IL-6 regulates MCP-1, ICAM-1 and IL-6 expression in human myoblasts

The inflammatory reaction ill autoimmune polymyositis and rejection of transplanted myoblasts is characterized by mononuclear cell infiltration. In other settings monocytes are locally recruited by all IL-6-induced IL-8-to-MCP-1 switch. IL-6, upon binding to soluble gp80 (sIL-6R), can interact with membrane-bound ubiquitously expressed gp130 and activate virtually all cells (transsignaling). We found that human myoblasts could use transsignaling to produce IL-6, MCP-1 and ICAM-1; the addition of sIL-6R, binding to IL-1 beta-induced IL-6, greatly increases IL-6 production. These in vitro data support the hypothesis that locally secreted IL-6 can target monocyte chemotaxis and leukocytes trafficking through an IL-6, MCP-1 and ICAM-1 modulation. (C) 2008 Elsevier B.V. Function and proteolytic generation of the soluble interleukin-6 receptor in health and disease

Summary: Interleukin-6 (IL-6) is a pleiotropic cytokine involved in various physiological and pathophysiological functions. It can signal through different pathways, with trans-signaling playing a crucial role in its pro-inflammatory properties. The soluble form of the IL-6 receptor (sIL-6R) is generated by different mechanisms, including proteolysis. Attempts to selectively block IL-6 trans-signaling are being made in pre-clinical animal models and clinical studies for inflammatory bowel disease.

Summary: Inflammation plays an important role in atherosclerotic cardiovascular disease, and there is a need for therapies targeting excessive inflammation. A fusion protein called olamkicept has been shown to reduce atherosclerosis independently of LDL cholesterol metabolism in mice, and its use in a patient with high-risk ASCVD also demonstrated a reduction in arterial wall inflammation.

Summary: This study reveals that combined hepatocellular carcinoma-cholangiocarcinoma (cHCC-CCA) originates from hepatic progenitor cells (HPCs), not hepatocellular carcinoma (HCC). IL-6 plays a significant role in the aging process of HPCs, suggesting potential therapeutic targets for cHCC-CCA tumors.

Genetic and therapeutic interventions targeting ADAM17 can ameliorate pancreatitis, reducing inflammatory cell infiltration, necrosis, and fibrosis.

Summary: Pulmonary emphysema is associated with dysregulated innate immune responses. In this study, the authors investigated the role of innate immune inflammasome complexes in promoting emphysema. They found that ASC, Caspase-1, and AIM2 were up-regulated in emphysematous patients and mouse models of emphysema. AIM2 was associated with elevated levels of IL-1 beta. Blockade of AIM2 and IL-1 receptor ameliorated emphysema in mice. IL-6 trans-signaling was also found to augment AIM2 expression in emphysema.

Summary: In this study, the researchers identified SEZ6 and gp130/IL6ST as physiological substrates of BACE1 in non-human-primate cerebrospinal fluid. They also found a reduction of gp130 in human CSF from a clinical trial with a BACE inhibitor and in plasma of BACE1-deficient mice. These findings provide insights into the relationship between BACE1 and gp130 in neuronal signaling and survival.

Summary: The organ-specific microbiome, particularly the skin microbiome, plays a crucial role in tissue homeostasis by regulating the inflammatory response. Short chain fatty acids (SCFA) induce regulatory T cells (Treg) in the skin and their signal is mediated by the receptor HCA2. Knock-out of HCA2 results in stronger inflammation and a switch of Treg from a suppressive to a proinflammatory type, indicating that the skin microbiome dictates the outcome of the inflammatory reaction.

Summary: Umbilical cord blood is an alternative source of hematopoietic stem cells for transplantation, but the low number of HSCs in cord blood is a limitation. This study found that the combination of HIL-6, SR1, and UM171 had a synergistic effect on expanding UCB-HSCs. Therefore, this combination is the best choice for obtaining HSCs for clinical transplantation.
