This paper reviews the main applications of NMR metabolomics of blood and urine in disease research, over the last 5 years. The broad range of disease types addressed attests the increasing interest within the academic and medical communities to explore the recognised potential of metabolomics to (1) provide insight into underlying disease pathogenesis and (2) unveil new metabolic markers for disease diagnosis and follow up. Importantly, most recent studies reveal an increasing awareness of possible limitations and pitfalls of the metabolomics approach, together with efforts for improved study design and statistical validation, which are crucial requisites for the sound development of NMR metabolomics and its progress into the clinical setting. PM2.5 chemical composition and health risks by inhalation near a chemical complex

Summary: This study collected PM2.5 samples near an industrial chemical pole and analyzed their OC, EC, trace elements, and organic constituents. The results showed that organic compounds, elemental carbon, and trace elements mainly originated from industrial emissions, nearby roads, and biomass burning. Some organic constituents were likely related to plasticizer ingredients and thermal stabilizers in industrial emissions. Additionally, principal component analysis revealed the contributions of different emission sources.

Clinical relevance of the blanking period on late recurrence after catheter ablation of atrial fibrillation

Summary: This study aimed to determine the predictors of early recurrence (ER) and late recurrence (LR) after catheter ablation (CA) for atrial fibrillation (AF) and establish the risk of LR in patients who experience ER. The results showed that the risk of LR after CA was significantly higher in patients with ER (five-fold increased risk). These findings highlight the importance of clarifying the clinical role of the blanking period.

Nutritional, rheological, sensory characteristics and environmental impact of a yogurt-like dairy drink for children enriched with lupin flour

Summary: Studies show that adding pulses to foods increases their nutritional value. This study focuses on lupin flour's effects on yogurt, finding that lupin-enriched yogurt is high in protein, fiber, and omega 3. Rheological tests also indicate that this yogurt is stiffer and more viscous. Sensory analysis shows no significant differences compared to controls, but bitterness, granularity, and after-taste are observed. Using lupin flour to enhance yogurt production could be an alternative for creating more nutritious products.

Summary: Studies on the therapeutic properties and metabolomic profile of camel urine are limited and lack standardization. Standardization and interdisciplinary approaches are needed to improve the interpretation and replicability of results. Sample enlargement can help evaluate intra- and interindividual variability and distinguish factors that may affect the composition and bioactivity of camel urine.

Effect of Pd2Spermine on Mice Brain-Liver Axis Metabolism Assessed by NMR Metabolomics

Summary: This study used Nuclear Magnetic Resonance metabolomics to compare the effects of Pd(2)Spermine and cisplatin on the brain and liver. The results showed that Pd(2)Spermine had a stronger impact on the brain metabolome compared to cisplatin. Both drugs did not cause neuronal damage or inflammation. Pd(2)Spermine seemed to enhance brain anti-inflammatory and antioxidant mechanisms, while cisplatin caused more liver damage and increased the need for liver regeneration processes.

An Intracellular Metabolic Signature as a Potential Donor-Independent Marker of the Osteogenic Differentiation of Adipose Tissue Mesenchymal Stem Cells

Summary: This paper presents an untargeted NMR metabolomics study on the proliferation and osteogenic differentiation of human adipose mesenchymal stem cells (hAMSCs), aiming to identify potential metabolic markers that are dependent on or independent of cell donors. By analyzing the polar endometabolome of hAMSCs from two donors with distinct characteristics, the study suggests an 18-metabolites signature for cell proliferation and a set of 11 metabolites for osteogenesis. These signatures have been validated in a third donor and may contribute to monitoring and predicting the proliferation and osteogenic ability of different donors.

Metabolomics Insights into the Differential Response of Breast Cancer Cells to the Phenolic Compounds Hydroxytyrosol and Luteolin

Summary: The aim of this study was to investigate the effects of two phenolic compounds, hydroxytyrosol (HT) and luteolin (LUT), on the metabolism of breast cancer cells of different molecular subtypes. The researchers used untargeted metabolomics to study the metabolic responses of MDA-MB-231 and MCF-7 cells to treatment with these phenols. The results showed that the effects of the phenolic compounds varied depending on the cell type and included changes in mitochondrial metabolism, glycolysis, lipid metabolism, and cellular redox status.

Metabolic Reprogramming of Breast Tumor-Educated Macrophages Revealed by NMR Metabolomics

Summary: Tumor-associated macrophages (TAMs) are the most abundant immune cells in the breast tumor microenvironment, and their immune-suppressive functions promote breast cancer progression. The metabolic crosstalk between tumor cells and TAMs is important in modulating the tumor microenvironment towards anti-tumor immunity. This study investigates how tumor-educated macrophages (TEM) reprogram their metabolism and phenotype in response to different metabolic conditions imposed by breast cancer cell subtypes, providing new insights into the metabolic plasticity of TEM in the breast cancer microenvironment.

Cardiovascular risk factors during pregnancy impact the postpartum cardiac and vascular reverse remodeling

Summary: Pregnant women with cardiovascular risk factors are at high risk of developing cardiovascular disease later in life. Our study found that pregnant women undergo cardiovascular remodeling during pregnancy, which reverses after delivery. Arterial hypertension, smoking, and obesity are independent predictors of increased left ventricular mass. Additionally, circulating biomarkers such as C-reactive protein and ST2/IL33 receptors may be associated with worse cardiac remodeling.

Impact of Sea Warming and 17-a-Ethinylestradiol Exposure on the Lipid Metabolism of Ruditapes philippinarum Clams

Summary: This study investigates the effects of exposure to 17-α-ethinylestradiol (EE2) on lipophilic extracts of Ruditapes philippinarum clams at 17 degrees C and 21 degrees C. The results show that EE2 exposure at 17 degrees C leads to a slight increase in membrane rigidity at low concentrations, while at higher concentrations, lipid metabolism stabilizes. In contrast, at 21 degrees C, lipid metabolism responds to EE2 at 125 ng/L, with the antioxidant docosahexaenoic acid (DHA) helping to mitigate oxidative stress. The study also identifies new potential markers of stress mitigation, such as high levels of phosphatidylcholine (PtdCho), polyunsaturated fatty acids (PUFAs), and linoleic acid, as well as low PUFA/glycine ratios.

Tumor Lipid Signatures Are Descriptive of Acquisition of Therapy Resistance in an Endocrine-Related Breast Cancer Mouse Model

Summary: The lipid metabolism adaptations of estrogen and progesteronereceptor-positive breast cancer tumors in relation to HD to HI tumor growth transition and HIR tumor acquisition of ET resistance were investigated. Results showed increased nonesterified cholesterol forms and phospholipids, and decreased triglycerides and fatty acids during tumor progression. These changes suggest a shift toward more activated cholesterol synthesis, β-oxidation, and phospholipid biosynthesis to sustain tumor growth. Certain compound levels and ratios were identified as potential markers for HD-HI-HIR progression.

A strategy takes Yiqing tablets as an example to carry out simpler multi-component quantification and use fingerprint technology for comprehensive quality consistency evaluation

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

Green sample preparation of medicinal herbs in closed digester block for elemental determination by ICP OES

Summary: This work presents a simple, cost-effective, and environmentally friendly method for determining the content of multiple elements in medicinal herbs using ICP OES. The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

Separation and characterization of related substances of Lurasidone hydrochloride by LC-QTOF-MS techniques

Summary: In this study, a reliable LC-QTOF-MS method was developed to separate and characterize process-related substances and degradation products of Lurasidone hydrochloride. The results showed that Lurasidone hydrochloride exhibited instability under certain stress conditions, and several previously unidentified compounds were discovered. The study emphasizes the importance of monitoring and controlling related substances in the manufacturing processes to optimize the process and ensure quality control.

Summary: This study investigated the cellular uptake of low, medium and high molecular weight polyethylene glycol (PEG) in MCF-7 cells using UPLC-MS/MS and CID techniques. The results showed that PEG600, PEG2K, PEG4K and PEG10K exhibited low toxicity and had limited cellular entry at concentrations of 5-1200 μg/mL. Additionally, the environmental friendliness of the developed analytical methods was evaluated.

Summary: This study employed network pharmacology-based analysis and reverse molecular docking to investigate the molecular targets and pathways associated with gynecological disorders and the potential therapeutic effects of fenugreek constituents. The results highlighted the potential of steroidal saponins, isoflavones, and flavones as active constituents of fenugreek with implications for lactation, reproductive processes, and estrogenic activity. Animal model study further supported the presence of these bioactive compounds and their effects on body weight, serum prolactin levels, serum estradiol levels, and ovarian histological structure.
