ITIH3 polymorphism may confer susceptibility to psychiatric disorders by altering the expression levels of GLT8D1

A recent genome-wide analysis indicated that a polymorphism (rs2535629) of ITIH3 showed the strongest association signal with susceptibility to psychiatric disorders in Caucasian populations. The aim of the study was to replicate the association of rs2535629 with schizophrenia and major depressive disorder (MDD) in Japanese subjects. A total of 611 patients with schizophrenia, 868 with MOD, and 1193 healthy controls were successfully genotyped for rs2535629. A significant difference in allele distribution was found between patients with schizophrenia and controls (odds ratio [OR] = 1.21, 95% confidence interval [Cl]: 1.05-1.39, P = 0.0077). A similar trend was found for patients with MDD (OR = 1.11,95% CI: 0.98-1.26, P = 0.092). The allele distribution in the combined patient group (schizophrenia and MDD) was significantly different from that of the control group (OR = 1.15, 95% CI: 1.03-1.28, P = 0.011). Gene expression microarray analysis of whole blood samples in 39 MDD patients and 40 healthy controls showed that rs2535629 has a strong influence on the expression levels of ITIH4 and GLT8D1. The expression levels of GLT8D1 were significantly higher in patients with MDD than in controls (P = 0.021). To our knowledge, the present study showed for the first time the association of rs2535629 with psychiatric disorders in an Asian population. Our findings suggest that rs2535629 influences the susceptibility to psychiatric disorders by affecting the expression level of GLT8D1. (C) 2013 Elsevier Ltd. All rights reserved.

Neurobiology of early life adversity: A systematic review of meta-analyses towards an integrative account of its neurobiological trajectories to mental disorders

Summary: Adverse childhood experiences (ACEs) can have long-lasting neurobiological effects, increasing the risk of developing mental disorders later in life. This systematic review integrates existing evidence from multiple fields and shows that individuals with ACEs exhibit blunted cortisol response to stress, low-grade inflammation indicated by increased C-reactive protein levels, exaggerated amygdalar response to negative emotions, and reduced hippocampal gray matter volume. These alterations are observed consistently in individuals with and without psychiatric diagnoses.

Discrepancy between Clinician-rated and Self-reported Depression Severity is Associated with Adverse Childhood Experience, Autistic-like Traits, and Coping Styles in Mood Disorders

The findings suggest that patients with adverse emotional experiences, autistic-like traits, and self-blame coping styles perceive greater distress than that evaluated objectively by clinicians. The results indicate the need for inclusion of subjective assessments to effectively evaluate depressive symptoms in patients deemed to have these psycho-behavioral concerns.

Suicidality in civilian women with PTSD: Possible link to childhood maltreatment, proinflammatory molecules, and their genetic variations

Summary: This study found that suicidal ideation and risk in PTSD patients were positively correlated with high-sensitivity C-reactive protein (hsCRP) and interleukin-6 (IL-6) levels. The risk alleles of CRP rs2794520 and IL6 rs1800796 were also associated with a higher risk of suicide. These findings suggest that genetic variations within inflammatory genes might be valuable in identifying PTSD patients at high risk of suicide.

Psychometric properties of the Distress Questionnaire-5 (DQ5) for measuring psychological distress in adolescents

Discovery and validation of protein biomarkers for monitoring the effectiveness of drug treatment for major depressive disorder

Summary: Major depressive disorder is a prevalent condition worldwide, but the proportion of patients receiving treatment has not increased. Biomarkers related to drug-treatment responses can be used to monitor the effectiveness of medication. Serum protein levels were compared among patients with depression who received medication, those who did not, and a control group. Eight biomarkers were identified, which can be used to monitor the effectiveness of drug treatment.

Functional and structural connectivity correlates of semantic verbal fluency deficits in first-episode psychosis
