Background and Objectives: The potential of salivary microorganisms to diagnose periodontal disease and to guide periodontal treatment is a research topic of current interest. This study aimed to determine whether the salivary counts of periodontopathic microbes correlated with the periodontal pocket counts of the same infectious agents, and whether the salivary counts of the test infectious agents could distinguish among individuals with periodontal health and various types of periodontal disease. Material and Methods: The study included 150 systemically healthy adults, of whom 37 were periodontally healthy, 31 had gingivitis, 46 had chronic periodontitis and 36 had aggressive periodontitis. Each study subject contributed microbial samples from the two deepest periodontal pockets of the dentition and from whole saliva. Aggregatibacter actinomycetemcomitans, Campylobacter rectus, Fusobacterium nucleatum, Porphyromonas gingivalis, Prevotella intermedia, Tannerella forsythia and Epstein-Barr virus were identified using the TaqMan real-time PCR methodology. Statistical analysis was performed using the Mann-Whitney U-test and the receiver operating characteristic statistics. Results: C. rectus, F. nucleatum, P. gingivalis, P. intermedia and T. forsythia occurred with significantly higher copy-counts in salivary samples from patients with gingivitis, chronic periodontitis and aggressive periodontitis than from periodontally healthy individuals. A. actinomycetemcomitans only showed higher salivary copy-counts in subjects with aggressive periodontitis compared with subjects with healthy periodontium, and the salivary copy-counts of Epstein-Barr virus did not reveal any significant difference among the four subject groups studied. The diagnostic sensitivity for periodontitis was 89.19 for P. gingivalis and for T. forsythia and 86.49 for P. intermedia, with specificities ranging from 83.78 to 94.59. The optimal copy-counts per mL saliva for identifying periodontitis were 40,000 for P. gingivalis, 700,000 for T. forsythia and 910,000 for P. intermedia. Conclusion: Salivary copy-counts of P. gingivalis, T. forsythia and P. intermedia appear to have the potential to identify the presence of periodontitis, whereas the salivary level of the other test infectious agents may possess little or no diagnostic utility. Longitudinal studies are warranted to determine the ability of salivary copy-counts of major periodontopathic bacteria to predict future periodontal breakdown.

Maternal omega-3 polyunsaturated fatty acids supplementation in pregnancy decreases MMP-1 levels in breastmilk: a cross-sectional study

Ozlem Naciye Sahin, Aysel Ozpinar, Muhittin Serdar

Summary: The study evaluated levels of MMP-1, TIMP-1, leptin, IL-6, and fatty acids in breastmilk from women who consumed omega-3. It found that MMP-1 levels were significantly lower in breastmilk from omega users, and positively correlated with omega 6:3 ratio, IL-6, and leptin levels. Maternal omega use may modify MMP-1 levels in breastmilk, impacting extracellular matrix remodelling and metabolic programming in early infancy.

Summary: This study aimed to investigate the plasma levels of tryptophan (Trp) and its metabolites in patients with primary Sjogren's syndrome (pSS). The study included 34 pSS patients and 42 healthy individuals, and serum Trp and kynurenine (Kyn) concentrations were measured. The results showed that Trp metabolism was altered in pSS patients, suggesting a possible link between Trp metabolism and the pathogenesis of pSS.

Summary: This study compares the concentrations of arginine and methylated arginine products in serum samples of COVID-19 patients to assess their association with disease severity. The results show that the concentrations of ADMA, SDMA, and L-NMMA in severe patients are significantly higher than in mild-moderate patients and the control group. Additionally, L-NMMA, CRP, and NLR are identified as independent risk factors for severe COVID-19 patients.

Summary: A novel method for measuring serum free light chains (FLCs) using immunoenrichment on micro-affinity chromatography in combination with matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS) detection was developed. The results showed good agreement between the proposed method and the FLC assay, making it a valuable tool for screening, prognostic stratification, and monitoring therapy responses in plasma cell disorders (PCDs).

Summary: This study aimed to evaluate the levels of ADMA, SDMA, and IL-1 beta in GCF from periodontitis patients. The results showed that the levels of IL-1 beta, ADMA, and SDMA in GCF were significantly higher in periodontitis patients compared to the control group, and they were positively correlated with clinical periodontal parameters. It suggests that ADMA and SDMA may be involved in the pathogenesis of periodontal disease.

Do arginine metabolites have a role in periodontitis due to smoking? A new perspective

Melis Yilmaz, Ekin Yay, Nur Atalay, Nur Balci, Sivge Kurgan, Hilal Toygar, Muhittin A. A. Serdar

Summary: This study aimed to determine the levels of arginine metabolites and IL-6 in saliva and serum of individuals with periodontitis and healthy individuals, and to evaluate their association with smoking status. The results showed that salivary concentrations of ADMA and SDMA were associated with periodontitis, and smoking increased the levels of arginine metabolites in the serum of periodontitis patients.

Summary: This study aimed to evaluate the levels of adipokines in the gingival crevicular fluid (GCF) of periodontitis patients and compare them before and after non-surgical periodontal treatment. The results showed that the levels of visfatin and chemerin were significantly higher in the periodontitis group compared to the healthy group. After non-surgical periodontal treatment, the levels of chemerin significantly decreased. These findings suggest that visfatin and chemerin may play a role in the periodontal disease process.

Does smoking influence tryptophan metabolism in periodontal inflammation? A cross-sectional study

Canan Onder, Nihan Akdogan, Sivge Kurgan, Nur Balci, Ceyhan Ceran Serdar, Muhittin A. Serdar, Meral Gunhan

Summary: The aim of this study was to investigate the effects of smoking and periodontal inflammation on tryptophan-kynurenine metabolism and their correlation with clinical periodontal parameters. The study found that smoking exacerbates inflammation in the periodontium, leading to increased tryptophan destruction and decreased IDO activity by suppressing the kynurenine pathway (KP) in serum. Therefore, kynurenine and its metabolites may serve as significant biomarkers connecting smoking and periodontal disease.

Summary: This study aims to reveal the role of gut microbiota in the pathogenesis of Alzheimer's Disease (AD), particularly regarding changes in the proteome and molecular pathways. The data indicates that the microbiota, proteome, and molecular changes exist long before the manifestation of disease symptoms. Moreover, disparities exist between the normal-aged flora and the gut microbiota of late-stage AD mice, emphasizing the potential value of identified phyla, proteins, and pathways as markers for the early and late stages of AD.
