Summary: In this study, the authors designed a hybrid material AuNPs@Fe-Zr-MOL consisting of gold nanoparticles (AuNPs) and metal-organic layers (MOLs) for the amplification of electrochemiluminescence (ECL) signal. The addition of Fe-Zr-MOL, a novel metal-organic framework with peroxidase-like activity, effectively enhanced the ECL signal of luminol. The planar structure of Fe-Zr-MOL exposed more active sites for luminol catalysis, improving its conductivity. Furthermore, the integration of AuNPs not only improved electron transport efficiency but also allowed for antibody conjugation, enabling the construction of a sandwich immuno-sensor for alpha fetoprotein (AFP) detection. The results demonstrated the effective amplification of ECL signal and immunodetection of AFP.

Summary: Label-free electrochemiluminescence (ECL) immunoassays (lf-ECLIA) have become an attractive method for biomarker detection due to their simplicity, speed, and cost-effectiveness. This study investigated the interaction between biomarkers and analytical interfaces constructed using a nanoluminophore and antibody-functionalized gold nanoparticles. The results showed that positively charged biomarkers increased the ECL intensity, while negatively charged biomarkers decreased the ECL intensity. A charge-dependent lf-ECLIA method was established based on the ECL signal changes, which exhibited different responses to positively charged and negatively charged biomarkers.

Summary: A sensitive electrochemiluminescence (ECL) immunosensor was developed using functionalized metal organic layers (MOLs) for the detection of carcinoembryonic antigen (CEA). The immunosensor utilized gold nanoparticles (AuNPs) grown on ultrathin two-dimensional MOLs, which acted as electron transfer accelerators and labels for antibody conjugation. The AuNPs@Ir-Zr-MOL nanocomposite exhibited high ECL efficiency and could detect CEA concentration in the range of 1.00 pg mL-1 to 100 ng mL-1 with a detection limit of 0.200 pg mL-1 (S/N = 3).
