Altered Distribution of Juxtaparanodal Kv1.2 Subunits Mediates Peripheral Nerve Hyperexcitability in Type 2 Diabetes Mellitus

Peripheral nerve hyperexcitability (PNH) is one of the distal peripheral neuropathy phenotypes often present in patients affected by type 2 diabetes mellitus (T2DM). Through in vivo and ex vivo electrophysiological recordings in db/db mice, a model of T2DM, we observed that, in addition to reduced nerve conduction velocity, db/db mice also develop PNH. By using pharmacological inhibitors, we demonstrated that the PNH is mediated by the decreased activity of K(v)1-channels. In agreement with these data, we observed that the diabetic condition led to a reduced presence of the K(v)1.2-subunits in juxtaparanodal regions of peripheral nerves in db/db mice and in nerve biopsies from T2DM patients. Together, these observations indicate that the T2DM condition leads to potassium channel-mediated PNH, thus identifying them as a potential drug target to treat some of the DPN related symptoms.

Modelling human blastocysts by reprogramming fibroblasts into iBlastoids

Delta and epsilon tubulin in mammalian development

Summary: Delta and epsilon tubulin, lesser-known relatives of alpha and beta tubulin, likely regulate centriole function across species, with their roles in mammals still mysterious. Mutations in 6- and epsilon-tubulin cause centriole destabilization, atypical mitosis, and cell death, indicating their importance in cellular processes beyond centrioles. They also interact with KATNAL2 during spermatogenesis, suggesting novel functions beyond the centriole.

Iatrogenic immune-mediated neuropathies: diagnostic, epidemiological and mechanistic uncertainties for causality and implications for clinical practice

Summary: Although many reports indicate that medical intervention may lead to acute and chronic immune-mediated neuropathies, only a few drugs such as antitumour necrosis factor alpha agents and immune checkpoint inhibitors are likely to have a causal relationship, while the causality remains largely unsubstantiated for most other suggested iatrogenic causes. Due to the lack of an accurate diagnostic biomarker, clinical assessment often takes precedence over ancillary investigations, casting doubt on the reliability of diagnosis.

Summary: A study conducted by expert physicians from the French Health Care Network Filnemus in 2020 showed a low prevalence of COVID-19 among NMD patients and limited short-term impact on NMD patients. Patients with diabetes, hypertension, or severe forms of NMD were identified as being at higher risk for developing a moderate or severe form of COVID-19. Contrary to expectations, corticosteroids or other immunosuppressants did not appear to increase the risk of more severe COVID-19 in patients with acquired NMD.

Summary: This study aims to assess the feasibility of early detection of Critical Illness Polyneuropathy (CIP) and Myopathy (CIM) in patients with septic shock and prolonged mechanical ventilation through peroneal nerve test (PENT) and sensory sural nerve action potential (SNAP) screening. The results showed that CIP and CIM are common in septic shock patients and can be detected before becoming symptomatic with simple bedside tools.

Summary: The spatial sorting of RNA transcripts in mouse preimplantation embryos plays a crucial role in gene expression and cell fate determination. Basally directed RNA transport is facilitated by microtubules and lysosomes. Despite higher accumulation in basal regions, higher translation activity occurs at the dispersed apical RNA foci, demonstrating spatial heterogeneities in RNA subtypes, organelle interactions, and translation events. Biased inheritance of RNA transcripts and differential translation capacity regulate cell fate allocation during embryonic development.
