Molecular imaging mass spectrometry for probing protein dynamics in neurodegenerative disease pathology

Recent advances in the understanding of basic pathological mechanisms in various neurological diseases depend directly on the development of novel bioanalytical technologies that allow sensitive and specific chemical imaging at high resolution in cells and tissues. Mass spectrometry-based molecular imaging (IMS) has gained increasing popularity in biomedical research for mapping the spatial distribution of molecular species in situ. The technology allows for comprehensive, untargeted delineation of in situ distribution profiles of metabolites, lipids, peptides and proteins. A major advantage of IMS over conventional histochemical techniques is its superior molecular specificity. Imaging mass spectrometry has therefore great potential for probing molecular regulations in CNS-derived tissues and cells for understanding neurodegenerative disease mechanism. The goal of this review is to familiarize the reader with the experimental workflow, instrumental developments and methodological challenges as well as to give a concise overview of the major advances and recent developments and applications of IMS-based protein and peptide profiling with particular focus on neurodegenerative diseases. By using advanced imaging techniques, the research identified distinct A beta signatures and key A beta species associated with neuritic dystrophy.

Plasma biomarker profiles in autosomal dominant Alzheimer's disease

Summary: The study describes plasma biomarkers in a Swedish cohort of patients with monogenic Alzheimer's disease. Plasma GFAP increases before P-tau181 and NfL, indicating that it reflects early Alzheimer's disease pathology. Plasma biomarkers may be non-invasive tools to detect Alzheimer's disease-related abnormalities. However, further validation is needed.

Longitudinal decline of plasma neurofilament light levels after antiretroviral initiation in people living with HIV

Summary: This retrospective follow-up study investigates the longitudinal change of plasma neurofilament light (NfL) levels after antiretroviral therapy (ART) initiation in people living with HIV. The results show that plasma NfL levels are higher at baseline and decline faster during the follow-up in patients with CD4(+) count 100 cells/μl or higher. There is no significant correlation between baseline HIV RNA levels, treatment regimen, and sex with plasma NfL levels.

Rebound HIV-1 in cerebrospinal fluid after antiviral therapy interruption is mainly clonally amplified R5 T cell-tropic virus

Summary: HIV-1 can persist in a latent reservoir in individuals receiving antiretroviral therapy, and rebound virus can be detected in the cerebrospinal fluid during treatment interruption. In this study, we found that high viral loads and clonally amplified viral lineages in the cerebrospinal fluid were correlated with the influx of white blood cells. Additionally, we did not observe rebound macrophage-tropic virus in the cerebrospinal fluid, indicating that the CNS may not be a source of this virus. We propose a model in which R5 T cell-tropic virus is released from infected T cells in the CNS during treatment interruption.

Tetramodal Chemical Imaging Delineates the Lipid-Amyloid Peptide Interplay at Single Plaques in Transgenic Alzheimer?s Disease Models

Summary: Beta-amyloid plaque pathology is a prominent feature of Alzheimer's disease, but the mechanisms linking it to the disease are not fully understood. A novel imaging approach was used to investigate the correlation between lipids and amyloid plaques in mouse models of AD. The results identified characteristic patterns of lipid-and peptide localizations in plaques and revealed different roles for specific lipids in plaque growth and amyloid fibrillation.

Plasma neurodegeneration biomarker concentrations associate with glymphatic and meningeal lymphatic measures in neurological disorders

Summary: In this study, associations between plasma neurodegeneration biomarker concentrations and measures of glymphatic and meningeal lymphatic functions were examined in individuals with neurological disorders. The results suggest that plasma concentrations of neurodegeneration biomarkers are associated with CSF clearance functions.

Correlative Chemical Imaging and Spatial Chemometrics Delineate Alzheimer Plaque Heterogeneity at High Spatial Resolution

Summary: We propose a novel correlative chemical imaging strategy that integrates multimodal matrix-assisted laser desorption/ionization (MALDI) mass spectrometry imaging (MSI), hyperspectral microscopy, and spatial chemometrics. Our approach overcomes challenges associated with data acquisition and alignment by using evolutionary image registration and multiblock orthogonal component analysis. This allows for the identification of covariations of biochemical signatures between and within imaging modalities. We demonstrate the potential of our method in delineating chemical traits of Alzheimer's disease pathology and improving image fusion for correlative MSI and fluorescence microscopy.

Biomarkers of Alzheimer's Disease and Cerebrovascular Disease in Relation to Depressive Symptomatology in Individuals With Subjective Cognitive Decline

Summary: This study found a significant association between depressive symptomatology and SCD-memory and SCD-concentration. Depressive symptomatology was not associated with AD and CVD biomarkers. The phosphorylated tau biomarker and depressive symptomatology predicted SCD-memory, while the A beta 42/40 ratio and depressive symptomatology predicted SCD-concentration.

TREM2 is down-regulated by HSV1 in microglia and involved in antiviral defense in the brain

Summary: Immunological control of viral infections in the brain, especially through the TREM2 pathway, is crucial for protection against herpes simplex virus type 1 (HSV1) infection and maintenance of brain integrity. The down-regulation of TREM2 expression in HSV1-infected microglia leads to impaired antiviral response and decreased ability to phagocytose infected neurons. This study highlights the importance of TREM2 in the immune defense against viral infections and raises questions about its role in neurological pathologies.

Investigating New Applications of a Photoswitchable Fluorescent Norbornadiene as a Multifunctional Probe for Delineation of Amyloid Plaque Polymorphism

Summary: This study discovered a new fluorescent probe, NBD1, which can effectively diagnose beta amyloid plaques in Alzheimer's disease and studied its spectral and switching properties. This finding opens up new avenues for using NBD-based fluorescent probes for super-resolution or dual-color imaging and multifunctional microscopy to study the heterogeneity of amyloid plaques.

Summary: Using diffusion tensor imaging along perivascular spaces, the study shows reduced glymphatic clearance in subjects with traumatic brain injury compared to healthy controls and its association with injury severity biomarker. The glymphatic system plays a crucial role in clearing waste products and toxins from the brain during sleep. Failure of glymphatic system is thought to contribute to protein deposition in neurodegenerative disorders. The study suggests that understanding glymphatic functioning after traumatic brain injury could lead to novel therapies for better recovery and reduced risk of neurodegeneration.

Cerebrospinal fluid and positron-emission tomography biomarkers for noradrenergic dysfunction in neurodegenerative diseases: a systematic review and meta-analysis

Summary: The noradrenergic system undergoes pathological modifications in aging and neurodegenerative diseases, and significant neuronal loss in Alzheimer's disease and Parkinson's disease. A meta-analysis found that individuals with Parkinson's disease had decreased levels of CSF noradrenaline and 3-methoxy-4-hydroxyphenylglycol, as well as noradrenaline transporters availability in the hypothalamus. In Alzheimer's disease dementia, only 3-methoxy-4-hydroxyphenylglycol levels were increased compared to controls. Further studies are needed to understand the association between these biomarkers and clinical symptoms and pathology.

Associations between liver function and cerebrospinal fluid biomarkers of Alzheimer's disease pathology in non-demented adults: The CABLE study
