The Use of Sonophoresis in the Administration of Drugs Throughout the Skin

Transdermal drug delivery offers an attractive alternative to the conventional drug delivery methods of oral administration and injection. However, the stratum corneum acts as a barrier that limits the penetration of substances through the skin. Application of ultrasound to the skin increases its permeability (sonophoresis) and enables the delivery of various substances into and through the skin. Ultrasound has been used extensively for medical diagnostics and to a certain extent in medical therapy (physiotherapy, ultrasonic surgery, hyperthermia). Nevertheless, it has only recently become popular as a technique to enhance drug release from drug delivery systems. A number of studies suggest the use of ultrasound as an external mean of delivering drugs at increased rates and at desired times. This review presents the main findings in the field of sonophoresis, namely transdermal drug delivery and transdermal monitoring. Particular attention is paid to proposed enhancement mechanisms and trends in the field of topical and transdermal delivery.

Summary: This study aimed to develop and characterize biodegradable polymeric microneedle arrays loaded with losartan potassium. Lecithin/gelatin nanoparticles were prepared and optimized, and a 3D printed mold was used to create the microneedle arrays. The optimized nanoparticle formulation and microneedle arrays showed potential for delivering losartan through the skin, providing an innovative pharmaceutical form for blood pressure control.

Through three stages of experimentation, the emulgel formulation and properties were determined. The formulated emulgel demonstrated appropriate technical characteristics for the treatment of mycetoma.

Transdermal Delivery Systems for Biomolecules

Summary: Transdermal drug delivery systems have become increasingly important since the late 1970s when approved by the FDA. They are used as an alternative resource for administering potent drugs in treatments like cancer, diabetes, vaccinations, hormonal disorders, and contraception. The latest technologies in transdermal biomolecule transporters include electrical methods, chemical penetration enhancers, and nanocarriers, allowing for efficient storage, transport, and release of proteins and peptides for better treatment outcomes in various diseases.

The study shows that the next generation nanoparticles have an enhanced anti-aging effect and successfully encapsulate various active ingredients for skin rejuvenation. These nanoparticles improve solubility, bioavailability, stability, permeability, and effectiveness, providing controlled drug release with minimized side effects.

Summary: The use of nanoparticles as drug delivery systems has become increasingly important in recent decades. Oral administration, despite its disadvantages, is still the most widely used route for therapeutic treatments and controlled-release systems based on nanoparticles synthesized from biodegradable natural polymers have been found to enhance oral delivery. Chitosan, with its various properties and roles in the pharmaceutical and health fields, is capable of encapsulating and transporting drugs within the body, improving drug efficacy, and forming nanoparticles through multiple mechanisms.

Ceftriaxone-Loaded Polymeric Microneedles, Dressings, and Microfibers for Wound Treatment

Summary: This study aimed to develop polymeric dressings, microfibers, and microneedles loaded with ceftriaxone to treat diabetic wounds and promote faster healing. The formulations were optimized and subjected to physicochemical tests. The results showed that the developed dressings, microfibers, and microneedles exhibited favorable bioadhesion, tear strength, erythema, hydration, pH, and drug release properties. In vitro studies demonstrated promising flux, permeation coefficient, and time lag values in wounded skin, and in vivo tests in diabetic rats showed healing time below 14 days. These formulations have great potential for addressing the challenges of chronic wounds, such as diabetic foot, and improving clinical outcomes.

Laponite Composites: In Situ Films Forming as a Possible Healing Agent

Summary: This study evaluates the performance of Laponite in lecithin/gelatin composites and maltodextrin/sodium ascorbate mixtures. Nanoparticles were prepared using the gelatin desolvation method and turned into films via the solvent-casting method. The optimal composites were developed with 8.8 mg of Laponite, reducing particle size and avoiding agglomeration. Drug release in LGL MAS was fitted to first-order and Korsmeyer-Peppas models.
