Aflatoxins (AFs) B-1, B-2, G(1) and G(2) were extracted from nuts by using pressurized liquid extraction (PLE) and the PLE extracts were analyzed using HPLC with fluorescence detection using photochemical post-column derivatization without further cleanup procedures. Several extraction parameters such as temperature (25, 40, 60 and 80 degrees C), pressure (500, 1.000, 1500 and 2000 psi), solvent extraction mixture (acetone, acetonitrile, ethyl acetate and methanol), number of cycles (I and 2), use of dispersing agents and cell size (5 and 11 mL) were investigated for their effects on the extraction performance. The results showed 60 degrees C, 1500 psi, acetonitrile, one cycle and a cell size of 5 mL as most favorable PLE operating conditions. The proposed analytical method provides LODs below the maximum levels established by European Union regulations and the recoveries of the four AFs were between 77 and 93% at spiking levels of 4, 2 and 0.5 mu g/kg for AFB(1) and AFG(1) and 1, 0.5 and 0.13 mu g/kg for AFB(2) and AFG(2). Validation was carried out using certified reference materials. PLE has been applied for the first time to the analysis of AFs in nuts and offers the possibility for fast simple and accurate quantitative determination of studied mycotoxins.

Characterization of the Biological Activities of a New Polyphenol-Rich Extract from Cinnamon Bark on a Probiotic Consortium and Its Action after Enzymatic and Microbial Fermentation on Colorectal Cell Lines

Alessandra De Giani, Stefania Pagliari, Jessica Zampolli, Matilde Forcella, Paola Fusi, Ilaria Bruni, Luca Campone, Patrizia Di Gennaro

Summary: Cinnamon polyphenols have antimicrobial activity against Staphylococcus aureus and Bacillus subtilis, but this activity is lost after in vitro digestion. However, they exhibit strong prebiotic potential for probiotic strains. Additionally, the secondary metabolites produced from cinnamon extract or its digested form have protective effects against colorectal tumor cells.

Natural Deep Eutectic Solvents (NADESs) Combined with Sustainable Extraction Techniques: A Review of the Green Chemistry Approach in Food Analysis

Ciro Cannavacciuolo, Stefania Pagliari, Jessica Frigerio, Chiara Maria Giustra, Massimo Labra, Luca Campone

Summary: Usual extraction processes for analyzing foods, supplements, and nutraceutical products involve massive amounts of organic solvents causing negative impact on the environment and human health. Natural deep eutectic solvents (NADES), a new class of green solvents, are sustainable, biodegradable, non-toxic and cost-effective, making them a valid alternative to conventional solvents. This review provides an overview of NADES, including their classification, preparation methods, physicochemical properties, toxicity, and their potential application as extractants of bioactive compounds in food analysis.

Antioxidant and Anti-Inflammatory Effect of Cinnamon (Cinnamomum verum J. Presl) Bark Extract after In Vitro Digestion Simulation

Stefania Pagliari, Matilde Forcella, Elena Lonati, Grazia Sacco, Francesco Romaniello, Pierangela Rovellini, Paola Fusi, Paola Palestini, Luca Campone, Massimo Labra, Alessandra Bulbarelli, Ilaria Bruni

Summary: Cinnamon bark is widely used for its organoleptic features in food and has been found to have beneficial effects on human health. Cinnamon extracts contain antioxidant, anti-inflammatory, and anticancer biomolecules. This study evaluated the antioxidant and anti-inflammatory bioactivity of digested cinnamon extract and found that the digestive process reduces the polyphenol content while maintaining other bioactive compounds and functional effects at the cellular level.

An analytical platform for the screening and identification of pyrrolizidine alkaloids in food matrices with high risk of contamination

Serena Rizzo, Rita Celano, Anna Lisa Piccinelli, Simona Serio, Mariateresa Russo, Luca Rastrelli

Summary: An analytical platform for the detection of pyrrolizidine alkaloids (PAs) in various products was proposed. The platform employed a wide-scope suspect screening method for characterization of PAs and a target screening method for high-throughput detection of 118 PAs. The platform showed high effectiveness in detecting and identifying both target and untarget PAs, as demonstrated by the screening of commercial samples.

LC-MS and GC-MS Data Fusion Metabolomics Profiling Coupled with Multivariate Analysis for the Discrimination of Different Parts of Faustrime Fruit and Evaluation of Their Antioxidant Activity

Ciro Cannavacciuolo, Stefania Pagliari, Chiara Maria Giustra, Sonia Carabetta, Werther Guidi Nissim, Mariateresa Russo, Paola Branduardi, Massimo Labra, Luca Campone

Summary: The comparative chemical composition of different parts of Faustrime fruits was analyzed using GC-MS and UHPLC-HRMS QTof. The data obtained were combined to define the fruit's complex antioxidant activity. The principal component analysis showed the occurrence of specific compounds in different parts of the fruit, such as alpha-bisabolol and alpha-trans-bergamotol in the pulp and albedo, hexanoic acid in the seeds, and various coumarins and phenolics in the peels. The higher radical scavenging activity of the pulp was attributed to the presence of citric acid.

Irena Mincheva, Zheko Naychov, Cristo Radev, Ina Aneva, Luca Rastrelli, Maria Kamusheva, Nikolay Nikolov, Ekaterina Kozuharova

Summary: Rhodopes Mountain is poorly studied in terms of ethnobotany. Our preliminary observation suggests that the local population still relies on previously collected medicinal plants. In this study, we aimed to evaluate the contemporary usage of medicinal plants and the associated health conditions in the Central and East Rhodopes. Through in-depth field research and interviews, we found that 92 informants mentioned the use of 114 plant species. The study shows that medicinal plants are most frequently mentioned in relation to diseases affecting the nervous, respiratory, digestive, and cardiovascular systems.

Target screening method for the quantitative determination of 118 pyrrolizidine alkaloids in food supplements, herbal infusions, honey and teas by liquid chromatography coupled to quadrupole orbitrap mass spectrometry

Serena Rizzo, Rita Celano, Anna Lisa Piccinelli, Mariateresa Russo, Luca Rastrelli

Summary: An analytical procedure for the screening and determination of pyrrolizidine alkaloids (PAs) in food supplements, herbal infusions, honey, and teas was successfully validated. The study found that 10% of the analyzed samples were contaminated above the maximum levels (MLs) set by European legislation. Contamination of herbal infusions of mixed plants raised potential health concerns, with 23% showing PA levels above ML. Although a high number of PAs not included in the regulation were detected in honey and herbal food supplements, their contribution to overall levels in honey was not significant.
