Summary: The expression of Receptor activator of NF-kappa B (RANK) is increased in podocytes of patients with diabetic nephropathy. Mouse models of podocyte-specific RANK depletion and overexpression showed that RANK depletion alleviated the development of kidney diseases, while RANK overexpression aggravated the condition. The findings suggest that RANK mediates the development of diabetic nephropathy by promoting glomerular oxidative stress and proinflammatory cytokine production.

Summary: This study investigated the role of p75 neurotrophin receptor (p75NTR) in cognitive dysfunction in Alzheimer's disease (AD). In vivo and in vitro models of AD were established using C57BL/6 mice and neuroblastoma cells treated with amyloid-beta1-42 (Aβ1-42), respectively. RNA-sequencing and bioinformatics analysis predicted the downstream genes of p75NTR. The interaction between p75NTR, nuclear factor kappa B (NF-κB), microRNA-210-3p (miR-210-3p), and phosphoethanolamine cytidylyltransferase 2 (PYCT2) was verified, and their effects on cognitive behaviors and biological characteristics of hippocampal neurons in AD mouse models were analyzed. p75NTR knockout alleviated cognitive dysfunction and reduced hippocampal neuron damage and apoptosis in AD-like mice. p75NTR upregulated miR-210-3p expression by activating NF-κB, thereby limiting PCYT2 expression. PCYT2 silencing in p75NTR-/- mice promoted neuronal apoptosis and aggravated cognitive dysfunction in AD mouse models. In summary, p75NTR accelerates cognitive dysfunction in AD through the NF-κB/miR-210-3p/PCYT2 axis.

Summary: Interstitial Cells of Cajal (ICC) are crucial for peristaltic contractions in the gastrointestinal and urinary tract. This study investigates the molecular signals of oxidative stress and apoptosis in ICC induced by tumor necrosis factor-alpha (TNF-alpha). The results demonstrate that TNF-alpha induces inflammation, oxidative stress, and cell apoptosis in ICC. Additionally, MEG3 mediates these effects through the miR-21/IKKB-NF-kappa B axis. This study enhances our understanding of ICC-related diseases at the molecular level.

Summary: This study investigated the protective effects of the natural alkaloid leonurine against iron-induced hepatotoxicity in rats. The results showed that leonurine suppressed ferroptosis, oxidative damage, and inflammation in the liver tissues. It also enhanced the antioxidant potential and improved histopathological abnormalities. Mechanistically, leonurine modulated the Nrf2 and NF-kappa B signaling pathways. These findings highlight the hepatoprotective activity of leonurine against iron-induced hepatotoxicity.

A study has revealed that the atypical I kappa B protein, Bcl3, plays a crucial role in enhancing the population of the p52:p52 homodimer within the NF-kappa B family. Bcl3 competes with other NF-kappa B subunits for efficient p52:p52 homodimer formation, leading to the upregulation of target genes involved in cell proliferation, migration, and inflammation. The aberrant activation of Bcl3 and p52 contributes to cancer.

Summary: The p75 neurotrophin receptor contains a death domain (DD) that plays a crucial role in apoptotic and inflammatory signaling. Conflicting findings and controversy exist regarding the oligomeric states of the p75NTR-DD. However, new evidence from biophysical and biochemical studies demonstrates the coexistence of symmetric and asymmetric dimers of the p75NTR-DD, which can equilibrate with the monomeric form in solution. This reversible behavior may be important for the intracellular signaling function of the p75NTR-DD.

Summary: Nerve growth factor (NGF) and its precursor, proNGF, play critical roles in neuronal survival and cognitive function. Dysregulation of proNGF may contribute to age-related memory loss and Alzheimer's disease. Increased reactive oxygen and nitrogen species (ROS/RNS) in the brain may impair proNGF transport and signalling, leading to vulnerability of the basal forebrain in AD. Antioxidant treatments may help restore proNGF signalling and reduce neurodegeneration and cognitive deficits in the basal forebrain.
