This general functional genomics approach holds promise for future research in identifying more transport proteins of scientific, clinical, and commercial interest.

Summary: In this study, the farnesyl diphosphate biosynthetic pathway and a-bisabolene synthase were compartmentalized in the yeast Yarrowia lipolytica peroxisome, and a systems metabolic engineering approach was employed to enhance the production of a-bisabolene. Through dynamic regulation and optimization, the engineered strain achieved the highest reported production of a-bisabolene from an engineered microbe. This work lays the foundation for peroxisome engineering of Y. lipolytica for sustainable a-bisabolene production.

Summary: This study found that plant pathogens can develop multidrug resistance (MDR) through metabolomic and efflux activities. The genes encoding ABC and MFS transporters were identified to be associated with MDR in Rhizoctonia solani, and their expression levels were higher in MDR mutant compared to the wild-type strain. Two of these transporters were found to be related to MDR and their roles were confirmed by detecting their protein products and high accumulation of an antifungal compound. These findings contribute to our understanding of fungicide resistance and the development of MDR.

Summary: A strategy is proposed to enhance the efficacy and selectivity of cytotoxic agents against cancer cells by utilizing the differences in morphology and microenvironment between tumor and healthy cells. The developed enhanced formulations of cytotoxic agents, including efflux inhibitors and ion channel inhibitors, demonstrate increased permeability in A549 cells and a protective effect on healthy HEK293T cells.

Summary: Soil is the major reservoir of antibiotic resistance genes (ARGs), particularly multidrug efflux pump genes, which have raised concerns due to their high prevalence in natural soil ecosystems. This study examined the influence of soil pH on the selection and composition of multidrug efflux pump genes in soil resistomes. The findings indicate that acidic soil environments significantly enrich multidrug efflux pump genes, reflecting the benefits of high soil proton activity on these genes.

Summary: This study develops a glycan-targeting photosensitizer, ACR-DMP, to fight against stubborn biofilms. ACR-DMP has high extracellular polymer penetrability and overcomes the hypoxic microenvironment, efficiently eradicating biofilms in vitro and in vivo. It not only changes membrane potential homeostasis and osmotic pressure balance, but also inhibits quorum sensing, two-component system, and efflux pump to conquer biofilm resistance.

Summary: Salvia officinalis L., Sambucus nigra L., Matricaria chamomilla L., Agrimonia eupatoria L., Fragaria vesca L., and Malva sylvestris L. are plants that have been used in European folk medicine for treating skin wounds and infections. This study investigated their potential as adjuvants for the treatment of antibiotic-resistant bacteria. The results showed that Malva sylvestris had the highest antimicrobial activity, while Fragaria vesca and Sambucus nigra demonstrated potential as efflux pump inhibitors. Overall, these plants have potential for improving antibiotic treatment.
