Solution equilibrium studies on anticancer ruthenium(II)-η6-p-cymene complexes of 3-hydroxy-2(1H)-pyridones

Ru-II(eta(6)-p-cymene) complexes of bidentate (0,0) alkoxycarbonylmethyl-3-hydroxy-2(1H)-pyridone ligands exhibit in vitro antitumor activity. We determined the stoichiometry and stability in aqueous solution of two examples by pH-potentiometry, H-1 NMR spectroscopy and UV-vis spectrophotometry and also characterized the proton dissociation processes of the ligands. Formation of mono-ligand complexes with moderate stability was found to predominate in the physiological pH range. Moreover, the chlorido/aqua co-ligand exchange processes of the [Ru-II(eta(6)-p-cymene)(L)(H2O)](+) species were also monitored and 55-65% of the aqua ligand was found to be replaced by chloride in 0.2 M KCl-containing aqueous solutions. Under basic conditions, the complexes decompose to dinuclear tri-hydroxido-bridged [Ru-2(I)(eta(6)-p-cymene)(2)(OH)(3)](+) and metal-free ligand and also a hydroxido species [Ru-II(eta(6)-p-cymene)(L)(OH)] was found. Furthermore, the ligands contain an ester functional group, which may hydrolyze at basic pH, which is however negligible at acidic or neutral pH. (C) 2012 Elsevier B.V. All rights reserved.

Platinum(terpyridine) complexes with N-heterocyclic carbene co-ligands: high antiproliferative activity and low toxicity in vivo

Summary: Pt(terpyridine) complexes are known for their ability to intercalate with DNA. The addition of an NHC co-ligand significantly increased the anticancer activity of the complex compared to its chlorido derivative. While the compound showed high efficacy, it was well-tolerated by zebrafish embryos, especially when compared to cisplatin. DNA interaction studies suggest that the mode of action is related to intercalation.

Evaluation of In Vitro Distribution and Plasma Protein Binding of Selected Antiviral Drugs (Favipiravir, Molnupiravir and Imatinib) against SARS-CoV-2

Summary: Uncertainties exist in plasma protein binding and blood distribution of the COVID-19 therapeutics favipiravir (FAVI), molnupiravir (MOLNU), and imatinib (IMA). This study investigated the proton dissociation processes, solubility, lipophilicity, and serum protein binding of these drugs. The results showed different degrees of lipophilicity at different pH values. MOLNU was hydrolyzed by blood serum proteins, while FAVI had weak binding to blood proteins and IMA showed high affinity binding to AGP.

Estradiol-Based Salicylaldehyde (Thio)Semicarbazones and Their Copper Complexes with Anticancer, Antibacterial and Antioxidant Activities

Summary: A series of novel estradiol-based salicylaldehyde (thio)semicarbazones and their Cu(II) complexes were synthesized and characterized. The Cu(II)-estradiol-semicarbazone complex structure was determined by X-ray crystallography. The Cu(II) complexes of estradiol-TSC and its N,N-dimethyl derivative showed high cytotoxicity against cancer cells and significant activity against Staphylococcus aureus bacteria strain. Estradiol-TSCs exhibited efficient antioxidant activity, which was decreased by complexation with Cu(II) ions. The exchange of estrone moiety to estradiol did not significantly affect the physicochemical and biological properties.

Design, Synthesis, and Evaluation of Biological Activity of Ferrocene-Ispinesib Hybrids: Impact of a Ferrocenyl Group on the Antiproliferative and Kinesin Spindle Protein Inhibitory Activity

Summary: Derivatives of ispinesib and its (S) analogue, containing ferrocenyl moieties or bulky organic substituents, were prepared to combine multiple biologically-active components in a single molecule. Several of these compounds demonstrated higher antiproliferative activity than ispinesib, with nanomolar IC50 values against cell lines. The improved activity may be attributed to synergic effects of various factors such as KSP inhibitory activity due to the ispinesib core and ability to generate ROS and induce mitotic arrest.

Ferrocene-Derived Palladium(II)-Based Metallosupramolecular Structures: Synthesis, Guest Interaction, and Stimulus-Responsiveness Studies

Summary: Using ferrocene-based ligand systems, a series of heterobimetallic architectures were designed to release and bind guest molecules. The complexes were characterized using various techniques and the disassembly and reassembly process of metallosupramolecular structures were investigated using NMR spectroscopy. Guest binding studies revealed interactions with anionic guests and the release of guests was induced by Cl- ions.

Anticancer Ru and Os complexes of N-(4-chlorophenyl) pyridine-2-carbothioamide: Substitution of the labile chlorido ligand with phosphines

Summary: The substitution of the labile chloride ligand with phosphorus donor ligands in half-sandwich MII(cym)Cl complexes showed that Ru and Os complexes with triphenylphosphine ligands exhibited the most potent anticancer activity in submicromolar range against four cancer cell lines. This suggests that the antiproliferative activity is mainly dependent on the lipophilic properties of the phosphine ligand.

In Situ Bioconjugation of a Maleimide-Functionalized Ruthenium-Based Photosensitizer to Albumin for Photodynamic Therapy

Summary: Maleimide-containing prodrugs can selectively react with circulating serum albumin, leading to longer blood circulation times and improved tumor accumulation. This study synthesized maleimide-functionalized complexes and investigated their interaction with albumin. The complexes showed lower cytotoxicity in noncancerous cells under dark conditions but did not result in a significant increase in tumor accumulation.

On-Resin Conjugation of the Ruthenium Anticancer Agent Plecstatin-1 to Peptide Vectors

Summary: Researchers conjugated a derivative of plecstatin-1 with peptides to enhance their targeting ability towards cancer cells. Metal-peptide conjugates were successfully synthesized and tested for their anticancer activity. Although the activity was relatively low, this study provides a potential strategy to design other metal complexes utilizing the targeting properties of peptides.

Critical evaluation of cell lysis methods for metallodrug studies in cancer cells

Summary: This study aimed to investigate different protocols for efficient and reproducible lysis of cancer cells and found that chemical lysis with the RIPA buffer was the most efficient method. Minimal differences were found for experiments measuring the overall cell uptake of the Ru complex.

Exploiting reduced-symmetry ligands with pyridyl and imidazole donors to construct a second-generation stimuli-responsive heterobimetallic [PdPtL4](4+) cage

Summary: A new sequential metalation strategy has been developed for the assembly of a more stable heterobimetallic [PdPtL4](4+) cage. By using a low-symmetry ditopic ligand (L) with imidazole and pyridine donor units, a [Pt(L)(4)](2+) open-cage complex was selectively formed and then reacted with Pd(ii) ions to assemble the cage C. Various spectroscopy and crystallography techniques confirmed the formation of the cage and its heterobimetallic structure. The cage C exhibited anionic guest molecule binding capability and demonstrated stimulus-responsive behavior.

Platinated azulenes: Structures and electronic spectroscopy

Summary: In this study, eight PtII-azulene complexes were synthesized to investigate the effect of metalation on the electronic structures, spectroscopy, and luminescence of organic chromophores. The results showed that the attachment of Pt substituents to azulene affected the HOMOs, LUMOs, and luminescence properties. The study provides insights into the role of metalation in modifying the properties of organic chromophores.

Phosphine oxide donor-based dinuclear rhenium(I) metallocycles

Summary: This study synthesized two types of neutral binuclear metallocycles using a one-pot solvothermal method and characterized their structures. The analysis revealed that these complexes adopt a binuclear heteroleptic cyclic architecture and belong to phosphine oxide donor (P=O)-based supramolecular coordination cyclic complexes.

Al- driven deoxydehydration (DODH) of glycols in the presence of pincer type mo-catalyst

Summary: This study demonstrates the effectiveness of zero-valent elements as reductants for the deoxydehydration reaction of polyols catalyzed by ONO-pincer ligated molybdenum dioxo. The insolubility and easy separability of the reductants and their oxidized products during the reaction are emphasized. Additionally, the choice of ligand has an impact on the catalytic performance of the reaction.

New ruthenium/phosphino carbazole systems for selective preparation of alcohols under hydroformylation conditions

Summary: Efficient Ru-catalyzed hydrogenation of aldehydes was achieved using carbazole-derived phosphine ligands, with the crucial role of N-H moiety in promoting the catalytic reaction. The method was successfully applied in the Co/Ru-catalyzed hydroformylation process, resulting in the production of long-chain alcohols with excellent yields.

Bis-oxazoline derivatives as ancillary ligands for bis-cyclometalated iridium complexes

Summary: Organometallic iridium complexes with two cyclometalated ligands and one bis-oxazoline derived ancillary ligand were studied. These complexes have emission spanning across the visible and near-ultraviolet region, with moderate to high photoluminescence quantum yields. The (ppy)2Ir(BOX-CN) complexes were resolved into Delta- and Lambda-diastereomers based on solubility differences.

Sonochemical synthesis of 1,2,4,5-tetrasubstituted imidazoles catalyzed by reusable reactive magnetic cobalt copper ferrite nanoparticles as a synergistic effect

Summary: In this research, a nanostructured CoCuFe2O4 catalyst was prepared using a brief and useful synthetic strategy. This catalyst was used for synthesizing 1,2,4,5-tetrasubstituted imidazole derivatives under ultrasonic irradiation. The approach offers advantages such as high efficiency, short reaction times, simplicity of the reaction medium, accelerated reaction rates, and easier separation and purification of products.

Summary: The activity of three Pd-PEPPSI catalyst precursors with N-heterocyclic carbene (NHC) ligands derived from caffeine and theophylline in Mizoroki-Heck reactions was tested. A low cost and energy consumption Ultra-microporous CPM-20(Co) for CO2/N2 and CO2/CH4 separation

Metal-organic frameworks (MOFs) are commonly used in adsorption due to their high surface area, porosity, and chemical properties. An ultra-microporous MOF CPM-20(Co) with mixed ligand was synthesized using inexpensive ligands. The material exhibited high selectivities and low regenerative energy, making it a promising option for CO2 separation in industries.
