Summary: Glucocorticoids (GCs) are commonly used in autoimmune diseases, but long-term or high-dose usage can lead to severe side effects, such as steroid myopathy and osteonecrosis. This case report describes a pemphigus vulgaris patient who developed muscle weakness and osteonecrosis after a cumulative dose of methylprednisolone. Genotyping showed the patient was sensitive to GCs. Tapering the GC dosage improved the patient's condition.

Summary: The COVID-19 pandemic has led to the emergence of new variants of SARS-CoV-2 and the development of several post-COVID conditions. This report describes a new post-COVID condition observed in patients who have recently recovered from COVID-19 and have received high dose steroid treatment - spontaneous de novo development of steroid induced avascular necrosis in the jaws. This condition is distinct from previously reported medication-related osteonecrosis of the jaws and further research is needed to understand its features.

Specifically, the rs10965059 polymorphism is correlated with a reduced risk of SONFH in individuals over 40 years old, male populations, and those with bilateral SONFH.

Summary: The study found that miR-224-5p was upregulated in GC-treated BMSCs, regulating the adipogenic and osteogenic differentiation of BMSCs by inhibiting Smad4 and activating the Smad4-Taz axis. The miR-224-5p antagomir could alleviate the inhibitory effects of GCs and promote bone formation in steroid-induced ONFH models, suggesting it as a potential novel therapeutic target for this condition.

Summary: This study aimed to establish the importance of risk factors in the development of MRONJ in cancer patients receiving zoledronic acid therapy. The analysis of 174 cancer patients revealed that treatment duration, chemotherapy, and hypertension were risk factors, while endocrine therapy and obesity were protective factors for MRONJ.

This pharmacovigilance study aimed to assess the risk of reporting osteonecrosis with TT compared to other medications. The results showed that TT was associated with a greater risk of reporting osteonecrosis compared to all other drugs and drugs for BPH.
