Stress Produces Aversion and Potentiates Cocaine Reward by Releasing Endogenous Dynorphins in the Ventral Striatum to Locally Stimulate Serotonin Reuptake

Activation of the dynorphin/kappa-opioid receptor (KOR) system by repeated stress exposure or agonist treatment produces place aversion, social avoidance, and reinstatement of extinguished cocaine place preference behaviors by stimulation of p38 alpha MAPK, which subsequently causes the translocation of the serotonin transporter (SERT, SLC6A4) to the synaptic terminals of serotonergic neurons. In the present study we extend those findings by showing that stress-induced potentiation of cocaine conditioned place preference occurred by a similar mechanism. In addition, SERT knock-out mice did not show KOR-mediated aversion, and selective reexpression of SERT by lentiviral injection into the dorsal raphe restored the prodepressive effects of KOR activation. Kinetic analysis of several neurotransporters demonstrated that repeated swim stress exposure selectively increased the V-max but not Km of SERT without affecting dopamine transport or the high-capacity, low-affinity transporters. Although the serotonergic neurons in the dorsal raphe project throughout the forebrain, a significant stress-induced increase in cell-surface SERT expression was only evident in the ventral striatum, and not in the dorsal striatum, hippocampus, prefrontal cortex, amygdala, or dorsal raphe. Stereotaxic microinjections of the long-lasting KOR antagonist norbinaltorphimine demonstrated that local KOR activation in the nucleus accumbens, but not dorsal raphe, mediated this stress-induced increase in ventral striatal surface SERT expression. Together, these results support the hypothesis that stress-induced activation of the dynorphin/KOR system produces a transient increase in serotonin transport locally in the ventral striatum that may underlie some of the adverse consequences of stress exposure, including the potentiation of the rewarding effects of cocaine.

Isolation of Highly Purified and Viable Retinal Endothelial Cells

Summary: This study established an optimized method for isolating neonatal retinal endothelial cells, resulting in high cell viability and purity. The CD31+/CD45- endothelial cell population isolated through fluorescence-activated cell sorting (FACS) showed enrichment in endothelial cell gene expression and maintained viability for 60 minutes post-FACS. This method is compatible with next-generation sequencing applications and will facilitate further research on mechanisms underlying vascular development and maturation.

A novel differentiation protocol presented here allows for the generation of well-defined hemogenic endothelial cells from human embryonic stem cells in a short timeframe, providing opportunities for further molecular studies and potential therapeutic applications.

Summary: The crosstalk between T cells, dendritic cells, and macrophages in leukocyte clusters within barrier tissues provides a new concept for T cell activation in the skin. It was found that macrophages produce IL-27 which induces IL-15 production from other cells within leukocyte clusters, enhancing the survival of skin T cells. Blocking the IL-27 pathway in CHS mice led to decreased expression of the T cell survival gene BCL2, resulting in a decline in dermal CD8(+) T cells and T cell cluster numbers.

Summary: This study finds that hematopoietic mosaic loss of Y chromosome (mLOY) is associated with increased risk of mortality and age-related diseases in men. The animal experiment results show that male mice reconstituted with bone marrow cells lacking the Y chromosome display increased mortality and age-related profibrotic pathologies including reduced cardiac function. Further research reveals that cardiac macrophages lacking the Y chromosome exhibit polarization toward a more fibrotic phenotype, and treatment with a specific antibody can alleviate cardiac dysfunction. A prospective study also reveals that mLOY in blood is associated with an increased risk for cardiovascular disease and heart failure-associated mortality.

Summary: We provide mechanistic insights into the role of Cx37 in regulating cell cycle modulators and growth arrest. Both the channel-forming and cytoplasmic tail domains of Cx37 are involved in up-regulating p27 and causing late G1 arrest. The cytoplasmic tail domain of Cx37 interacts with activated ERK, leading to stabilization of pERK nuclear target Foxo3a and subsequent up-regulation of p27 transcription.
