Flavonoids target a variety of pathophysiological mechanisms and are therefore increasingly considered as compounds encompassed with therapeutic potentials in diseases such as cancer, diabetes, arteriosclerosis, and neurodegenerative diseases and mood disorders. Hops (Humulus lupulus L) is rich in flavonoids such as the flavanone 8-prenylnaringenin, which is the most potent phytoestrogen identified so far, and the prenylchalcone xanthohumol, which has potent tumor-preventive, anti-inflammatory and antiviral activities. In the present study, we questioned whether hops-derived prenylflavonoids and synthetic derivatives thereof act on neuronal precursor cells and neuronal cell lines to induce neuronal differentiation, neurite outgrowth and neuroprotection. Therefore, mouse embryonic forebrain-derived neural precursors and Neuro2a neuroblastoma-derived cells were stimulated with the prenylflavonoids of interest, and their potential to activate the promoter of the neuronal fate-specific doublecortin gene and to stimulate neuronal differentiation and neurite outgrowth was analyzed. In this screening, we identified highly neuroactive compounds, which we termed enhancement of neuronal differentiation factors (ENDFs). The most potent molecule, ENDF1, was demonstrated to promote neuronal differentiation of neural stem cells and neurite outgrowth of cultured dorsal root ganglion neurons and protected neuronal PC12 cells from cobalt chloride-induced as well as cholinergic neurons of the nucleus basalis of Meynert from deafferentation-induced cell death. The results indicate that hops-derived prenylflavonoids such as ENDFs might be powerful molecules to promote neurogenesis, neuroregeneration and neuroprotection in cases of chronic neurodegenerative diseases, acute brain and spinal cord lesion and age-associated cognitive impairments. Summary: Pericytes (PCs) play a crucial role in vascular development and stability. In this study, two inducible reporter mouse models were used to investigate the expression efficiency of the tdTomato reporter gene in PCs. The results showed that both models successfully labeled PCs, but with different induction efficiencies. These models represent promising tools for fate-mapping studies of PCs, although one model showed additional labeling of Muller cells, which may hinder the analysis of reporter-positive PCs.

Neuron-glia (mis)interactions in brain energy metabolism during aging

Summary: Life expectancy is increasing, resulting in a growing aging population. One of the factors related to inefficient brain function during aging is hypometabolism. This review focuses on the metabolic interactions between glial cells and neurons, particularly during aging. Aging alters the main metabolic exchange machinery, leading to inefficient levels of nutrients, detrimental mitochondrial activity, high reactive oxygen species levels, reduced ATP production, and a highly inflammatory environment. Ageing is a crucial risk factor for developing neurodegenerative/neuroinflammatory diseases.

Multi-Level Analysis of Adipose Tissue Reveals the Relevance of Perivascular Subpopulations and an Increased Endothelial Permeability in Early-Stage Lipedema

Additionally, the secretome of SVF cells from lipedema patients was shown to induce endothelial barrier dysfunction in early-stage lipedema, as evidenced by increased leakiness of healthy human primary endothelial cells.

Summary: The study investigated the activation and function of DDR1 in OLs by collagen IV. The results showed that collagen IV could induce DDR1 phosphorylation in OLs derived from both neural stem cells and human oligodendroglial cells. The protein expression of DDR1 decreased during OL differentiation. The addition of collagen IV did not change the number of DDR1+/MBP+ cells but accelerated OL branching.

It is found that the dysfunction of HCN channels contributes to the pathophysiology of axotomized M1LV neurons, but its impact varies among neurons and interacts with other mechanisms.

Metabolic Fingerprinting of Different Sideritis Taxa Infusions and Their Neurogenic Activity

Summary: In recent years, research has shown that Sideritis extracts can improve memory. However, their potential to promote the generation of new neurons through neural stem cell differentiation has not been explored. This study evaluates the neurogenic effects of different Sideritis infusions on neural stem and precursor cells, as well as their impact on cell viability. Metabolic fingerprints were also recorded using various analytical techniques. The results suggest that Sideritis infusions have potent neurogenic potential and may be useful in the treatment of neurodegenerative diseases.

Summary: In this study, the participation and transdifferentiation of endogenous pericyte-derived cells in optic nerve injury were investigated using a lineage tracing reporter mouse model. The results showed the presence of pericyte-derived cells in the lesion, which increased in number over time. The non-vascular associated pericyte-derived cells in the lesion core suggest their participation in the injury response.

Regulatory effect of Epigallocatechin-3-O -gallate on circular RNA expression in mouse liver

Summary: This study investigates the impact of maternal vitamin D levels during pregnancy on infantile eczema. The findings suggest that lower prenatal maternal vitamin D levels increase the risk of eczema in infants aged 0-1 year. Furthermore, the study indicates that infants with eczema have lower expression of the Foxp3 gene in cord blood and decreased placental FOXP3 protein expression. These results suggest that vitamin D may affect the development of infantile eczema through the regulation of gene and protein expression in cord blood and placenta.

The study reveals that FTO is associated with hypothalamic leptin resistance and provides new insight into its role in obesity.

Natural products targeting the Notch pathway have shown potential as chemopreventive and anti-cancer agents, providing a feasible solution to fight against cancer by either alone or in combination with current therapeutic agents.
