Summary: This study synthesized a library of IgG N-glycans covering all possible biantennary and bisected structures by reprogramming biosynthetic assembly lines and deciphered their unique binding specificities with different proteins using glycan microarray technology. This synthetic IgG N-glycan library can be used as standards for N-glycan structure identification in complex biological samples and the microarray data enrich N-glycan glycomics for biomedical applications.

Summary: The highly branched tetradecasaccharide repeating unit and shorter sequences of GLSWA-1 from Ganoderma lucidum, which possess immune-enhancing activities, were prepared using a one-pot glycan assembly strategy that features orthogonal one-pot glycosylation, one-pot assembly of oligosaccharides, and modular and convergent [4+5+5] assembly of the highly branched tetradecasaccharide.

Summary: This review provides an update on the application of matrix-assisted laser desorption/ionization (MALDI) mass spectrometry to the analysis of carbohydrates and glycoconjugates. It covers the literature up to 2020 and includes methods appropriate to MALDI analysis. The review is divided into three sections, focusing on general aspects, applications to various structural types, and other areas where MALDI is extensively used. The reported work demonstrates the increasing use of new techniques and the significant impact of MALDI imaging.

Summary: Parasitic nematodes cause chronic disablement in humans worldwide. Current diagnostics for surveillance of filarial diseases have limitations, and improved assays are needed. We studied the glycan antigens of the parasite Brugia malayi and found that they triggered IgG responses. Our findings suggest potential for developing disease-specific diagnostics for filarial disease control.

Summary: The binding of glycans to glycans plays a significant role in xenogeneic self-recognition and adhesion in sponges, cell compaction in mouse embryos, and in vitro tumor cell adhesion in mammals. This intermolecular recognition process is considered a new paradigm in cellular interactions. Glycan structures in sponges have unique species-specific sequences, and their binding mechanism relies on specific interactions of small oligosaccharide motifs.

Summary: This paper presents a novel approach to synthesize 5-hydroxymethylfurfural (HMF) using metal-exchanged Keggin heteropolyacid salts as catalysts, resulting in improved yield and reduced environmental impact. The most efficient catalyst, Cu3/2PW12O40, showed high Lewis acidic strength allowing for coordination with water molecules and generation of H3O+ ions in the reaction medium.

Summary: The purpose of this study was to investigate the effects of different extraction methods on the yield, chemical structures and antioxidant activity of Dictyo-phora indusiata polysaccharides. The results showed that ultrasonic-microwave-assisted extraction (UMAE) had the greatest damage to the cell wall of the polysaccharides and the best comprehensive antioxidant capacity. Different extraction methods had no significant effect on the types of glycosidic bonds and sugar rings, but there were differences in absolute molecular weight and molecular conformation. UMAE had the highest polysaccharides yield, indicating its potential for modification and application in the functional food industry.

Summary: This study evaluated the combined effect of microwave-assisted extraction and stem additions on the chemical composition of Bonarda wines. The results showed that stem additions increased tannin content, while microwave-assisted extraction improved phenolic extraction and polymeric pigments formation, enhancing wine color. In addition, the combined strategies increased polysaccharides extraction and modified the volatile profile of wines.

Summary: A sample preparation method based on microwave assisted free radical degradation (MFRD) was proposed to establish structural-fingerprinting of polysaccharides for quality assessment. The MFRD condition was optimized and the structures of the degradation products were analyzed. A low-polymerization compositional fingerprinting was successfully constructed using high-performance anion-exchange chromatography. Compared to conventional methods, MFRD significantly improves sample preparations and is applicable to various polysaccharide samples.
