Breast cancer brain metastases express the sodium iodide symporter

Breast cancer brain metastases are on the rise and their treatment is hampered by the limited entry and efficacy of anticancer drugs in this sanctuary. The sodium iodide symporter, NIS, actively transports iodide across the plasma membrane and is exploited clinically to deliver radioactive iodide into cells. As in thyroid cancers, NIS is expressed in many breast cancers including primary and metastatic tumors. In this study NIS expression was analyzed for the first time in 28 cases of breast cancer brain metastases using a polyclonal anti-NIS antibody directed against the terminal C-peptide of human NIS gene and immunohistochemical methods. Twenty-five tumors (84%) in this retrospective series were estrogen/progesterone receptor-negative and 15 (53.6%) were HER2+. Overall 21 (75%) cases and 80% of HER2 positive metastases were NIS positive. While the predominant pattern of NIS immunoreactivity is intracellular, plasma membrane immunopositivity was detected at least focally in 23.8% of NIS-positive samples. Altogether, these findings indicate that NIS expression is prevalent in breast cancer brain metastases and could have a therapeutic role via the delivery of radioactive iodide and selective ablation of tumor cells.

Summary: Immune effector cell-associated neurotoxicity syndrome (ICANS) is a common condition following CAR T therapy and other cancer cell therapies, but its underlying pathophysiology and neuropathology are not fully understood. This study presents the cellular and tissue neuropathologic analysis of a patient with severe ICANS after anti-CD19 CAR T therapy, revealing a pattern of multifocal demyelinating leukoencephalopathy associated with severe ICANS. The loss of region-specific oligodendrocyte lineage cells is proposed as a potential cellular and pathophysiologic factor in severe ICANS. Elucidation of the mechanistic processes underlying ICANS is crucial for minimizing neurotoxicity in CAR T-cell and immunotherapy treatments.

Purification and characterization of human neural stem and progenitor cells

Summary: The human brain undergoes rapid development during mid-gestation, and neural stem and progenitor cells (NSPCs) play a key role in this process. However, the lack of precise purification methods has hindered the functional study of NSPCs. In this study, researchers developed a method to isolate ten distinct NSPC types from the developing human brain using cell-surface markers. These isolated cells were able to differentiate into various neural lineages, providing a valuable framework for studying the functions of different cell types in human neurodevelopment.

Summary: Multiple sclerosis (MS) is an immune-mediated demyelinating disease of the central nervous system, and imaging strategies using TREM1-PET have the potential to aid in the diagnosis and monitoring of therapeutic responses. In this study, the effectiveness of TREM1 PET imaging in the EAE mouse model of MS was validated, and it was found to be more sensitive than the established TSPO-PET imaging method. TREM1-PET imaging has the potential to play a role in the diagnosis of MS and monitoring of therapeutic responses to drug treatment.

Synthetic whole-slide image tile generation with gene expression profile-infused deep generative models

Summary: In this study, a novel approach using deep generative models, infused with gene expression profiles, is proposed to generate high-quality whole-slide image (WSI) tiles. The RNA-GAN model showed superior performance compared to traditional GANs, with preference by expert pathologists and shorter training time. Moreover, RNA-GAN demonstrated the ability to generalize to unknown gene expression profiles.

Pediatric Brain Tumours: Lessons from the Immune Microenvironment

Summary: Despite advances in tumour molecular subtyping, pediatric brain tumours (PBTs) still pose a significant challenge and cause high mortality rates. Tumour immunotherapy has emerged as a promising approach for treating childhood tumours, including PBTs, by leveraging the immune landscape of the developing brain. This review focuses on understanding the immune microenvironments of common PBTs and how they shape responses to immunotherapy, with the goal of informing future treatment designs.

CHEK2 mutations in pediatric brain tumors

Summary: This study reviewed 3 pediatric patients with CNS tumors having CHEK2 mutations. Germline testing in 1 case confirmed the hereditary nature of CHEK2 mutation. The study suggests that CHEK2 may contribute to carcinogenesis and reporting similar cases may help identify a new molecular driver in pediatric CNS tumors.
