The methylator phenotype in microsatellite stable colorectal cancers is characterized by a distinct gene expression profile

The CpG island methylator phenotype (CIMP) in colorectal tumours can be recognized by an increased frequency of aberrant methylation in a specific set of genomic loci. Because of the strong association of CIMP with high microsatellite instability (NISI-H), the identification of CIMP+ tumours within microsatellite stable (MSS) collorectal cancers may not be straightforward. To overcome this potential limitation, we have built an improved seven-locus set of methylation markers that includes CACNA1G, IGF2, RUNX3, HTR6, RIZ1, MINT31, and MAP1B. This new set of CIMP markers revealed a bimodal distribution of methylation frequencies in a group of 95 MSS colorectal cancers, which allowed a clearer separation between CIMP classes. Correlation of MSS CIMP+ tumours with bio-pathological traits revealed significant associations with location to the proximal colon, mucinous histology, BRAF mutation, and chromosomal stability. A potential trend towards an adverse prognosis of CIMP+ cases was associated with the high frequency of BRAF mutations present within this cohort of tumours. Microarray analysis revealed that CIMP+ tumours are characterized by a unique expression profile, a result that confirms that CIMP+ tumours represent a truly distinct molecular class within MSS colorectal cancers. Overexpression of ultraconserved region 83-induces lung cancer tumorigenesis

Summary: The expression of non-coding RNAs is dysregulated in human cancers, particularly the transcribed ultraconserved regions (T-UCRs). In lung cancer, uc.83-, an antisense ultraconserved element, is upregulated and plays a role in cell growth. AKT and ERK 1/2 signaling pathways are involved in the oncogenic function of uc.83-. Inhibition of uc.83- could be a potential therapeutic treatment for non-small cell lung cancer.

Sickle Cell Trait and SARS-CoV-2-Induced Rhabdomyolysis: A Case Report

Summary: This study presents a case of rhabdomyolysis caused by SARS-CoV-2 infection. The patient had unknown sickle cell trait and developed acute kidney injury, requiring acute hemodialysis. Pathological examination confirmed rhabdomyolysis injury, but no viral traces were found in the kidney biopsy samples. The mechanisms of SARS-CoV-2-induced rhabdomyolysis are discussed, and it is suggested to test for sickle cell trait in African patients.

PML at mitochondria-associated membranes governs a trimeric complex with NLRP3 and P2X7R that modulates the tumor immune microenvironment

Summary: Uncontrolled inflammatory response from the tumor microenvironment significantly contributes to cancer progression. A trimeric complex at the mitochondria-associated membranes, involving P2X7 receptor-NLRP3 inflammasome interaction, is regulated by the tumor suppressor PML. Downregulation of PML leads to an exacerbated immune response and boosted tumor growth. Mislocalization of PML results in uncontrolled NLRP3 activation and cytokine production, fueling cancer development and worsening prognosis in various human cancers. Mechanistic insights into the PML-P2X7R-NLRP3 axis provide new opportunities for targeted therapeutic approaches in human carcinogenesis.

The results revealed a higher percentage of SARS-CoV-2 NP positive placental samples in the group positive for SARS-CoV-2 at delivery compared to the negative group.

Identifying effective diagnostic and prognostic biomarkers, such as microRNAs (miRNAs), for LSCC is crucial for disease management and improving clinical outcomes. This systematic review provides an overview of tissue and circulating miRNAs and their potential role in diagnosing and predicting LSCC.

Summary: The aim of this systematic literature review was to investigate the effects of b/tsDMARDs on the synovial membrane of PsA patients and identify biomarkers of response to therapy. A meta-analysis of 22 studies showed a significant reduction in CD3+ lymphocytes and CD68+ macrophages in synovial biopsies from patients treated with bDMARDs for 4-12 weeks. Reduction in CD3+ was correlated with clinical response. The most consistent finding in the literature was the reduction in CD3+/CD68+ sublining cells during the first 3 months of treatment with TNF inhibitors.

A Systematic Review of Diagnostic and Prognostic Biomarkers for Head and Neck Cancer of Unknown Primary: An Unmet Clinical Need

Summary: Head and neck cancer of unknown primary (HNCUP) refers to cervical lymph node metastases without an identifiable primary tumor. The management of HNCUP patients is challenging due to controversial guidelines in diagnosis and treatment. This systematic review presents available data on diagnostic and prognostic molecular biomarkers for HNCUP, with HPV and EBV being the only biomarkers currently used in clinical practice. Improved molecular profiling and development of tissue-of-origin classifiers are necessary for better diagnosis, staging, and therapeutic management of HNCUP patients.
