In this review, the set-up and data interpretation of experimental designs (screening, response surface, and mixture designs) are discussed. Advanced set-ups considered are the application of D-optimal and supersaturated designs as screening designs. Advanced data interpretation approaches discussed are an adaptation of the algorithm of Dong and the estimation of factor effects from supersaturated design results. Finally, some analytical applications in separation science, on the one hand, and formulation-, product-, or process optimization, on the other, are discussed. (C) 2011 Elsevier B.V. Development of a capillary electrophoresis method for the separation of flavonolignans in silymarin complex

Summary: A capillary electrophoresis (CE) method was developed and validated for the baseline separation of structurally similar flavonolignans in silymarin complex. The method showed good separation efficiency, linear calibration curves, and accurate results under the experimental conditions. The CE analysis results were comparable to the reference HPLC method for determining flavonolignans in dietary supplements.

Evaluating micellar liquid chromatographic methods on octadecyl particle-based and monolithic columns to predict the skin permeation of drug and cosmetic molecules

Summary: A micellar liquid chromatographic method was developed to model skin permeability of pharmaceutical and cosmetic compounds. The method allowed for analysis of a large test set of compounds with just one mobile-phase composition, providing useful models with similar or even slightly better performance parameters on a monolithic column as compared to a particle-based column. The method on the monolithic column also allowed for faster analysis, making it ideal for estimating skin permeability.

Natural plant toxins in honey: An ignored threat to human health

Summary: Consumers often believe that natural food is harmless, however naturally occurring toxins in food represent a health risk to humans. Toxic honey, collected by bees from poisonous plants, contains plant toxins such as grayanotoxins and triptolides that can be fatal if consumed. Understanding the toxicity mechanisms of different toxins, preventing toxic honey from entering the food chain, and developing effective antidotes and treatments are critical challenges.

Fabrication of a molecularly imprinted monolithic column via the epitope approach for the selective capillary microextraction of neuropeptides in human plasma

Summary: In this study, a novel molecularly imprinted monolithic column was successfully synthesized for the selective capillary microextraction of neumtensin and neummedin N. The column exhibited excellent adsorption capacity and selectivity towards the target peptides, and the proposed method proved to be effective for the specific analysis of NT and NmN in human plasma samples.

Summary: In this study, the compounds Nemorosine A (1) and fargesine (2) were explored for their pharmacological effects on neurodegenerative disorders using a combined in silico-in vitro-in vivo approach. The compounds showed agonistic effects on the 5-HT2A receptor and demonstrated potential in reducing proteotoxicity and neurodegeneration in Caenorhabditis elegans models. These results contribute to the understanding of the compounds' multitarget profiles and support their potential in the treatment of neurodegenerative disorders.

Comparison of supercritical fluid chromatographic methods to predict the skin permeability of pharmaceutical and cosmetic compounds

Summary: Supercritical fluid chromatography (SFC) was investigated as an alternative method to liquid chromatography for predicting the skin permeability of pharmaceutical and cosmetic compounds. Nine different stationary phases were used to screen a test set of 58 compounds. Multiple linear regression (MLR) models were found to perform better than partial least squares (PLS) models. The best correlation with skin permeability data was achieved using retention factors from a cyanopropyl (CN) column. A simple MLR model including the CN-column retention, octanol-water partition coefficient, and number of atoms showed good fit and predictive features.

Improving the LC-MS/MS analysis of neuromedin U-8 and neuromedin S by minimizing their adsorption behavior and optimizing UHPLC and MS parameters

Summary: Neuromedin U (NmU) and neuromedin S (NmS) are neuropeptides belonging to the neuro-medin family. Liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) is the preferred analytical technique for peptide quantification, but quantification limits for these peptides in biological samples remain challenging due to nonspecific binding. This study focuses on solving the adsorption problem and improving the sensitivity of the LC-MS/MS method for NmU and NmS.

Quality Control and Authentication of Argan Oils: Application of Advanced Analytical Techniques

Summary: In addition to its nutritional and therapeutic benefits, Argan oil is highly regarded for its unique bio-ecological and botanic characteristics. It has been traditionally used to treat various health issues and is widely commercialized. However, the intentional adulteration of Argan oil with cheaper oils affects its quality and poses potential health risks to consumers. Analytical techniques are employed to ensure the quality, safety, and authenticity of Argan oil. This review provides a comprehensive overview of studies on the quality assessment of Moroccan Argan oil using both targeted and untargeted approaches, coupled with chemometric techniques to detect adulteration.

Evaluation of Multivariate Filters on Vibrational Spectroscopic Fingerprints for the PLS-DA and SIMCA Classification of Argan Oils from Four Moroccan Regions

Summary: The aim of this study was to develop an analytical method for determining the geographical origin of Moroccan Argan oil using near-infrared (NIR) or mid-infrared (MIR) spectroscopic fingerprints. Unsupervised and supervised classification methods were evaluated to distinguish between Argan oils from different regions. The use of multivariate filters improved the accuracy of the classification models. The findings suggest that these models can be applied in routine monitoring to identify the geographical origin of Argan oils, reducing analysis costs and time.

Modelling the enantiorecognition of structurally diverse pharmaceuticals on O-substituted polysaccharide-based stationary phases

Summary: This study aims to develop models for predicting the retention, separation, and elution sequence of enantiomers in diverse pharmaceuticals. Quantitative Structure Retention Relationships (QSRR) models are built using molecular descriptors and retention. The models show that only achiral descriptors can accurately predict the global retention, while chiral descriptors are not suitable for predicting enantioseparation and elution sequence. Models with both chiral and achiral descriptors can effectively predict retention, but their ability to predict elution sequence and separation varies for different chromatographic systems.

Analytical strategies for herbal Cannabis samples in forensic applications: A comprehensive review

Summary: Cannabis sativa L. is widely used as a recreational drug due to its desired psychotropic effects, and its medical properties have attracted attention. This review discusses the challenges in analyzing herbal Cannabis, and the need for sensitive techniques to identify and quantify the components of interest. It provides detailed information on the analysis of cannabinoids in seizures for forensic purposes, and the advantages and drawbacks of existing methods. The application of chemometrics in interpreting complex data is also explained.

A strategy takes Yiqing tablets as an example to carry out simpler multi-component quantification and use fingerprint technology for comprehensive quality consistency evaluation

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

Green sample preparation of medicinal herbs in closed digester block for elemental determination by ICP OES

Summary: This work presents a simple, cost-effective, and environmentally friendly method for determining the content of multiple elements in medicinal herbs using ICP OES. The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

Separation and characterization of related substances of Lurasidone hydrochloride by LC-QTOF-MS techniques

Summary: In this study, a reliable LC-QTOF-MS method was developed to separate and characterize process-related substances and degradation products of Lurasidone hydrochloride. The results showed that Lurasidone hydrochloride exhibited instability under certain stress conditions, and several previously unidentified compounds were discovered. The study emphasizes the importance of monitoring and controlling related substances in the manufacturing processes to optimize the process and ensure quality control.

Summary: This study investigated the cellular uptake of low, medium and high molecular weight polyethylene glycol (PEG) in MCF-7 cells using UPLC-MS/MS and CID techniques. The results showed that PEG600, PEG2K, PEG4K and PEG10K exhibited low toxicity and had limited cellular entry at concentrations of 5-1200 μg/mL. Additionally, the environmental friendliness of the developed analytical methods was evaluated.

Unraveling the mechanisms of Fenugreek seed for managing different gynecological disorders: steroidal saponins and isoflavones revealed as key bioactive metabolites

Summary: This study employed network pharmacology-based analysis and reverse molecular docking to investigate the molecular targets and pathways associated with gynecological disorders and the potential therapeutic effects of fenugreek constituents. The results highlighted the potential of steroidal saponins, isoflavones, and flavones as active constituents of fenugreek with implications for lactation, reproductive processes, and estrogenic activity. Animal model study further supported the presence of these bioactive compounds and their effects on body weight, serum prolactin levels, serum estradiol levels, and ovarian histological structure.
