a promising predictive marker for the prognosis of gastric cancer. J. Surg. Recent application of artificial intelligence on histopathologic image-based prediction of gene mutation in solid cancers

Mohammad Rizwan Alam, Kyung Jin Seo, Jamshid Abdul-Ghafar, Kwangil Yim, Sung Hak Lee, Hyun-Jong Jang, Chan Kwon Jung, Yosep Chong

Summary: The status of predicting gene mutations on histologic images using artificial intelligence (AI) models was assessed through a systematic review. The study found that while the accuracy of predicting cancer driver gene mutations in specific organs was relatively high, the overall accuracy of gene mutation prediction needs improvement. Therefore, further validation with larger datasets is necessary before AI models can be used in clinical practice to predict gene mutations.

Summary: After the outbreak of the SARS-CoV-2 pandemic, a novel mRNA vaccine (BNT162b2) was developed at an unprecedented speed. Although widespread immunity has been achieved through vaccination and infections, even individuals who have recovered from SARS-CoV-2 infection are recommended to receive BNT162b2 vaccination due to its effectiveness in lowering the risk of recurrent infection. However, the BNT162b2 vaccine has been reported to increase the risk of myocarditis. This study tracked changes in chromatin dynamics in peripheral blood mononuclear cells (PBMCs) of a patient who developed myocarditis after BNT162b2 vaccination, revealing alterations in interferon signaling and RUNX2/3 activity.

Devising a deep neural network based mammography phantom image filtering algorithm using images obtained under mAs and kVp control

Summary: This study investigates the use of a deep neural network based algorithm to filter mammography phantom images that will pass or fail. Based on 543 phantom images from a mammography unit, VGG16 based phantom shape scoring models were created, and filtering algorithms were designed to filter out failed or passed phantom images. External validation using 61 phantom images from two different medical institutions showed that the scoring models achieved F1-scores of 0.69 (95% confidence interval 0.65, 0.72) for multi-class classifiers and 0.93 (95% CI 0.92, 0.95) for binary-class classifiers, with an area under the receiver operating characteristic curve of 0.97 (95% CI 0.96, 0.98). Out of the 61 phantom images, 42 (69%) were filtered by the algorithms without the need for further assessment by a human observer. This study demonstrates the potential of using deep neural network based algorithms to reduce the human workload in mammographic phantom interpretation.

AGE-RAGE axis culminates into multiple pathogenic processes: a central road to neurodegeneration

Reshmee Bhattacharya, Mohammad Rizwan Alam, Mohammad Azhar Kamal, Kyung Jin Seo, Laishram Rajendrakumar Singh

Summary: Advanced glycation end-products (AGEs) are toxic compounds that modify proteins and interact with their receptor RAGE, leading to various pathological consequences in the brain. Recent research has shown the involvement of the AGE-RAGE axis in neurodegenerative diseases, such as disruption of blood brain barrier and dysregulation of antioxidant enzymes. Specific inhibitors of TIR domains of TLR or RAGE receptors are considered to be important for the therapeutic intervention of these diseases. Clinical perspectives have also been discussed.

Summary: In this case study, a 49-year-old female patient with menorrhagia was initially suspected to have cervical squamous cell carcinoma based on magnetic resonance imaging (MRI) findings. However, histological examination confirmed the lesion to be angioleiomyoma after hysterectomy. This is the first reported case of angioleiomyoma presenting with MRI findings suggestive of uterine SCC. Recognizing the imaging mimics of angioleiomyomas can be beneficial for accurate diagnosis and appropriate treatment strategies.

Summary: This study aimed to develop a nomogram for estimating the risk of extraperigastric lymph node metastasis (LNM) in early gastric cancer (EGC). The possible risk factors for extraperigastric LNM were determined through comparing the clinicopathological characteristics of positive and negative LNM groups and conducting univariate and multivariate analyses. A risk prediction model was developed based on these factors, and it showed good agreement in internal validation.
