Proteomic analysis of endothelial cell secretome: A means of studying the pleiotropic effects of Hmg-CoA reductase inhibitors

The clinical benefits of 3-hydroxy-3-methyl-3-glutaryl coenzyme A (HMG-CoA) reductase inhibitors (statins) are closely related to their cholesterol-lowering properties. However, the inhibition of HMG-CoA reductase may also lead to pleiotropic effects due to the ability to inhibit the synthesis of non-steroidal isoprenoid compounds, thus exerting extra-beneficial effect in preventing atherosclerosis beyond their effect on the lipid profile. To identify new drug targets by means of which statins can promote some of their beneficial effects we used a global proteomic approach to analyse the secretome of endothelial cells, a major class of proteins that control a multitude of biological and physiological processes. Differentially expressed proteins were identified using a data-independent, label-free, mass spectrometry-based method. A total of 273 proteins were identified, including 112 that were differentially expressed: 29 uniquely expressed in control cells, 14 uniquely expressed in statin-treated cells, 51 down-regulated by statin, and 18 up-regulated by statin. Gene ontology analysis revealed modulated biological processes related to proteolysis, cellular component organisation and biogenesis, and response to stress. The findings were validated by biochemical assays, thus confirming the effectiveness of our proteomic approach. In conclusion, this study underlines the role of proteomics for the discovery of novel and unpredictable targets of statins. (C) 2012 Elsevier B.V. All rights reserved.

Summary: Macrophages are versatile cells that can adapt their characteristics and functions to different environments. They play important roles in maintaining homeostasis and are also involved in various human diseases, including atherosclerosis. Proteomics has been widely used to study macrophages, and analyzing the proteome of different macrophage phenotypes can provide insights into their roles in atherosclerosis and identify potential therapeutic targets. In this review, we discuss different proteomic techniques used to profile macrophage phenotypes and summarize findings that contribute to understanding macrophage functions and developing strategies to combat atherosclerosis.

Infrared Macrothermoscopy Patterns-A New Category of Dermoscopy

Summary: The authors developed a new non-invasive dermatological infrared macroimaging analysis technique (MacroIR) that evaluates microvascular, inflammatory, and metabolic changes in skin and mucosal lesions. The technique combines naked eye observation, polarized light dermatoscopy (PLD), and MacroIR to compare results. Ten cases were evaluated using a smartphone coupled with a dermatoscope and a macro lens integrated far-infrared transducer into specific software. MacroIR imaging showed promise in improving dermatological diseases diagnosis.

Pharmacometabolomics for the Study of Lipid-Lowering Therapies: Opportunities and Challenges

Summary: Lipid-lowering therapies are widely used to prevent ASCVD and related mortality. Omics technologies have been employed to study the mechanisms and effects of these drugs, aiming to improve treatment efficacy and safety. Pharmacometabolomics focuses on studying the effects of drugs on metabolic pathways and the variation of treatment response. This review summarizes significant metabolomic studies on lipid-lowering therapies, including statins, fibrates, and novel drugs and nutraceutical approaches. Integrating pharmacometabolomics data with other omics approaches can help understand the biological mechanisms of lipid-lowering drugs and improve treatment efficacy while reducing side effects.

Exploring Facial Thermography Patterns in Women with Chronic Migraine

Development of lymph node metastasis-related prognostic markers in breast cancer

Summary: A prognostic model of BC based on LNM-related genes was established in this study, providing guidance for prognosis evaluation and precise treatment of BC. The genes related to lymph node metastasis in BC are largely unknown and need further exploration. Searching for potential lymph node metastasis-related genes of BC will provide meaningful biomarkers for BC treatment. Our model, based on TCGA-BRCA data, effectively predicted patient outcomes and classified BC patients. The identified feature genes might have a predictive function in immunotherapy. The results of this study provide a new reference for the prognosis and treatment of BC patients with lymph node metastasis.
