CD147, also known as extracellular matrix metalloproteinase inducer, is a widely distributed cell surface glycoprotein that belongs to the immunoglobulin superfamily. CD147 has been proved to be enriched on the surface of many tumor cells, promoting tumor growth, invasion and metastasis by its stimulation effect on adjacent fibroblasts to produce matrix metalloproteinases. In this study, we aimed to explore the expression pattern of CD147 in glioblastoma (GBM) and investigate whether it could be used to assess subsequent prognosis of patients. For that, we recruited a total of 206 patients with pathologically confirmed GBM and 36 normal control brain tissue specimens. The expression of CD147 in GBM and normal tissues was investigated by immunohistochemistry assay. Genetic factors including MGMT and IDH1 mutation were also investigated to justify the prognostic significance of CD147. Results showed that CD147 expression was increased in GBM compared with that in normal tissues. Kaplan-Meier analysis showed that increased CD147 expression was associated with poor overall survival of patients with GBM. Moreover, Cox's proportional hazards model revealed that CD147 expression was an independent and significant prognostic marker of overall survival in GBM patients. These results proved that CD147 expression was relatively abundant in GBM and can be potentially used to predict prognosis and treatment response in GBM patients.

The Role of m6A Regulator-Mediated Methylation Modification and Tumor Microenvironment Infiltration in Glioblastoma Multiforme

Liang Wang, Haiyan Cao, Ying Zhong, Peigang Ji, Fan Chen

Summary: This study reveals the important role of m6A modifications in GBM tumor microenvironment (TME) cell infiltration and stemness characteristics. The m6A modification patterns are correlated with TME cell infiltration characteristics, stemness characteristics, tumor clinical outcome, immune infiltration, and stemness. Assessing the m6A modification pattern of individual tumors can enhance our understanding of TME infiltration and stemness characteristics and contribute to the development of immunotherapeutic strategies.

Summary: This study found that different immune subtypes have a significant impact on the survival of glioma patients. Patients with a high immune response subtype had a shorter survival, and the immune microenvironment of these patients showed enhanced immune cell infiltration. Furthermore, 132 genes related to glioma immunity were identified, and the expression levels of seven core genes were significantly correlated with the prognosis of glioma patients. These findings could be used to stratify glioma patients based on immune subgroup analysis and guide clinical treatment regimens.

Research Progress on Intracranial Lymphatic Circulation and Its Involvement in Disorders

Fan Chen, Xuan Xie, Liang Wang

Summary: The lymphatic system plays an important role in the circulatory system, with functions including immune defense, maintaining internal environment stability, and regulating tissue pressure. Recent studies have confirmed the presence of lymphatic vessels lined with endothelial cells in the CNS, particularly in the meninges. These discoveries have significant clinical implications for infectious diseases, autoimmunity, and tumor immunology.

Wnt signaling regulates MFSD2A-dependent drug delivery through endothelial transcytosis in glioma

Yuan Xie, Liqun He, Yanyu Zhang, Hua Huang, Fan Yang, Min Chao, Haiyan Cao, Jianhao Wang, Yaling Li, Lingxue Zhang, Lele Xin, Bing Xiao, Xinxin Shi, Xue Zhang, Jiefu Tang, Lene Uhrbom, Anna Dimberg, Liang Wang, Lei Zhang

Summary: Inhibiting the Wnt signaling pathway can increase Temozolomide (TMZ) delivery and sensitize glioma to chemotherapy. Transcriptome analysis revealed that Wnt signaling inhibition enhances vascular transcytosis by upregulating PLVAP and downregulating MFSD2A. Additionally, Wnt signaling inhibition also enhances caveolae-mediated transcytosis in tumor endothelial cells (ECs). These findings suggest that Wnt signaling regulates MFSD2A-dependent TMZ delivery through a caveolae-mediated pathway, highlighting Wnt signaling as a promising therapeutic target for GBM treatment.

Prognostic and predictive factors of secondary gliosarcoma: A single-institution series of 18 cases combined with 89 cases from literature

Jinghui Liu, Chen Li, Yuan Wang, Peigang Ji, Shaochun Guo, Yulong Zhai, Na Wang, Meng Xu, Julei Wang, Liang Wang

Summary: This study assessed the clinicopathological features and outcomes of secondary gliosarcomas (SGS) through retrospective analysis of data. The results showed that initial diagnosis of GBM was a poor prognostic factor for SGS. Patients with initial non-GBM diagnosis who underwent gross total resection and chemoradiation had better overall survival outcomes. Patients initially diagnosed with GBM who received both chemoradiotherapy and active therapy after disease progression to SGS had significantly longer overall survival.

Real-time imaging of traumatic brain injury using magnetic induction tomography

Summary: In this study, an improved magnetic induction tomography (MIT) data acquisition system was used to monitor traumatic brain injury (TBI) and distinguish different injury levels. The experiment used a pneumatically controlled cortical impactor to strike the parietal lobe of anesthetized rabbits and establish different TBI models. The MIT data acquisition system continuously monitored the brain for an hour and successfully observed targets with increased conductivity in the reconstructed images, indicating that MIT may facilitate early TBI monitoring and distinguish varying degrees of injuries.

Summary: This study identified the elevated expression of CKAP4 in glioblastoma multiforme (GBM) and investigated its role in the development of the disease. CKAP4 was found to influence the malignant behavior of GBM through the regulation of the FOXM1 signaling pathway. The CKAP4-based prognostic model developed in this study could potentially guide individualized treatment decisions for glioma patients.
