Background and Objectives: The molecular genetic alterations leading to gastric malignancy are largely unknown. This study aimed to unravel the genomic DNA copy number aberrations (CNAs) profile during gastric tumorigenesis. Methods: In this study, we performed genomic profiling in a set of 50 intestinal type gastric carcinomas by a PCR-based relative quantification method, multiple ligation-dependent probe amplification (MLPA) with 112 cancer-related gene loci selected throughout each human chromosome as probes of MLPA assay. Results: Numerous chromosomal DNA CNAs, including gains of 3p22, 4q25, 8q24, 11p13, and 20q13, and losses of 1p36 and 9p21, were identified by MLPA assay as recurrent DNA CNAs in gastric cancer. Moreover, we found the median numbers of gains, losses, and total CNAs were significantly higher in lymph node metastasis positive patients than in cases without metastasis. And gain of 11p13 and losses of 9p21.3, 11q13.3, 17q25.3, and 22q11.23 were associated with lymph node metastasis (P<0.05). Finally, two major groups, including G1 + 2 with a large number of CNAs and 03 + 4 with a small number of CNAs, can be successfully distinguished by hierarchical cluster analysis. Conclusions: Our results proved MLPA is a reliable and efficient method to evaluate DNA copy number changes in gastric cancers. J. Surg. Oncol. 2011;103:124-132. (C) 2010 Wiley-Liss, Inc.

Summary: YAP1 is highly upregulated in peritoneal carcinomatosis tumor cells, contributing to cancer stem cell properties and metastasis. Inhibition of YAP1 can significantly slow tumor growth and eliminate peritoneal carcinomatosis in preclinical models. Pharmacological inhibition of YAP1 can reduce cancer stem cell-like properties and suppress tumor growth, especially in combination with cytotoxic drugs in vivo models.

Increased Expression of LIPC Is Associated with Aggressive Phenotype of Borrmann Type 4 Gastric Cancer

Jin-yu Huang, Wei-lan Zhang, Ya-nan Xing, Wen-bin Hou, Song-cheng Yin, Zhen-ning Wang, Yu-en Tan, Ying-ying Xu, Zhi Zhu, Hui-mian Xu

Summary: High expression of LIPC is associated with poor clinical outcomes and plays an important role in regulating cell migration and invasion in Borrmann type 4 gastric cancer. High LIPC expression correlates with lymph node metastasis, advanced TNM stage, and poor overall survival in Borrmann type 4 gastric cancer patients. By reducing LIPC expression, cell invasion and migration are suppressed and specific gene expression is altered in Borrmann type 4 gastric cancer.

Superiority of log odds of positive lymph nodes (LODDS) for prognostic prediction after gastric cancer surgery: a multi-institutional analysis of 7620 patients in China

Pengfei Gu, Jingyu Deng, Zhe Sun, Zhenning Wang, Wei Wang, Han Liang, Huimian Xu, Zhiwei Zhou

Summary: The study compared different staging systems for predicting the prognosis of gastric cancer patients, finding that the LODDS staging system was superior to other lymph-node classifications. For predicting the prognosis of GC patients, the modified TLM staging system showed better discriminative ability and optimistic prognostic stratification compared to the 8th TNM or TRM staging systems.

Summary: This study found that higher levels of tRF-3017A were associated with significantly higher lymph node metastasis in gastric cancer patients, and tRF-3017A may play a role in promoting the migration and invasion of gastric cancer cells by silencing the tumor suppressor NELL2.

Summary: Genetic alterations in ARID1A were found in over 30% of gastric adenocarcinoma cases, and the loss of ARID1A leads to activation of the mTOR pathway and increased sensitivity to mTOR inhibitors, suggesting a new therapeutic strategy for targeting the mTOR pathway in patients with gastric adenocarcinoma with ARID1A deficiency.

Survival landscape of different tumor regression grades and pathologic complete response in rectal cancer after neoadjuvant therapy based on reconstructed individual patient data

Jia-Yi Li, Xuan-Zhang Huang, Peng Gao, Yong-Xi Song, Xiao-Wan Chen, Xing-Er Lv, Yv Fu, Qiong Xiao, Shi-Yv Ye, Zhen-Ning Wang

Summary: The study found that patients with rectal cancer achieving pathologic complete response (pCR) had significantly higher 10-year overall survival and 5-year disease-free survival compared to those in the non-pCR group. Improvement in tumor regression after neoadjuvant therapy was associated with increased overall survival and disease-free survival, and the pCR group had longer overall survival and disease-free survival than the non-pCR group. The results suggest a positive relationship between better tumor regressions and improved survival benefits, highlighting the importance of achieving pCR in the treatment of rectal cancer.
