Summary: A concise synthetic route with good overall yield was developed for the total synthesis of five pyrrolo-azepine containing alkaloids, including aspastipuline and portulacatone, from a commercially available starting material, ethyl 1H-pyrrole-2carboxylate, via a key step of TFAA mediated intramolecular Friedel-Crafts acylation. These alkaloids exhibit significant anticancer properties and moderate antiinflammatory activities.

Summary: In this study, the addition of alpha-ketoamide top-quinone methide initiated by dialkylphosphite in the presence of organic base DBU was explored. The coupling of dialkylphosphites to alpha-ketoamides followed[1,2]-phospha-Brook rearrangement, generating alpha-phosphonyloxy enolates that were subsequently seized by p-quinonemethides. This two-step one-pot 1,6-conjugate addition method provides effective access to a series of isatin-incorporated phosphate-bearing 1,6-adducts with high yield and selectivity.

Summary: This study successfully obtained linear poly-cardanol (LPCA) through a Friedel-Crafts alkylation reaction, and found that LPCA exhibited elastomer characteristics when co-crosslinked with liquid polyisoprene rubber (LIR). LPCA showed the potential to partially or completely replace rubber.

The trans-selective installation of the homoallylic alcohol side-chain at C-15 allowed the successful total syntheses of antirhine and its known epimer.

Summary: In this study, a catalytic conversion of B-X to B-H bond was achieved using a small amount of [Ph3C](+) as an initiator with Et3SiH. This method was successfully applied to six haloboranes. Additionally, C-F bond activation of benzyl fluorides was observed in the presence of arenes, leading to the formation of Friedel-Crafts products using B-H-9-BBN in the presence of Et3SiH. The protocol provides a simple, cost-effective, and mild route to 1,1-diarylmethanes from benzyl fluorides with high yields (up to 99%).

Summary: This research presents the total synthesis of (+)-pancratistatin in 10 linear steps using a known aldehyde as a chiral source. The synthetic route involves a highly stereoselective intermolecular Michael addition and intramolecular Henry reaction to construct a cyclohexane ring with 6 successive stereocenters. All of the synthetic steps are reliable and efficient and can be easily scaled up, allowing for pharmacological tests of (+)-pancratistatin. Importantly, this study discovered a new pharmacological mechanism of action where (+)-pancratistatin is able to inhibit the activity of topoisomerase I, suggesting its potential as a new type of Topo I inhibitor.

Summary: Here we report an asymmetric organocatalyzed method for the Friedel-Crafts reaction between activated phenols and trihaloacetaldehydes. A screening of 41 compounds identified a catalyst structure based on 3,5-dinitrobenzamide of 9-amino-epi-cinchonidine as the lead catalytic molecule. Under optimized conditions, the catalyst provided trihalohydroxyalkylated adducts in good yields and with high enantiomeric ratios. The reaction scope was determined on 29 entries and several follow-up transformations of the enantioenriched products were accomplished.

Summary: The total syntheses of Stemona alkaloids sessilifoliamides B and D, as well as the second synthesis of sessilifoliamide C, were successfully completed from a simple pyrrole substrate. The bicyclic lactam core was prepared on a gram scale via a Bronsted acid mediated cyclization and controlled oxidation with Dess-Martin periodinane. Yield percentages for the synthesis of sessilifoliamides B, C, and D were 24%, 13%, and 17% respectively, over multiple steps.

The synthetic strategy involved a copper-catalyzed [3 + 2] annulation reaction and a Pictet-Spengler cyclization.
