Summary: This study described a Catharanthus roseus mutant that accumulates high levels of monoterpenoid indole alkaloids (MIAs). The mutant exhibited stunted growth and deficiency in chlorophyll biosynthesis, along with lesion formation. The lesions were enriched in MIAs and resulted from upregulation of genes involved in the MIA pathway. Furthermore, the accumulation of akuammicine, a minor constituent of the mutant, was found to be inducible by jasmonate and may have a role in plant defense.

The results showed that salicylic acid treatment increased the expression of genes in the Terpenoid Indole Alkaloids pathway, leading to enhanced synthesis of alkaloids. The abundance of transcription factor families also responded to salicylic acid stimulation.

Summary: This study aimed to improve the production of secondary metabolites in Catharanthus roseus and investigate the effects of PEG-induced drought stress on physiological and biochemical characteristics. The results showed that increasing PEG concentration led to higher levels of malondialdehyde and proline in the Alba cultivar compared to the Rosea cultivar. The Alba cultivar also exhibited higher activity of antioxidant enzymes. The relative expression of vinblastine biosynthesis genes was significantly increased in both cultivars, especially in the medium with 5% PEG. Similarly, the relative expression of CrMPK3 and catalase genes increased significantly in all PEG-containing media. These findings suggest that PEG can induce the accumulation of vinblastine through a signaling pathway involving MAPK and antioxidant enzymes.

OsRR6 expression is regulated by exogenous cytokinin and various abiotic stress treatments, affecting root growth, anthocyanin accumulation and cytokinin sensitivity in plants. OsRR6(OX) plants showed upregulated defense response and anthocyanin metabolism genes, indicating a positive role of OsRR6 in imparting abiotic stress tolerance.

Summary: The rewiring of lipid metabolism in cancer cells is associated with their increased aggressiveness, making the targeting of lipid metabolism an appealing therapeutic strategy. Lipin-1, a central regulator of lipid homeostasis, has been highlighted for its potential role in cancer progression, both in cancer cells and the tumor microenvironment. Inhibiting Lipin-1 may serve as an adjuvant therapy to enhance the effects of anti-cancer treatments.

Summary: The aim of this study was to identify the active principle from the leaves of Catharanthus roseus against three mosquito species. The chloroform extract showed the highest larvicidal activity, and a triterpenoid called ursolic acid was isolated as the active constituent. Derivatives of ursolic acid were prepared and tested for their larvicidal activity, with the acetyl derivative showing the highest activity against all three mosquito species. This is the first report of ursolic acid from C. roseus with larvicidal activity.
