Summary: In this study, six PRL-R variants associated with prolactinomas were identified, with two variants showing higher frequency in patients compared to the genomic database. These variants were associated with male sex, higher serum PRL levels, larger tumors, and cabergoline resistance. These findings suggest the involvement of the prolactin/prolactin receptor system in prolactinoma tumorigenesis and resistance to cabergoline.

Various therapies, both pharmacological and nonpharmacological, have been suggested to improve sexual dysfunction, with evidence-based treatment available for erectile dysfunction (ED).

Summary: Prolactin plays an important role not only in reproductive physiology, but also in neurohormonal appetite regulation and metabolism. Elevated prolactin levels are associated with worsening metabolic disease, but low prolactin levels could also be metabolically unfavorable. This review discusses the pathophysiology of prolactin-related metabolic changes, as well as the less recognized effects of prolactin on adipose tissue, pancreas, liver, and small bowel. The effects of dopamine agonists on the metabolic profiles of patients with hyperprolactinemia are also discussed.

Summary: Sexual dysfunction (SD) is a common non-motor symptom of Parkinson's disease (PD) that is often overlooked. The mechanism of SD in PD is not well understood, but studies have shown that dopamine (DA) in the hypothalamus plays a role in regulating sexual behavior. In this study, a Drosophila melanogaster model of sporadic SD in PD was developed and observed based on abnormalities in courtship behavior markers. The model exhibited SD without mobility defects, and there was no observable degeneration of dopaminergic neurons in the brain. This study sheds light on the molecular basis of SD in PD and provides an opportunity for screening potential therapeutic molecules.

Transsphenoidal surgery (TSS) can achieve early remission in half of DA-resistant patients, especially in microprolactinoma cases. Tumor invasiveness and preoperative PRL levels are significant predictors of hyperprolactinemia recurrence after TSS, with early postoperative remission being an independent predictor of tumor recurrence.

Sequence2Script is a web-based tool designed to assist users in efficiently translating PGx testing results into evidence-based prescribing recommendations. This tool supports 97 gene-drug pairs and allows users to adjust recommendations, generating a clinical report summarizing patient genotype and prescribing recommendations. The hope is that Sequence2Script will help standardize the use of PGx testing results and reduce barriers to implementation in practice.
