Summary: Cranial radiation therapy is effective for childhood brain cancers, but it can lead to cognitive impairment. This study found that radiation arrested the cell cycle and impacted differentiation of neural stem cells (NSCs). Proteomic analysis revealed that radiation suppressed protein transport and neuron projection development. Additionally, oxidative modifications were observed in proteins of differentiating NSCs. Some of the identified proteins were associated with neurodegenerative diseases. These findings have implications for understanding neurodegeneration and developing preventive strategies.

Summary: The study found that microvascular endothelial cells play a crucial role in promoting neural stem cell clustering and migration, by affecting the expression of N-cadherin to promote cluster formation. This mechanism may enhance the therapeutic response of neural stem cells to sites of injury.

Summary: This paper introduces a model called MultiVelo, which extends the RNA velocity framework using differential equations to incorporate epigenomic data. The model improves the accuracy of cell fate prediction by estimating the temporal relationship between chromatin accessibility and gene expression. The study reveals different classes of genes and cell states, and identifies time lags between transcription factor expression and binding site accessibility, as well as disease-associated SNP accessibility and linked gene expression.

Summary: Tcf4 regulates neuronal positioning, with its deficiency leading to clustering of neurons. By controlling the expression of cell adhesion molecules, Tcf4 plays a role in neuronal positioning. Mutations in TCF4 result in disrupted neuronal functions, and are associated with schizophrenia and Pitt-Hopkins syndrome.

Summary: In tissue repair, cell behaviors can be modulated by sensing and responding to the topographical and biochemical cues provided by the surrounding microenvironment. Herein, we designed and fabricated a class of uniaxially aligned nanofiber yarns welded with electrosprayed microparticles to guide the directional growth of axons and the migration of neural stem cells. The migration of neural stem cells was significantly accelerated by the synthetic effects provided by the physical and biological signals.

Summary: Stem cells and their derivatives hold promise for tissue replacement therapy, but their heterogeneous characteristics pose challenges. Targeting specific cell surface markers with antibodies has proven efficient in isolating cell populations, with proteomics playing a key role in identifying these markers. Single cell proteomics and surface markers associated with progenitor stages show potential for use in regenerative medicine.

Summary: Spinal cord injury affects millions of people worldwide, and there is currently no cure. Utilizing endogenous neural stem cells (NSCs) to replace lost cells and promote structural repair is a promising approach. However, there are still many unanswered questions regarding the heterogeneity of NSCs, their interaction with the environment, and factors that enhance their response.

Summary: In this study, a composite material based on poly(L-lactide-co-glycolide) (PLGA) enriched with sol-gel bioactive glasses (SBG) was investigated. The results showed that the CM from the A1/PLGA composite significantly increased the migration rate of human bone marrow stromal cells (hBMSCs) and upregulated the expression of important biological markers.

Summary: Research has identified the long noncoding RNA Pnky as a key regulator of neural stem cell (NSC) migration in mice. Pnky interacts with splicing factors and mRNA export adaptors to modulate the splicing and export of target mRNAs, subsequently influencing NSC migration. This study provides a theoretical basis for understanding the regulation of NSC migration.
