The thalamus plays pivotal roles in the central nervous system as relay center for organizing information, such as auditory and visual senses front diverse brain regions and their re-distribution to the cerebral cortex. Brain diseases including schizophrenia, Parkinson's disease, epilepsy, and bipolar disorder have been associated with the thalamus, We performed a shotgun proteome analysis of iTRAQ-labeled tryptic peptides of human mediodorsal thalamus protein extracts coming from two healthy male and two healthy female subjects. The shotgun workflow consisted of IEF fractionation, RP LC and MALDI-TOF/TOF mass spectrometric analysis. We were able to identify 542 proteins that are involved in different biological processes and from diverse cellular localizations. A considerable fraction of these proteins had not been identified by traditional proteomics methods such as 2-DE. The thalamus proteome contributes to the knowledge of the human brain proteome and future applications in basic and clinical research.

Lower Emotion Regulation Competencies Mediate the Association between Impulsivity and Craving during Alcohol Withdrawal Treatment

Daniela Reichl, Niklas Enewoldsen, Kiona K. Weisel, Sebastian Saur, Lukas Fuhrmann, Catharina Lang, Matthias Berking, Mathias Zink, Andreas Ahnert, Peter Falkai, Thomas Kraus, Thomas Hillemacher, Felix-N Muller, Stephan Lins, Dominikus Bonsch, Linda Kerkemeyer, Sabine Steins-Loeber

Summary: There is evidence that craving mediates the relationship between Impulsive Personality Traits (IPTs) and relapse during the treatment of an Alcohol Use Disorder (AUD). Lower emotion regulation competencies were found to mediate the relationship between attentional as well as non-planning IPTs and craving. These findings contribute to a deeper understanding of the underlying processes and may inform tailored interventions for AUD treatment.

Summary: Increased activity of DYRK1A may play a role in the abnormal metabolism of APP, and it regulates the intracellular distribution of APP in neurons by affecting axonal transport. These findings provide insights into the potential therapeutic intervention of using DYRK1A inhibitors to restore APP axonal transport in Down's syndrome and Alzheimer's disease.

Induced pluripotent stem cell-derived astrocytes from patients with schizophrenia exhibit an inflammatory phenotype that affects vascularization

Pablo Trindade, Juliana Minardi Nascimento, Barbara S. Casas, Tomas Monteverde, Juciano Gasparotto, Camila Tiefensee Ribeiro, Sylvie Devalle, Daniela Sauma, Jose Claudio Fonseca Moreira, Daniel Pens Gelain, Lisiane O. Porciuncula, Veronica Palma, Daniel Martins-de-Souza, Stevens K. Rehen

Summary: This study examined the proteome, inflammatory responses, and secretome effects of astrocytes derived from schizophrenia patients. The findings suggest that these astrocytes exhibit molecular and functional abnormalities, affecting immune function and vascularization. Reduced expression of NF-kappa B p65 subunit was observed, and the secretion of interleukin 8 (IL-8) was particularly elevated. The conditioned medium from these astrocytes could reduce the diameter of newly grown vessels.

Neurodevelopmental disturbances in schizophrenia: evidence from genetic and environmental factors

Andrea Schmitt, Peter Falkai, Sergi Papiol

Summary: Schizophrenia is a neurodevelopmental disorder influenced by genetic and environmental risk factors. The interaction between these factors during prenatal, perinatal, and adolescence periods contribute to the onset of symptoms in early adulthood. These factors lead to disturbances in brain connectivity and molecular/cellular abnormalities.

Mitochondrial, cell cycle control and neuritogenesis alterations in an iPSC-based neurodevelopmental model for schizophrenia

Giuliana S. S. Zuccoli, Juliana M. M. Nascimento, Pedro M. M. Moraes-Vieira, Stevens K. K. Rehen, Daniel Martins-de-Souza

Summary: Schizophrenia is a severe neurodevelopmental psychiatric disorder affecting around 1% of the global population. Proteomic studies on neural stem cells and neurons derived from induced pluripotent stem cells from schizophrenia patients have revealed differentially expressed proteins related to mitochondrial function, cell cycle control, DNA repair, and neuritogenesis. Functional analysis of neural stem cells also showed alterations in mitochondrial oxygen consumption and higher levels of intracellular reactive oxygen species in schizophrenia-derived cells.
