Consumer complaints against the blandness of modern lean meat and the frequent reference to the more strongly flavored meat that was available years ago have prompted reconsideration of high fat-depositing typical pig breeds Casertana and Large White pig breeds are characterized by a different tendency toward fat accumulation as they exhibit opposite genetic and physiological traits with respect to the energy metabolism These physiological differences were investigated in longissimus lumborum muscles through proteomics (2-DE MS/MS) and microarray approaches Data were analyzed for pathway and network analyses as well as GO term enrichment of biological functions As a result, Casertana showed a greater amount of proteins involved in glycolitic metabolism and mainly rely on fast-mobilizable energy sources Large White overexpressed cell cycle and skeletal muscle growth related genes Metabolic behavior and other implications are discussed

Plasma levels of carboxylic acids are markers of early kidney dysfunction in young people with type 1 diabetes

Timothy Vigers, Carissa Vinovskis, Lu-Ping Li, Pottumarthi Prasad, Hiddo Heerspink, Angelo D'Alessandro, Julie A. Reisz, Federica Piani, David Z. Cherney, Daniel H. van Raalte, Kristen J. Nadeau, Meda E. Pavkov, Robert G. Nelson, Laura Pyle, Petter Bjornstad

Summary: This study compared plasma metabolites between youth with and without type 1 diabetes mellitus (T1DM) and found lower levels of metabolites associated with amino acid oxidation and the tricarboxylic acid cycle in individuals with T1DM. These metabolites were related to kidney function and may serve as potential biomarkers for improving kidney bioenergetics.

Sphingosine 1-phosphate has a negative effect on RBC storage quality

Ariel Hay, Travis Nemkov, Fabia Gamboni, Monika Dzieciatkowska, Alicia Key, Matthew Galbraith, Kyle Bartsch, Kaiqi Sun, Yang Xia, Mars Stone, Michael P. Busch, Philip J. Norris, James C. Zimring, Angelo D'Alessandro

Summary: Blood storage leads to the decrease of ATP and DPG in red blood cells, which play crucial roles in erythrocyte physiology, oxygen kinetics, and posttransfusion survival. Supplementing S1P to stored RBC units can improve energy metabolism and posttransfusion recovery.

Bernard, John G. Gribben, Kevin Rouault-Pierre

Summary: Patients with myelodysplastic syndrome and ring sideroblasts (MDS-RS) suffer from symptomatic anemia due to ineffective erythropoiesis caused by SF3B1 mutations. Mis-splicing of COASY induced by these mutations affects heme biosynthesis and erythropoiesis. Supplementation with COASY substrate may serve as a potential treatment for anemia in MDS-RS patients.

Metabolic Signatures of Performance in Elite World Tour Professional Male Cyclists

Travis Nemkov, Francesca Cendali, Davide Stefanoni, Janel L. Martinez, Kirk C. Hansen, Inigo San-Millan, Angelo D'Alessandro

Summary: This study aimed to explore the metabolic characteristics of elite athletes before and after exercise in a World Tour cycling race through metabolomics analysis. The results showed significant differences in blood metabolites between aerobic and anaerobic exercise. Additionally, similar metabolic features were observed during climbing and sprinting stages in the World Tour race, and an increase in fatty acid oxidation capacity was correlated with competitive performance.

Histone deacetylase inhibitors synergize with sildenafil to suppress purine metabolism and proliferation in pulmonary hypertension

Hui Zhang, Angelo D'Alessandro, Min Li, Julie A. Reisz, Suzette Riddle, Akshay Muralidhar, Todd Bull, Lan Zhao, Evgenia Gerasimovskaya, Kurt R. Stenmark

Summary: This study investigates the effects of sildenafil on fibroblasts derived from patients with pulmonary hypertension (PH). Sildenafil showed specific effects on purine metabolites, especially adenosine, adenine, and xanthine. However, its impact on cell stress markers was limited. Combination treatment of sildenafil with HDAC inhibitors showed promising inhibitory effects on proliferation and metabolic reprogramming in PH-fibroblasts.

A Multi-Omic Mosaic Model of Acetaminophen Induced Alanine Aminotransferase Elevation

Andrew A. Monte, Alexis Vest, Julie Haines, Danielle Berninzoni, Claire Hart, Layne Dylla, Angelo D'Alessandro, Kennon J. Heard, Cheyret Wood, Jack Pattee

Summary: This study used metabolomic and genomic data to predict liver injury and regeneration in patients taking therapeutic doses of acetaminophen. Several metabolites were found to be under genetic control and predictive of ALT elevation due to acetaminophen. These findings confirm the importance of mitochondrial energy production and the urea cycle in acetaminophen-induced liver injury.
