Summary: The NF-kappa B family of dimeric transcription factors regulate various biological functions. A study has revealed that the atypical I kappa B protein, Bcl3, plays a crucial role in enhancing the population of the p52:p52 homodimer within the NF-kappa B family. Bcl3 competes with other NF-kappa B subunits for efficient p52:p52 homodimer formation, leading to the upregulation of target genes involved in cell proliferation, migration, and inflammation. The aberrant activation of Bcl3 and p52 contributes to cancer.

Macrophages appear to have a leading role in producing inflammatory factors through NF-kB in the intima, while leukocytes may have a dominant role in the subintima of advanced OA. Blocking NF-kB expression in macrophages and leukocytes could be a potential therapeutic target for modifying the disease.

Summary: The expression of Receptor activator of NF-kappa B (RANK) is increased in podocytes of patients with diabetic nephropathy. Mouse models of podocyte-specific RANK depletion and overexpression showed that RANK depletion alleviated the development of kidney diseases, while RANK overexpression aggravated the condition. The findings suggest that RANK mediates the development of diabetic nephropathy by promoting glomerular oxidative stress and proinflammatory cytokine production.
