Summary: This study shows that overexpression of c-Myc in gastric parietal cells can induce gastric adenoma in mice, and this tumorigenesis process is promoted through the AKT/mTOR pathway. Inhibition of AKT or mTOR can suppress the proliferation of c-Myc overexpressing GC cells and the initiation of gastric tumorigenesis in transgenic mice.

Summary: Human bone marrow-derived mesenchymal stromal cells (MSCs) regulate macrophage activation through upregulation of the Akt/mTORC1 signaling pathway. Inhibition of Akt or mTORC1 attenuates MSCs' effects on macrophage cytokine production, while activation of Akt or mTORC1 enhances these effects. MSCs with cyclooxygenase-2 knockdown have a weaker modulation on macrophage cytokine production.

Summary: Fear-motivated avoidance extinction memory is reconsolidated upon recall through hippocampal BDNF-dependent pathway, which involves activation of mTOR in dorsal CA1. Inhibition of mTOR post-recall hinders memory persistence, while coadministration of recombinant BDNF impedes mTOR inhibition from affecting the memory trace. This suggests that mTOR signaling is essential for fear-motivated avoidance extinction memory reconsolidation and BDNF may act downstream mTOR in a protein synthesis-independent manner.

Summary: This study demonstrates the potential of hesperetin as a mTOR inhibitor in both in silico and in vitro analyses. Hesperetin showed significant inhibitory effects on the phosphorylation of mTOR and downstream molecules in PA-1 and MCF-7 cancer cell lines in a dose-dependent manner.

Summary: Down syndrome (DS) is associated with memory deficits due to hippocampal-related issues, with experimental models showing alterations in synaptic plasticity pathways. Enhanced mGluR-LTD and increased mushroom spines in hippocampal neurons in Ts1Cje mice contribute to memory deficits. Prenatal rapamycin treatment shows positive effects on both phenotypes, suggesting therapeutic potential for DS intellectual disability.

Summary: The isolation of synaptoneurosomes (SNs) is a useful method for studying synaptic events. However, the preparation of SNs is challenging due to variations in synapse size and density in different regions of the central nervous system and different experimental species. In this study, the researchers applied a method originally designed for cortical and hippocampal SNs to isolate SNs from spinal cord tissue in a mouse model of amyotrophic lateral sclerosis (ALS). They found that the isolation of spinal cord SNs required a different gradient fraction compared to cortical/hippocampal SNs. Using proteomics analysis, they also identified an important regulator of local synaptic translation, MNK1, which may play a role in the early stages of ALS.
