Tumor Necrosis Factor-Alpha Antagonist Administration Recovers Skeletal Muscle Dysfunction in Ovariectomized Rats

Skeletal muscles deteriorate after ovariectomy. Molecular pathway of this deterioration has not been defined. Tumor necrosis factor (TNF)-alpha activation is assumed to trigger muscle atrophy and administration of its antagonist is hypothesized to recover this atrophy in rats. Slow-twitch soleus and fast-twitch extensor digitorum longus muscle functions were investigated in intact, ovariectomized (OVX), and OVX plus 10 mu g/g/week TNF-alpha antagonist administered female rats. Maximum isometric twitch and tetanic contraction responses were lower in the OVX groups. Maximum isometric twitch amplitudes recovered in the extensor digitorum longus but not in the soleus muscles after TNF-alpha antagonist administration. The decrease in responses to tetanic stimulations recovered in the OVX TNF group at frequencies higher than 20 Hz in both muscle types. OVX animals body weight was 21% higher than intact animals. Muscle weight to body weight ratios of the OVX groups were higher than the control group which recovered after TNF-alpha antagonist administration. Findings suggest that the functional loss in OVX rat muscles is TNF-alpha pathway dependent. Skeletal muscle atrophy and function after OVX recovered by TNF-alpha antagonist administration. Summary: This study evaluated the effects of single and combined exposures to BPA and/or DEHP during the prenatal and lactational period on the male reproductive system of rats in later stages of life. The results suggest that prenatal and lactational exposure to BPA and DEHP may have adverse effects on the male reproductive system in later stages of life, especially after combined exposure.

Boron Nano-hydroxyapatite Composite Increases the Bone Regeneration of Ovariectomized Rabbit Femurs

Summary: Boron combined with nano-hydroxyapatite can stimulate new bone formation. Injecting boron-containing nano-hydroxyapatite composites with hyaluronan into rabbit femurs can increase bone mineral density and new bone formation. This treatment shows promise in preventing femoral fractures in osteoporosis patients.

Summary: This study found that the HEp-2 human laryngeal squamous cell carcinoma cell line releases the major endocannabinoids AEA and 2-AG, and their exogenous supplementation inhibits tumor cell proliferation in vitro. 2-AG exhibited a stronger anti-proliferative effect compared to AEA, suggesting that they may function through different pathways.

Differential role of prolyl hydroxylases on mitochondrial function of HL-1 cells in a model of iron deficiency

Mesenchymal stem cells promote spermatogonial stem/progenitor cell pool and spermatogenesis in neonatal mice in vitro

Comparative analysis of magnetically activated cell sorting and ultracentrifugation methods for exosome isolation

Thin-Walled Commercially Pure Titanium Structures: Laser Powder Bed Fusion Process Parameter Optimization

Summary: The aim of this study is to develop a comprehensive process for the production of Cp-Ti bulk and thin structures using L-PBF technology. By varying the process parameters, the optimal parameters for producing high density Cp-Ti parts were determined. The results showed that L-PBF is a favorable manufacturing technology for Cp-Ti parts with almost full density, good mechanical properties, and good dimensional accuracy, even on thin geometries.

A pilot study: Nano-hydroxyapatite-PEG/PLA containing low dose rhBMP2 stimulates proliferation and osteogenic differentiation of human bone marrow derived mesenchymal stem cells

Summary: In this study, we combined nano-hydroxyapatite-polyethylene glycol/poly(lactic acid) (nHAp-PEG/PLA) with recombinant human bone morphogenetic protein 2 (rhBMP2) and evaluated the effects of 0.5, 1.0, and 3.0 μg/mL rhBMP2 on the proliferation, differentiation, and osteogenic genes of human bone marrow mesenchymal stem cells (hBMMSCs). The results showed that nHAp-PEG/PLA composites with 1.0 μg/mL and 3.0 μg/mL rhBMP2 promoted hBMMSC proliferation and differentiation.

Development and validation of a sensitive assay for the quantification of arachidonoylcyclopropylamide (ACPA) in cell culture by LC-MS/MS

Summary: Synthetic and natural cannabinoid derivatives have potential as drug candidates for various diseases, including cancer. ACPA, a synthetic cannabinoid, shows anticancer effects on lung, pancreatic, and endometrial cancer. Therefore, it is crucial to quantify ACPA in biological samples for monitoring its effects. However, the short half-life and lack of UV signal of ACPA present challenges in its quantification. In this study, a sensitive and selective LC-MS/MS method was developed for quantification of ACPA in cell culture medium and intracellular matrix.
